Two different types of -substituted alkanethiol/disulfide compounds have been used to prepare monolayer architectures on gold serving as platforms for the immobilization of receptor probe molecules – antibodies. These are: (i) carboxylic acid alkanethiols post-reacted with amino biotin to generate streptavidin surfaces, and (ii) N-hydroxysuccinimide-terminated disulfide surfaces. The properties of the monolayers, with and without attached receptor probe molecules, were analysed using infrared spectroscopy, ellipsometry, fluorescence scanning and atomic force microscopy. Several experimental parameters, such as condensation reagents, additives, probe and target concentrations and immobilization time, were systematically varied to determine the dynamic range and to optimize the sensitivity and signal-to-noise ratio of the biochip platforms. Fluorescence screening using Cy5-labelled antigens finally demonstrated that both surfaces could be successfully employed to immobilize the antibodies. A reaction system consisting of terephthalic acid, NaOH, inorganic Mn(II) or Mn(III) salt, and salicylidene alkylimine resulted in dinuclear manganese complexes (salpn)(2)Mn(2)(mu-phth)(CH(3)OH)(2) (1, salpn = N,N'-1,3-propylene-bis(salicylideneiminato); phth = terephthalate dianion), (salen)(2)Mn(2)(mu-phth)(CH(3)OH)(2) (2, salen = N,N'-ethylene-bis(salicylideneiminato)), (salen)(2)Mn(2)(mu-phth)(CH(3)OH)(H(2)O) (3), and (salen)(2)Mn(2)(mu-phth) (4), while the absence of NaOH in the reaction led to a mononuclear Mn complex (salph)Mn(CH(3)OH)(NO(3)) (5, salph = N,N'-1,2-phenylene-bis(salicylideneiminato)). In addition, a trinuclear mixed metal complex H[Mn(2)Na(salpn)(2)(mu-OAc)(2)(H(2)O)(2)](OAc)(2) (6) was obtained from the reaction system by using maleic acid instead of terephthalic acid. Five-coordinate Mn ions were found in 4 giving rise to an intermolecular interaction and constructing a one-dimensional linear structure. Antiferromagnetic exchange interactions were observed for 1-3, and a total ferromagnetic exchange of 4 was considered to stem from intermolecular magnetic coupling. (1)H NMR signals of phenolate ring and alkylene (or phenylene) backbone of the diamine are similar to those reported in the literature, and the phth protons are at -2.3 to -10.1 ppm. Studies on structure, bond valence sum analysis, and magnetic properties indicate the oxidation states of the Mn ions in 6 to be +3, which are also indicated by ESR spectra in dual mode. Ferromagnetic exchange interaction between the Mn(III) sites was observed with J = 1.74 cm(-1). A quasireversible redox pair at -0.29V/-0.12V has been assigned to the redox of Mn(2)(III)/Mn(III)Mn(II), implying the intactness of the complex backbone in solution.  相似文献

Summary. A symmetrical C(10)-selena-bilirubin analog, 8,12-bis(2-carboxyethyl)-7,13-dimethyl-2,3,17,18-tetraethyl-10-selenabiladiene-ac-1,19(21H,24H)-dione was synthesized from 8-(2-carboxyethyl)-2,3-diethyl-7-methyl-(10H)-dipyrrin-1-one in one step by reaction with diselenyl dichloride. The selena-rubin exhibited UV-vis and NMR spectroscopic properties similar to those of the parent mesobilirubin, and like bilirubin and mesobilirubin, it adopts an intramolecularly hydrogen-bonded conformation, shaped like a ridge-tile but with a steeper pitch. The longer C–Se bond lengths (2.2Å) and smaller bond angles at C–Se–C (88°), as compared to C–CH2–C (1.5Å, 106°), lead to an interplanar angle between the two dipyrrinones of only 72°, which is considerably less than that of bilirubin (100°) and close to that (74°) of its 10-thia-rubin analog. Despite the conformational distortion, the sensitivity of Se toward oxidation and the typically weak C–Se bond, the selena-rubin is metabolized in normal rats, like bilirubin, to acyl glucuronides, which are secreted into bile. In mutant (Gunn) rats lacking bilirubin glucuronosyl transferase (UGT1A1), glucuronide or other metabolites of the selena-rubin were not detected in bile, demonstrating the importance of hepatic glucuronidation for its biliary excretion.  相似文献

The mesoionic compound, 1,4,5-triazole-2-thiolate, is a versatile ligand, capable of binding metal centres in different ways. The present study shows how it reacts with the bridging platinum(II) complex μ-dichlorobis[(chlorotriethylphosphineplatinum(II)], [Pt2(PEt3)2Cl4], originating a bimetallic complex. Both the sulphur atom and nitrogen 3 behave as basic sites, coordinating different platinum centres. An interesting isomerization process, shown by 31P{1H} n.m.r. spectroscopy, occurs at the S-bonded platinum moiety, resulting in a fluxional behaviour of the complex. Simple tandemly organized (GTG)n/(CAC)n sequences are spread throughout the human chromosomes. The most informative DNA fingerprints for the testing of pedigrees and/or paternity were obtained with the simple triplet repeat probe (GTG)5 or its complement (CAC)5. These hypervariable simple-repeat fragments are stably inherited in a Mendelian fashion. Using these highly discriminating probes, all human individuals could, theoretically, be differentiated, except for genetically identical monozygotic twins. Examples from actual case work are reported and pertinent advantages of this methodology are discussed.  相似文献

The determination of uranium in natural waters by XRF analysis after its preconcentration with some organic precipitants is described.109Cd source is used for the excitation of L-series X-rays of uranium. Separation yield, kinetics of coprecipitation and the possibility of preparing thin-layer samples for XRF analysis were studied in detail for uranium concentration with α-nitroso-β-naphtol, methylene blue with ammonium thiocyanate, tannin with urotropine and other precipitants. A procedure using coprecipitation of uranium with α-nitroso-β-naphtol from 500 ml samples of surface or waste, waters is proposed.  相似文献

A kinetic method for the determination of Te based on its inhibitory effect on the PdII-catalysed reaction between pyronine G and H2PO2- is described. The influence of experimental variables on the rate of the process and the potential interfering effect of a large number of ions has been studied. Under the selected experimental conditions: 6 x 10(-5) M pyronine G; 0.6 M H2PO2-; pH 2.6, adjusted with Britton-Robinson buffer; 0.80 microgram ml-1 of PdII; and a temperature of 22 +/- 0.2 degrees C, Te was determined in the concentration range 0.08-0.85 microgram ml-1. The method was applied to the determination of Te in waters and lead concentrates.  相似文献

In this work a modified adsorption method was used to form firstly a saturated Cd or Pb adlayer on platinum single crystal (100) surface, then its surface structure, the electrochemical stability and the desorption-readsorption CV properties were studied. The HCOOH oxidation has been used as a probe to determine the electrocatalytic propertics of Pt (100) with different surface concentration of Cd or Pb adatoms. The results show that Cd adatoms on Pt(100) inhibit both HCOOH dissociation adsorption (to form poisoning intermediates) and HCOOH oxidation via the reactive intermediate, and that Pb adatoms on Pt(100) inhibit only HCOOH disociation adsorption but catalyse HCOOH oxidation via the reactive intermediate, in this case the more Pb adatoms on Pt(100), the higher te catalytic effect. The highest enhanced catalytic effet has been found with the saturated Pb adlayer on Pt(100) (the coverage of Pb, calibrated by hydrogen adsorption, is then equal to 1.0), i.e. with a surface structure of C(2×2)-bridged Pb. The difference of influence to the hydrogen adsorption-desorption process and the different catalytic effect on HCOOH oxidation of Cd and Pb adatoms on Pt(100) were attributed to the intrinsic properties of Cd and Pd elements and also to their interaction with Pt(100). This results indicate that the electrocatalytic mechanism of Cd and Pb adatom on Pt(100) depends on the geometrical surface structure effect and, more importantly, on the electro structure effect during the interaction of adatoms with Pt(100) surface atoms.  相似文献
