Elaeis guineensis Jacq. has gained a reputation in the food industry as an incredible crop capable of supplying the world’s largest edible oil production. In Ecuador, an important oil palm-producing country, this crop is affected in a high percentage by the bud rot disease, which is responsible for palm death. The main objective of the investigation was dedicated to understanding the palm defense mechanism facing bud rot disease, translated in the induction of reactive oxygen species, activation of defensive machinery comprising enzymatic and non-enzymatic antioxidative components, secondary metabolites, carotenoids accumulation in the palm during all stages of disease infection. For this, a survey was conducted in different oil palm plantations in the Esmeraldas province, one of the most representative for its highest incidence of bud rot disease. The survey completed DPPH, FRAP, ABTS, and other spectrophotometric analyses to underline the biochemical, biological, and physiological palm response front of bud rot incidence. The palm defense strategy in each disease stage could be represented by the phenolic compound’s involvement, an increment of antioxidant activity, and the high enzymatic activity of phenylalanine ammonia-lyase (PAL). The results of the investigation made understandable the palm defense strategy front of this disease, respectively, the antioxidative defense and the palm secondary compounds involved. 相似文献

Nucleic acid therapeutics (NATs), such as mRNA, small interfering RNA or antisense oligonucleotides are extremely efficient tools to modulate gene expression and tackle otherwise undruggable diseases. Spherical nucleic acids (SNAs) can efficiently deliver small NATs to cells while protecting their payload from nucleases, and have improved biodistribution and muted immune activation. Self-assembled SNAs have emerged as nanostructures made from a single DNA-polymer conjugate with similar favorable properties as well as small molecule encapsulation. However, because they maintain their structure by non-covalent interactions, they might suffer from disassembly in biologically relevant conditions, especially with regard to their interaction with serum proteins. Here, we report a systematic study of the factors that govern the fate of self-assembled SNAs. Varying the core chemistry and using stimuli-responsive disulfide crosslinking, we show that extracellular stability upon binding with serum proteins is important for recognition by membrane receptors, triggering cellular uptake. At the same time, intracellular dissociation is required for efficient therapeutic release. Disulfide-crosslinked SNAs combine these two properties and result in efficient and non-toxic unaided gene silencing therapeutics. Series of self-assembled and mono-dispersed bovine serum albumin (BSA)-conjugated ZnS/CuS nano-composites with different Zn/Cu ratios had been successfully synthesized by a combination method of the biomimetic synthesis and ion-exchange strategy under the gentle conditions. High-resolution transmission electron microscopy observation, Fourier transform infrared spectra and zeta potential analysis demonstrated that BSA-conjugated ZnS/CuS nano-composites with well dispersity had the hierarchical structure and BSA was a key factor to control the morphology and surface electro-negativity of final products. The real-time monitoring by atomic absorption spectroscopy and powder X-ray diffraction revealed that the Zn/Cu ratio of nano-composites could be controlled by adjusting the ion-exchange time. In addition, the metabolic and morphological assays indicated that the metabolic proliferation and spread of rat pheochromocytoma (PC12) cells could be inhibited by nano-composites, with the high anti-cancer activity at a low concentration (4 ppm). What were more important, Zn and Cu in nano-composites exhibited a positive cooperativity at inhibiting cancer cell functions. The microscope observation and biochemical marker analysis clearly revealed that the nano-composites-included lipid peroxidation and disintegration of membrane led to the death of PC12 cells. We give a precise mathematical formulation of a variational model for the irreversible quasi-static evolution of brittle fractures proposed by G. A. Francfort and J.-J. Marigo, and based on Griffith's theory of crack growth. In the two-dimensional case we prove an existence result for the quasi-static evolution and show that the total energy is an absolutely continuous function of time, although we cannot exclude the possibility that the bulk energy and the surface energy may present some jump discontinuities. This existence result is proved by a time-discretization process, where at each step a global energy minimization is performed, with the constraint that the new crack contains all cracks formed at the previous time steps.
