The highly crowded 3,3-bis(di-tert-butylmethylsilyl)-1,2-bis(tri-tert-butylsilyl)cyclotrisilene (3) was newly designed as a precursor of the cyclotrisilenylium ion and prepared by the reaction of 2 equiv of dilithiosilane (tBu2MeSi)2SiLi2 (1) with 2,2,3,3-tetrabromo-1,1,1,4,4,4-hexa-tert-butyltetrasilane. The reaction of 3 with triphenylmethylium tetraarylborate in toluene produced (di-tert-butylmethylsilyl)bis(tri-tert-butylsilyl)cyclotrisilenylium ion (4+), which was isolated in the form of the tetraarylborate salt as extremely air- and moisture-sensitive yellow crystals, representing the first isolable silicon congener of the cyclopropenylium ion. The molecular structure of 4+.TSFPB- (TSFPB- = tetrakis[4-(tert-butyldimethylsilyl)-2,3,5,6-tetrafluorophenyl]borate) was established by X-ray crystallographic analysis, showing that the three-membered ring constitutes an almost equilateral triangle with average Si-Si bond lengths of 2.216(3) A. The X-ray crystal structure and spectral data show that 4+ is not only a free silyl cation but also a 2pi electron aromatic species with delocalization of the positive charge over the three-membered skeleton.  相似文献

Thymidine analogues bearing a variety of functional groups at the C5-position via an amino-linker arm were prepared and the substrate activity for PCR using thermophilic KOD Dash DNA polymerase was examined. The enzyme accepted the thymidine analogues bearing pyridine, imidazole, biotin, a cationic-charged guanidinium, a cationic-charged amino, mercaptopyridyl and phenanthrolne groups at the C5-position, forming the corresponding PCR product. However, a thymidine analogue bearing a carboxyl group at the C5-position was a poor substrate and the corresponding PCR products could not be obtained. The thymidine analogue bearing a mercapto group was also a poor substrate for the enzyme, because it dimerized by disulfide linkage under PCR conditions. The enzyme hardly accepts the thymidine analogues with a negatively-charged carboxyl group or a bulky group as a substrate. KOD Dash DNA polymerase, having a broader substrate specificity than any other DNA polymerase, will expand the variety of modified DNAs that can be prepared by PCR.  相似文献

The imaging of ultrafine Au, Pd, CdS, and ZnS particles prepared in reverse micelles has been studied by atomic force microscopy (AFM). Mica substrates, derivatized with a monolayer of amine or thiol-terminated silanes, were used to immobilize the particles. The substrates were exposed to reverse micellar solutions containing the particles and were then immersed in appropriate solvent media to remove surfactants. This resulted in a partial coating of the surfaces by the particles. The particle size was estimated as the height of protrusion, shown on the AFM images. The size values for the Pd and CdS particles, thus obtained, were found to be almost identical to those obtained by transmission electron microscopy (TEM), whereas those for the Au and ZnS particles were larger than those obtained by TEM. Scanning electron microscopy showed that the Au particles tended to aggregate on the surfaces, while Pd particles were isolated from one another. Copyright 2000 Academic Press.  相似文献

Branched tris‐DNA, in which two oligonucleotides of the same sequence and one other oligonucleotide of a different sequence are connected with a rigid central linker, was prepared chemically by using a DNA synthesizer. Two branched tris‐DNA molecules with complementary DNA sequences form dimer and tetramer as well as linear and spherical oligomer complexes. The complex formation was studied by UV/thermal denaturation, enzyme digestion, gel electrophoresis, and AFM imaging.  相似文献

A novel series of 1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid derivatives were synthesized and biologically evaluated. (S)-2-Benzyl-7-[2-(5-methyl-2-phenyloxazol-4-yl)ethoxy]-1,2,3,4-tetrahydroisoquinoline-3-carboxylic acid (10, KY-021) was identified as a novel peroxisome proliferators-activated receptor (PPAR) gamma agonist, which showed potent activity in human PPAR gamma (EC50=11.8 nM). KY-021 reduced plasma glucose and triglyceride levels at 3 mg/kg/d for 7 d in male KK-Ay mice. KY-021 also decreased plasma triglyceride levels at 0.3-3 mg/kg/d for 6 d, and improved oral glucose tolerance at 1 and 3 mg/kg/d for 7 d in male Zucker fatty rats. Maximal plasma concentration of KY-021 after oral administration at 10 mg/kg was 6.6 microg/ml and 2.1 microg/ml in male ICR mice and male SD rats, respectively. Repeated oral administration of KY-021 at 30 mg/kg/d for 10 weeks had little toxicity in male SD rats. These results demonstrated that KY-021 has great potential as an efficacious and safe drug for diabetes.  相似文献

A quantitative comparison between theory and experiment has been carried out for a collection-mode near-field optical microscope. A 30-nm (lateral dimension) cylindrical dielectric sample was imaged. This image was compared with the result of theoretical calculations that used a classical macroscopic nonglobal model based on the excitation theorem. Good agreement was obtained, with image inversion, edge enhancement, and edge asymmetry correctly predicted.  相似文献

ABSTRACT

We prepared crosslinked azotolane liquid-crystalline polymer (LCP) films doped with a stilbene derivative (two-photon chromophore) utilizing an interpenetrating polymer network (IPN) structure. The IPN films bend toward the light source upon irradiation with femtosecond laser pulses at 600 nm, which can excite the stilbene derivative by two-photon absorption. The bending speed of the IPN films increases with the square of the laser pulse intensity, which is compelling evidence for the two-photon processes.  相似文献
