Simultaneous estimation of four proton pump inhibitors—lansoprazole,omeprazole, pantoprazole and rabeprazole: development of a novel generic HPLC‐UV method and its application to clinical pharmacokinetic study

A highly selective, sensitive and accurate HPLC method has been developed and validated for the estimation of four proton‐pump inhibitors (PPI), lansoprazole (LPZ), omeprazole (OPZ), pantoprazole (PPZ) and rabeprazole (RPZ), with 500 µL human plasma using zonisamide as an internal standard (IS). The sample preparation involved simple liquid–liquid extraction of LPZ, OPZ, PPZ and RPZ and IS from human plasma with ethyl acetate. The baseline separation of all the peaks was achieved with 0.1% triethylamine (pH 6.0):acetonitrile (72:28, v/v) at a flow rate of 1 mL/min on a Zorbax C₈ column. The total chromatographic run time was 11.0 min and the simultaneous elution of IS, OPZ, RPZ, PPZ and LPZ occurred at approximately 2.42, 4.45, 5.02 and 9.37 min, respectively. The method was proved to be accurate and precise at linearity range of 20.61–1999.79 ng/mL with a correlation coefficient (r) of ≥0.999. The limit of quantitation for each of the PPI studied was 20.61 ng/mL. The intra‐ and inter‐day precision and accuracy values were found to be within the assay variability limits as per the FDA guidelines. Lipase activity of Lecitase® Ultra: characterization and applications in enantioselective reactions

The general properties of Lecitase® Ultra, a phospholipase manufactured and marketed by Novozymes, Denmark, have been studied after purification by ultrafiltration. The enzyme has a molecular mass of 35 KD, pH-optimum of 8.5, and appears to possess a single active site which exhibits both the lipase and phospholipase activities that increase in the presence of Ca²⁺ and Mg²⁺ ions. The enzyme is inhibited by heavy metal ions and surfactants, and does not accept p-nitrophenyl acetate and glycerol triacetate. Substrates, such as glycerol tributyrate and p-nitrophenyl palmitate, esters of N-acetyl-α-amino acids and α-hydroxy acids are readily accepted. Amino acids with aliphatic residues, such as alanine, isoleucine, and methionine, are hydrolyzed with high enantioselectivity for the l-enantiomer (E >100), but amino acids with aromatic residues such as phenylalanine and phenylglycine, and esters of α-hydroxy acids are hydrolyzed with low enantioselectivity (E = 1–5). Immobilization of the enzyme in a gelatin matrix (gelozyme) leads to a marginal improvement in the enantioselectivity for these substrates. However, a dramatic improvement in enantioselectivity is observed for ethyl 2-hydroxy-4-oxo-4-phenylbutyrate (E value increases from 4.5 to 19.5 with S-selectivity). Similarly, glycidate esters, such as ethyl trans-(±)-3-phenyl glycidate and methyl trans-(±)-3-(4-methoxyphenyl) glycidate, are selectively hydrolyzed with a remarkable selectivity towards the (2S,3R)-enantiomer providing unreacted (2R,3S)-glycidate esters (ee >99%, conversion 52–55%) by the immobilized enzyme.     

Evaluation of one-centre electron interaction integrals over slater type atomic orbitals

Simplified expressions for one-centre electron interaction integrals in general and Slater-Condon parameters as well as repulsion integrals of the type (aa|aa) in particular have been obtained over Slater-type atomic orbitals. Results calculated using these expressions are in agreement with those due to other authors.     

Role of tunneling in electron accretion on negatively charged dust particles

In this Letter the authors have shown that the tunneling of electrons through a potential energy barrier around a negatively charged particle in a dusty plasma must be taken into account in the evaluation of the accretion current. Both the linear and nonlinear screenings have been considered.     

Direct amplitude detection of Zernike modes by computer-generated holographic wavefront sensor: Modeling and simulation

A fast holographic wavefront sensor is proposed using a computer-generated hologram (CGH). This CGH is a multiplexed hologram of different Zernike mode–amplitude combinations, and is designed in such a manner as to get the corresponding spots on the detector according to the presence and strength of a particular aberration. Interference between the aberrated wavefront (with a single mode–amplitude combination) and the Fourier transform of an image with single bright pixel (defined as dot image) is numerically calculated for one hologram. Different mode–amplitude combination and corresponding different positions of bright pixels (dots) are taken to compute various holograms and then all the holograms are multiplexed to get the final hologram. When the aberrated wavefront with a particular mode–amplitude combination is incident onto the multiplexed hologram, the corresponding dot is generated in the Fourier plane. A lens performs the Fourier transform in optical domain and provides the instant detection of amplitude of the respective Zernike mode. The main advantage of the scheme is to avoid the need of any computations, which makes it really fast. The simulation results are presented with the cross-talk analysis for few Zernike terms.
