Thyroid autoimmunity in Graves’ disease (GD) is accompanied by Graves’ orbitopathy (GO) in 40% of the cases. Orbital fibroblasts (OF) play a key role in the pathogenesis and cigarette smoking is a known deteriorating factor. Alongside conventional cigarettes (CC) new alternatives became available for smokers, including heated tobacco products (HTP) and E-cigarettes (ECIG). We aimed to study the cellular effects of smoke extracts (SE) in orbital fibroblasts. Primary OF cultures from GO and NON-GO orbits were exposed to different concentrations of SE (1%, 50%) and the changes were followed using Real Time Cell Electronic Sensing (RT-CES). Untreated GO and NON-GO cells had different maximum cell index (CI) values of 3.3 and 2.79 respectively (p < 0.0001). CC, HTP and ECIG treated NON-GO fibroblasts exhibited peak CIs of 2.62, 3.32 and 3.41 while treated GO cells’ CIs were higher, 5.38, 6.25 and 6.33, respectively (p < 0.0001). The metabolic activity (MTT) decreased (p < 0.001) and hyaluronan production doubled (p < 0.02) after 50% of CC SE treatment in all cell cultures. GO fibroblasts were more sensitive to low concentration SE then NON-GO fibroblasts (p < 0.0001). The studied SEs exerted different effects. RT-CES is a sensitive technique to detect the effects of very low concentration of SE on fibroblasts.  相似文献

A modularly built bisubstrate inhibitor , the natural product pepticinnamin E (shown on the right) was sythesized for the first time. In the case of in vitro assays it inhibits the enzyme farnesyltransferase with respect to both the peptide substrate and farnesylpyrophosphate (KI = 30 and 8 μM , respectively). The inhibitory activity is decisively influenced by the central tripeptide unit and the absolute configuration of the non-proteinogenic amino acid incorporated therein.  相似文献

Steamed zeolites exhibit improved catalytic properties for hydrocarbon activation (alkane cracking and dehydrogenation). The nature of this practically important phenomenon has remained a mystery for the last six decades and was suggested to be related to the increased strength of zeolitic Bronsted acid sites after dealumination. We now utilize state-of-the-art infrared spectroscopy measurements and prove that during steaming, aluminum oxide clusters evolve (due to hydrolysis of Al out of framework positions with the following clustering) in the zeolitic micropores with properties very similar to (nano) facets of hydroxylated transition alumina surfaces. The Bronsted acidity of the zeolite does not increase and the total number of Bronsted acid sites decreases during steaming. O5Al(VI)-OH surface sites of alumina clusters dehydroxylate at elevated temperatures to form penta-coordinate Al1O5 sites that are capable of initiating alkane cracking by breaking the first C-H bond very effectively with much lower barriers (at lower temperatures) than for protolytic C-H bond activation, with the following reaction steps catalyzed by nearby zeolitic Bronsted acid sites. This explains the underlying mechanism behind the improved alkane cracking and alkane dehydrogenation activity of steamed zeolites: heterolytic C-H bond breaking occurs on Al-O sites of aluminum oxide clusters confined in zeolitic pores. Our findings explain the origin of enhanced activity of steamed zeolites at the molecular level and provide the missing understanding of the nature of extra-framework Al species formed in steamed/dealuminated zeolites.  相似文献

First, several reminiscences are recalled about the author's encounters with such a beautiful person as Professor Jean-Louis Calais. Since Jean-Louis was very much interested—among other things—in the correlation problem of solids, different methods are reviewed for the correlation correction of the energy bands of 1D and 2D periodic systems as well for the levels of 1D nonperiodic chains. The results obtained show a very good agreement with experiment for the fundamental gap and exciton spectra of different polymers, for the mechanical properties of polyethylene, as well for other not-here-described physical properties of different polymeric systems. A theoretical model, based on the binomial (Bernoullian) distribution function, was employed for the prediction of functional group distribution in an ethylene-propylene copolymer randomly grafted by maleic anhydride. Using the experimentally determined graft level and molecular weight distribution function, the fraction of polymer molecules with given number of functional groups was calculated. The result was checked experimentally by a fluorescence method based on the excimer formation of pyrene fluorophores attached to the anhydride pendants. The time-resolved fluorescence from the pyrene-labeled copolymer yielded the fraction of polymer molecules with a single functional group. The fraction of singly labeled molecules was compared to the calculated functional group distribution and a reasonable agreement was found between the two. The distribution of grafted maleic anhydride was found to be apparently random among polymer molecules. The distribution of distances was calculated between randomly attached consecutive functional groups along the polymer backbone also. The result indicated that the distance distribution function (similar to a decaying exponential) is dominated by short distances. © 1996 John Wiley & Sons, Inc.  相似文献
