We describe a new method for the fabrication of surface-chemical gradients. A film of titanium dioxide is brought into close proximity to a uniformly monolayer-covered surface and exposed to UV light to produce oxygen radicals. The use of a gradated grayscale mask between the UV source and the TiO2 allows the production of surface-chemical gradients via oxidation of the monolayer. The technique is demonstrated on gold surfaces bearing alkanethiol SAMs. Oxidation and subsequent replacement of the oxidized thiols has been used to produce surface-chemical gradients with lengths on the submillimeter to centimeter scales. The oxidation, removal, and replacement of the thiols during the process have been demonstrated by means of XPS. This oxidative process may be applied to other surface chemistries. Similarly, other shapes and slopes of gradients may be produced, depending on the photomask employed.  相似文献

This study reports a new microfluidic system with three integrated functional devices for pumping, mixing and separation of bio-samples by utilizing micro-electro-mechanical-systems technology. By using antibody-conjugated magnetic beads, the developed system can be used to purify and enrich virus samples such that the subsequent detection of viruses can be performed with a higher sensitivity. The target viruses were first captured by the antibody coated onto the magnetic beads by using a rotary micromixer which performed the incubation process. The viruses were then purified and enriched by a magnetic field generated by planar microcoils. The integrated microfluidic system can perform the whole purification and enrichment process automatically using a rotary micropump and appropriate microvalves. In addition, a numerical simulation was also employed to optimize the design of the microcoils and to investigate the magnetic field strength and distribution. The simulation results were consistent with experimental observations. Finally, the developed system was used to successfully perform the purification and enrichment of Dengue viruses. The detectable limit of Dengue viruses was found to be as low as 10(2) pfu ml(-1) by using this approach. Therefore, the integrated microsystem can perform incubation, transportation, mixing and purification of virus samples, possibly making it a promising platform for future biological and medical applications.  相似文献

Centrifugal precipitation chromatography and a high-speed counter-current chromatography system equipped with a spiral tubing support rotor (spHSCCC) were successfully applied for the identification and isolation of carotenoid cleavage-like enzymes from Enteromorpha compressa (L.) Nees. This is the first study separating active enzymes from a complex natural matrix by spHSCCC. The target enzymes were identified after fractionation of the proteins in an acetone Tris-buffer gradient by centrifugal precipitation chromatography. Also, an aqueous two-phase solvent system consisting of PEG 1000 and mono- and dibasic potassium phosphate was used for the isolation of the enzymes by spHSCCC. The purified fractions contained two proteins of 65 and 72 kDa, respectively. The enzymes could cleave β-carotene and β-apo-8'-carotenal to produce β-ionone.  相似文献

We consider the use of pulse-shaped broadband femtosecond lasers to optically cool rotational and vibrational degrees of freedom of molecules. Since this approach relies on cooling rotational and vibrational quanta by exciting an electronic transition, it is most easily applicable to molecules with similar ground and excited potential energy surfaces, such that the vibrational state is usually unchanged during electronic relaxation. Compared with schemes that cool rotations by exciting vibrations, this approach achieves internal cooling on the orders-of-magnitude faster electronic decay timescale and is potentially applicable to apolar molecules. For AlH(+), a candidate species, a rate-equation simulation indicates that rovibrational equilibrium should be achievable in 8 μs. In addition, we report laboratory demonstration of optical pulse shaping with sufficient resolution and power for rotational cooling of AlH(+).  相似文献

The helix-forming nature of β-1,3-glucan polysaccharides is a characteristic that has potential for producing gene carriers, bio-nanomaterials and other chiral nanowires. Herein, carboxylic curdlan (CurCOOH) bearing the β-1,3-polyglucuronic acid structure was successfully prepared from β-1,3-glucan polysaccharide curdlan (Cur) by one-step oxidation using a 4-acetamido-TEMPO/NaClO/NaClO(2) system as the oxidant. The resulting high-molecular-weight CurCOOH was proved to bear the 6-COOH group in 100% purity. The optical rotatory dispersion (ORD) spectra indicated that the obtained CurCOOH behaves as a water-soluble single-strand in various pH aqueous media. This advantage has allowed us to use CurCOOH as a polymeric host to form various macromolecular complexes. For example, complexation of CurCOOH with single-walled carbon nanotubes (SWNTs) resulted in a water-soluble one-dimensional architecture, which formed a dispersion in aqueous solution that was stable for several months, and much more stable than SWNTs complexes of the similar negatively-charged polyacrylic acid (PAA) and polymethacrylic acid (PMAA). It was shown that in the complex, SWNTs are effectively wrapped by a small amount of CurCOOH, enabling them to avoid electrostatic repulsion. This pH-responsive CurCOOH formed a very stable complex with cationic water-soluble polythiophenes (PT-1), which was stabilized not only by the hydrophobic interaction but also by the electrostatic attraction between trimethylammonium cations in PT-1 and dissociated anionic COO(-) groups in CurCOOH. The included PT-1 became CD-active only in the neutral to basic pH region, and the positive Cotton effect suggested that the conjugated main chain is twisted in the right-handed direction. We also found that CurCOOH can interact with polycytidylic acid (poly(C)) only under high NaCl concentrations, the binding and release of which could be controlled by a change in the salt concentration. We believe, therefore, that CurCOOH bearing a dissociable COOH group can act as a new potential polymeric host to construct novel polymeric complexes applicable for gene carriers, biosensors, chiral polymer assemblies, etc.  相似文献

[(Tosylamino)alkyl]naphthalenols have efficiently been synthesized by nucleophilic addition of naphthalen‐2‐ol with N‐tosyl imines (derived from both aromatic and aliphatic aldehydes) in the presence of BF3?OEt2 as a catalyst at room temperature. A novel counter-current chromatographic system is developed by mounting a vortex column on a type-I coil planet centrifuge. The column is fabricated from a high-density polyethylene disk (16 cm diameter and 5 cm thick) by making multiple holes of various diameters (3-12.5 mm) each arranged in a circle and connected with narrow transfer ducts. The performance of this vortex column is tested with three different two-phase solvent systems with a broad range in hydrophobicity. The results indicated that the smallest diameter column (3mm diameter, 120 units with 42.8 ml capacity) yielded the best separation with the height equivalent to a theoretical plate of 2 cm compared with 20 cm required by the conventional multilayer coil column of high-speed CCC. By avoiding the use of an Archimedean Screw Force, the system shows a low column pressure which would permit safe operation of a large preparative column without a risk of leakage of solvent and column damage.  相似文献

Abstract

The stability of parathion-methyl, methiocarb, DDT and 2,4-D, adsorbed on the polymeric PLRP-S phase packed in small stainless steel precolumns was examined, with a view to propose the use of these precolumns as alternative means for the transport of water samples. First, water samples spiked with the studied pesticide at low μg/l concentration levels were extracted and preconcentrated in the precolumns, using appropriate conditions for a total recovery. Then, the precolumns were stored at room temperature (15—20°C) or at 35°C for different time periods. At the end of the respective period each precolumn was coupled to an HPLC column via a switching valve and was on-line analyzed by reversed phase chromatography with UV detection. The four pesticide recoveries after one week in the precolumn at room temperature were higher than 90%. The same was true at 35°C except for DDT, which suffered a 30% degradation in one week. Further studies showed that DDT and parathion-methyl were stable at least for five weeks in precolumns stored at room temperature. Methiocarb also was stable for this period but storing the precolumns at 4°C.  相似文献
