Assays were developed for DNA or polymyxin B (PMB) based on enhanced resonance Rayleigh scattering (RRS) and resonance nonlinear scattering (including second order scattering and frequency doubling scattering) that result from the interaction of PMB with DNA. A minor-groove binding mechanism is suggested from the results obtained with RRS and from absorption and circular dichroism spectroscopy. The types of interaction and reasons of RRS enhancement are discussed. Linear relationships do exist over a wide range between the intensity of enhanced scattering and the concentrations of either DNA or PMB. When ctDNA is used as a probe to determine PMB, the detection limit (3σ) is 9.8 ng mL?1. When PMB is used as a probe to determine DNA, the detection limit (3σ) is in the range from 3.8 to 9.0 ng mL?1.

In this article, we developed a native biofilm (NBF) bioreactor used for biochemical oxygen demand mediated method (BODMed). There were two innovations differed from previous BODMed assay. Firstly, the immobilization of microorganisms was adopted in BODMed. Secondly, the NBF was introduced for BOD measurement. The NBF bioreactor has been characterized by optical microscopy. A culture condition of NBF with 24 h, 35 °C and pH 7 was optimized. Furthermore, a measuring condition with 35 °C, pH 7 and 55 mM ferricyanide in 1 h incubation were optimized. Based on the optimized condition, the real wastewater samples from local sewage treatment plant had been measured. Performances of the NBFs proposed at different culture conditions were recorded for 110 d, and the results indicated that long-term storage stability was obtained. With the proposed method, an uncontaminated native microbial source solution can be obtained from a wastewater treatment plant. In this way, we can ensure that the microbial species of all in the NBF are same as that in the target to be measured.  相似文献

Gold nanoparticles-poly(luminol) (Plu-AuNPs) hybrid film and multi-walled carbon nanotubes with incorporated β-cyclodextrin modified glassy carbon electrode (β-CD-MWCNTs/Plu-AuNPs/GCE) was successfully prepared for simultaneous determination of dopamine (DA) and uric acid (UA). The surface of the modified electrode has been characterized by X-ray photo-electron spectroscopy (XPS), energy dispersive X-ray spectroscopy (EDS), field-emission scanning electron microscope (SEM) and transmission electron microscope (TEM). Cyclic voltammetry (CV), electrochemical impedance spectroscopy (EIS) and differential pulse voltammetry (DPV) have been used to investigate the β-CD-MWCNTs/Plu-AuNPs composite film. Gold nanoparticles anchored into poly(luminol) film exhibited catalytic activity for DA. MWCNTs with incorporated β-CD can greatly promote the direct electron transfer. In 0.10 M phosphate buffer solution (PBS, pH 7.0), the DPV response of the β-CD-MWCNTs/Plu-AuNPs/GCE sensor to DA is about 8-fold as compared with the Plu-AuNPs/GCE sensor, and the detection limit for DA is about one order of magnitude lower than the Plu-AuNPs/GCE sensor. The steady-state current response increases linearly with DA concentration from 1.0 × 10−6 to 5.6 × 10−5 M with a low detection limit (S/N = 3) of 1.9 × 10−7 M. Moreover, the interferences of ascorbic acid (AA) and uric acid (UA) are effectively diminished. The applicability of the prepared electrode has been demonstrated by measuring DA contents in dopamine hydrochloride injection.  相似文献

Several highly luminescent, aryl-substituted mesobenzanthrones (7H-benz[de]anthracen-7-ones) were prepared by a simple, two-step synthesis: addition of a carboxylated benzyne to a cyclopentadienone followed by an intramolecular Friedel-Crafts acylation. These compounds exhibit brilliant, yellow-green luminescence with quantum yields ranging from 0.01 to 1, depending on the aryl substituents present, and their photophysical behavior was elucidated by experimental and computational methods.  相似文献

为发现防治蚜虫的新型活性化合物,以蚜虫报警信息素E-β法尼烯(EBF)为先导,设计合成了20个未见文献报道的含吡唑环甲酰胺基EBF类似物,所有化合物结构均通过1H NMR,IR及HRMS确证.初步生物活性研究表明,部分目标化合物对五日龄豆蚜(Aphis craccivora Koch)在600mg/L时表现出较好的杀虫活性,其中5a及5k对豆蚜活性与商品化药剂氟虫腈及先导化合物EBF相当,甚至优于EBF.对影响目标化合物的活性因素进行了初步探讨.  相似文献

A mercury coordination polymer [Hg3(TizT)2I6]n (Mr = 1921.72, TizT = 2,4,6- tri(imidazole-1-yl)-1,3,5-triazine) containing a 40-membered macrocycle which was constructed by four TizT ligands and four mercury(II) iodide molecules had been synthesized by the reaction of HgI2 with TizT. The complex was characterized by elemental analysis, FT-IR, 1H NMR spectra and X-ray crystallography. The crystal of the complex belongs to the monoclinic system and C2/c space group with a = 35.840(5), b = 8.169(5), c = 14.980(5) , β = 104.466(5)°, Z = 4, V = 4247(3) 3, Dc = 3.006 g·cm-3, μ = 15.223 mm-1, F(000) = 3384, Rint = 0.0504, wR = 0.0833 and constructs a chair-like conformation of cyclohexane one by one, which forms a 1-D polymer through the fashion of fused ring aromatic hydrocarbon. The hydrogen bonds and π-π interactions shape the 2-D network structure. The two compounds excited weak fluorescence.  相似文献

The in vitro suitable action distance between umbilical cord blood-derived hematopoietic stem/progenitor cells and its feeder cell, human adipose-derived stem cells, during their co-culture, was investigated through a novel transwell co-culture protocol, in which the distance between the two culture chambers where each cell type is growing can be adjusted from 10 to 450 μm. The total cell number was determined with a hemacytometer, and the cell morphology was observed under an inverted microscope each day. After 7 days of co-culture, the fold-expansion, surface antigen expression of CD34(+) and CFU-GM assay of the hematopoietic mononuclear cells (MNCs) were analyzed. The results showed that there was an optimal communication distance at around 350 μm between both types of stem cells during their in vitro co-culture. By using this distance, the UCB-MNCs and CD34(+) cells were expanded by 15.1?±?0.2 and 5.0?±?0.1-fold, respectively. It can therefore be concluded that the optimal action distance between stem cells and their supportive cells, when cultured together for 7 days, is of around 350 μm.  相似文献

The great performance of a fibrous bed bioreactor (FBB) is mainly dependent on the cell adhesion and immobilization into the fibrous matrix. Therefore, understanding the mechanism and factors controling cell adhesion in the fibrous matrix is necessary to optimize the FBB setup and further improve the fermentability. The adhesion behavior of a strain of Clostridium tyrobutyricum isolated from an FBB was studied, which was proven to be affected by the different environmental conditions, such as growth phase of cells, pH, ionic strength, ionic species, and composition of media. Our results also suggested that electrostatic interactions played an important role on bacteria adhesion into the fibrous matrix. This study demonstrated that the compositions of fermentation broth would have a significant effect on cell adhesion. Consequently, a two-stage glucose supply control strategy was developed to improve the performance of FBB with higher viable cell density in the operation of the FBB setup.  相似文献

This work describes a novel methodology for unequivocal identification of chemical constituents in Zhimu‐Baihe herb‐pair (ZMBHHP). Compounds were removed from ZMBHHP by ultrasonic extraction with 70% ethanol, and then analyzed by fast high‐performance liquid chromatography (HPLC) coupled with time‐of‐flight mass spectrometry (TOFMS). The accurate‐mass capability of the TOF analyzer allowed reliable confirmation of the identity of the detected compounds, normally with mass errors below 3 ppm in routine analysis. This mass accuracy was sufficient to verify the elemental compositions of the chemical constituents in ZMBHHP. With dynamic adjustment of fragmentor voltage in TOFMS, an efficient ion transmission was achieved to obtain the best sensitivity and abundant fragmentation. By accurate mass measurements for each molecular ion and subsequent fragment ions, a reliable identification and differentiation of 24 saponins, 3 xanthones, 1 anthraquinone and 2 alkaloids was described here, including four groups of isomers. It is concluded that this fast and sensitive HPLC/ESI‐TOFMS technique is powerful in qualitative analysis of complex herbal medicines in terms of sensitivity, selectivity, resolving power, time savings and lower solvent consumption. Furthermore, the data gathered in this study may be helpful for understanding the synergistic nature of this herb pair in traditional Chinese medicine (TCM) and further pharmacokinetic studies of ZMBHHP.
