The 'apparent' hydrolysis of alkyl esters during electrospray ionization

Electrospray ionization (ESI) mass spectrometry was employed to obtain both molecular weight confirmation and structural information for a series of novel alkenyldiarylmethane (ADAM) analogs. The mass spectral data were intended for use during the structure elucidation of ester hydrolysis products formed during an in vitro metabolism study of a series of novel ADAM analogs. The data on the precursor molecules show the presence of the molecular ion peak, [M+H](+), as well as a peak consistent with the hydrolysis product of the original ester ([MH-ROH+H(2)O](+)). However, chemical ionization mass spectrometry, elemental analysis and (1)H NMR data indicated the presence of only the intact diester compounds, suggesting that the formation of the hydrolysis product was an instrumental artifact, i.e., in-beam hydrolysis during ESI or a result of longer ion residence times of the ion trap mass analyzer.     

A conformationally flexible tetrahydroxycalix[4]arene adopting the unusual 1,3-alternate conformation

The use of a chiral solvating agent enabled the determination of the NMR-silent ring-inversion process of the ketocalixarene 3. Spectroscopic and crystal data indicate that 3 adopts the unusual 1,3-alternate conformation.     

Versatile and facile synthesis of diverse semisynthetic tetracycline derivatives via Pd-catalyzed reactions

A diverse collection of tetracycline derivatives has been synthesized utilizing Heck, Suzuki, and other palladium-coupling reactions via tetracycline arenediazonium and iodoarene salts. Large numbers of tetracyclines are now possible via these reactions, including numerous upper periphery derivatives of doxycycline, minocycline, sancycline, and methacycline modified at positions C7, C9, and C6-C13 on the tetracycline naphthacene ring. Application of palladium-coupling reactions to the tetracyclines has yielded new tetracycline classes with differing structural attributes, greatly increasing the structural diversity of this family of antibiotics, one of the last of the early antibiotic families to be expanded by organic and medicinal chemistry.     

Synthesis of bridged sugar amino acids: a new entry into conformationally locked δ- and ε-amino acids

The synthesis of three methylene bridged sugar amino acids is described. Key transformations in the synthetic strategy are a CO-insertion on fully protected ribose, an aldol condensation with formaldehyde and an oxetane forming cyclisation step. A novel Leu-enkephalin analogue containing the δ-SAA 1 was prepared using standard solution phase peptide chemistry.     

RESS for the preparation of fluorinated porphyrin nanoparticles

Rapid Expansion of Supercritical Solutions (RESS) was used to produce clean, surfactant-free nanoparticles (average size = 60 nm) of a fluorinated tetraphenylporphyrin from supercritical solutions with CO2.     

Beta-lactams or gamma-lactams by 4-exo-trig or 5-endo-trig anionic cyclisation of lithiated acrylamide derivatives

Substituted acrylamide derivatives of benzylamine are lithiated alpha to nitrogen by LDA. The benzyllithium thus formed undergoes either 5-endo-trig anionic cyclisation, formally by intramolecular conjugate addition to the acrylamide, to yield 5-membered lactams, or, if the acrylamide bears a beta-electron withdrawing group, 4-exo-trig cyclisation to a beta-lactam.     

Dyad beads and the combinatorial discovery of catalysts

Dyad beads, bearing both a substrate and a catalyst, were prepared to enable direct split and mix bead based screening for catalysis.     

Wavelength dependent photo-controlled differential release of compounds from solid phase resin

A method to effect photo-mediated differential release of bead-based compound libraries using a tuneable laser in combination with chromatically orthogonal photolabile linkers is described.     

Increasing the crystallisation temperature to access new spin clusters: conversion of [Ni8(cit)6(OH)2(H2O)2]10- to [Ni8(cit)6(OH)2]10-

The role of temperature in the formation of high nuclearity nickel(II) citrate spin clusters is explored, revealing how changes in structure and hence magnetic properties can be triggered through desolvation and ligand reorganisation.
