Coatings based on dendritic polyglycerol (dPG) were investigated for their use to control nonspecific protein adsorption in an assay targeted to analyze concentrations of a specific protein. We demonstrate that coating of the sample vial with dPG can significantly increase the recovery of an antibody after incubation. First, we determine the concentration dependent loss of an antibody due to nonspecific adsorption to glass via quartz crystal microbalance (QCM). Complementary to the QCM measurements, we applied the same antibody as analyte in an surface plasmon resonance (SPR) assay to determine the loss of analyte due to nonspecific adsorption to the sample vial. For this purpose, we used two different coatings based on dPG. For the first coating, which served as a matrix for the SPR sensor, carboxyl groups were incorporated into dPG as well as a dithiolane moiety enabling covalent immobilization to the gold sensor surface. This SPR-matrix exhibited excellent protein resistant properties and allowed the immobilization of amyloid peptides via amide bond formation. The second coating which was intended to prevent nonspecific adsorption to glass vials comprised a silyl moiety that allowed covalent grafting to glass. For demonstrating the impact of the vial coating on the accuracy of an SPR assay, we immobilized amyloid beta (Aβ) 1-40 and used an anti-Aβ 1-40 antibody as analyte. Alternate injection of analyte into the flow cell of the SPR device from uncoated and coated vials, respectively gave us the relative signal loss (1 − RUuncoated/RUcoated) caused by the nonspecific adsorption. We found that the relative signal loss increases with decreasing analyte concentration. The SPR data correlate well with concentration dependent non-specific adsorption experiments of the analyte to glass surfaces performed with QCM. Our measurements show that rendering both the sample vial and the sensor surface is crucial for accurate results in protein assays.  相似文献

In this study we report the novel polymeric resin poly(N-vinyl imidazole/ethylene glycol dimethacrylate) for the purification and isolation of phenolic acids. The monomer to crosslinker ratio and the porogen composition were optimized for isolating phenolic acids diluted in acetonitrile at normal phase chromatography conditions, first. Acetonitrile serves as polar, aprotic solvent, dissolving phenolic acids but not interrupting interactions with the stationary phase due to the approved Hansen solubility parameters. The optimized resin demonstrated high loading capacities and adsorption abilities particularly for phenolic acids in both, acetonitrile and aqueous solutions. The adsorption behavior of aqueous standards can be attributed to ion exchange effects due to electrostatic interactions between protonated imidazole residues and deprotonated phenolic acids. Furthermore, adsorption experiments and subsequent curve fittings provide information of maximum loading capacities of single standards according to the Langmuir adsorption model. Recovery studies of the optimized polymer in the normal-phase and ion-exchange mode illustrate the powerful isolation properties for phenolic acids and are comparable or even better than typical, commercially available solid phase extraction materials. In order to prove the applicability, a highly complex extract of rosemary leaves was purified by poly(N-vinyl imidazole/ethylene glycol dimethacrylate) and the isolated compounds were identified using UHPLC–qTOF-MS.  相似文献

This paper describes the use of dendritic polyglycerol as a new high-loading polymeric support. The soluble polyether skeleton allows the parallel synthesis of small libraries on a large scale (1-5 mmol). Purification of polymer-bound products is easily achieved by a parallel dialysis apparatus, which was developed to separate up to 12 reaction mixtures simultaneously. The terminal 1,2-diol groups of polyglycerol (loading capacity: 4.1 mmol diol/g) can be directly coupled with carbonyl compounds without additional linker groups. At the same time the polyglycerol support acts as a polymeric ketal protecting group. The coupling of the carbonyl compounds occurs in high yields, and effective loading capacities of up to 3.5 mmol of ketone/g can be reached. The obtained polymeric acetals can easily be characterized by standard analytical techniques, such as NMR, IR, UV, and SEC. The versatility of this new polymeric support for solution-phase organic synthesis is demonstrated by two efficient polymer-supported syntheses: nucleophilic substitutions of gamma-chloroketones with amines and Suzuki-coupling on p-bromobenzaldehyde. The acid-catalyzed acetal cleavage with a solid-phase acidic ion-exchange resin in methanol demonstrates the orthogonal use of these soluble polymeric supports with conventional solid-phase reagents. Cleavage of products occurs in high yields, and almost complete recovery (>95%) of the polyglycerol support has been demonstrated after phase separation or ultrafiltration.  相似文献

In this review, important aspects of λ3-2H-azaphosphirene metal complexes are discussed in relation to synthesis, physical properties and synthetic applications; ab-initio calculations on relative energies of CH2NP isomers and of λ3-2H-azaphosphirene metal complexes (Cr, Mo, W) are also presented. Currently, there are three routes to this unsaturated three-membered ring system, which are discussed in detail: two of them use a rearrangement of metal carbene complexes, whereas the other relies on [2+1] cycloaddition reactions of electrophilic terminal phosphanediyl complexes and carbonitriles. The structural data show characteristics of a very strained heterocyclic ring system, which partially helps to understand the reactivity of this heterocycle complex in solution. The synthetic potential of λ3-2H-azaphosphirene metal complexes is illustrated by selected examples, which demonstrate their ability to serve, under very mild conditions as precursor for various new building blocks, i.e. nitrilium phosphanylide complexes, electrophilic terminal phosphanediyl complexes and phosphavinyl-nitrene complexes.  相似文献

1,2‐Diorganylsubstituted derivatives of hydrazinium azide were examined in order to investigate their higher volatility and higher sensitivity to initiation compared to 1,1‐diorganylsubstituted hydrazinium azide derivatives. The compounds were synthesized from the respective hydrazines by reaction with HN3 and characterized by elemental analysis, vibrational (IR, Raman) and multinuclear NMR spectroscopy (1H, 13C, 14N). Their sensitivity to friction, shock, electrostatic impact and heat was examined and the explosion products were investigated. The crystal structure of pyrazolidinium azide was determined.  相似文献

The novel N,P,C‐cage complexes 5 a – f and 6 a – f have been obtained by the reaction of the P‐pentamethylcyclopentadienylphosphinidene complex 2 , generated thermally from 2H‐azaphosphirene complex 1 , with N‐methyl‐C‐arylcarbaldimines 3 a – f . Li/Cl phosphinidenoid complex 8 reacted with 3 a , b to give N,P,C‐cage complexes 6 a , b , whereas with 3 c – f , complexes 6 c – f were obtained in negligible amounts only. Both types of ligand N,P,C‐cage structures 5 and 6 were found to be in an unprecedented equilibrium, with 5 a , f as the predominant species. Transient electrophilic terminal phosphinidene complexes 10 a – f serve as intermediates in both ligand interconversions ( 5 a , f ? 6 a , f ), as evidenced through trapping reactions with phenylacetylene and N‐methyl‐C‐phenylcarbaldimine, thus leading to the novel N,P,C‐cage complexes 13 b and 15 . DFT calculations predicted a small difference in the relative energies of the two types of N,P,C‐cage ligands, and a remarkable stabilisation of the aminophosphinidene complex 10 as the common precursor, thereby providing an insight into this surprising 5‐ring–3‐ring interconversion. In depth analysis of intermediate 10 revealed the occurrence of both through‐bond (conventional inductive/mesomeric effects) and through‐space (non‐covalent interactions) mechanisms, which amount to 67.8 and 14.4 kcal mol?1, respectively, and account for the remarkable stabilisation of this intermediate.  相似文献

We synthesized melemium hydrogensulfate H3C6N7(NH2)3(HSO4)3 by reaction of melem with 70 % sulfuric acid. The crystal structure was elucidated by single‐crystal XRD (P21/n (no. 14), Z = 4, a = 10.277(2), b = 14.921(3), c = 11.771(2) Å, β = 99.24(3)°, V = 1781.5(6) Å3). H3C6N7(NH2)3(HSO4)3 is the first compound displaying a triple protonation of melem., In this contribution an overview of accessible melemium sulfates depending on the concentration of sulfuric acid is given. Two additional melemium sulfates were identified this way.  相似文献

Fluorescent silica nanoparticles (SiNPs) were prepared by covalent attachment of fluorophores to the amino-modified surface of SiNPs with a typical diameter of 15 nm. The SiNPs are intended for use in novel kinds of fluorescence resonance energy transfer (FRET)-based affinity assays at the interface between nanoparticle and sample solution. Various labels were employed to obtain a complete set of colored SiNPs, with excitation maxima ranging from 337 to 659 nm and emission maxima ranging from 436 nm to the near infrared (710 nm). The nanoparticles were characterized in terms of size and composition using transmission electron microscopy, thermogravimetry, elemental analysis, and dynamic light scattering. The surface of the fluorescent SiNPs was biotinylated, and binding of labeled avidin to the surface was studied via FRET in two model cases. In the first, FRET occurs from the biotinylated fluorescent SiNP (the donor) to the labeled avidin (the acceptor). In the second, FRET occurs in the other direction. Aside from its use in the biotin–avidin system, such SiNPs also are believed to be generally useful fluorescent markers in various kinds of FRET assays, not the least because the fluorophore is located on the surface of the SiNPs (and thus always much closer to the second fluorophore) rather than being doped deep in its interior.  相似文献

This paper describes the behavior of various generations of polyglycerol dendrimers that contain a perfluorinated shell. The aggregation in organic solvents is based on supramolecular fluorous–fluorous interactions, which can be described by means of 19F NMR spectroscopy. In order to study the interaction and aggregation phenomena of dendrimers with perfluorinated shell and perfluoro‐tagged guest molecules we investigated [G3.5]‐dendrimer with a perfluorinated shell in the presence of perfluoro‐tagged disperse red. Noteworthy, the interaction intensities varied in an unexpected manner depending on the equivalents of perfluoro‐tagged guest molecules added to the dendrimers in solution which then formed supramolecular complexes based on fluorous–fluorous interactions. We found that these complexes aggregated around residual air in the solvent to form stable micron‐sized bubbles. Their sizes correlated with the interaction intensities measured for certain dendrimer–guest molecule ratios. Degassing of the solutions led to a quasi phase separation between organic and fluorous phase, whereby the dendrimers formed the fluorous phases. Regassing the sample with air afforded bubbles of the initial size again.  相似文献
