Amphotericin B (AmB 1) is known to assemble together and form an ion channel across biomembranes. The antibiotic consists of mycosamine and macrolactone moieties, whose relative geometry is speculated to be determinant for the drug's channel activity and sterol selectivity. To better understand the relationship between the amino-sugar orientation and drug's activity, we prepared conformation-restricted derivatives 2-4, in which the amino and carboxyl groups were bridged together with various lengths of alkyl chains. K+ influx assays across the lipid-bilayer membrane revealed that ergosterol selectivity was markedly different among derivatives; short-bridged derivative 2 almost lost the selectivity, while 3 showed higher ergosterol preference than AmB itself. Monte Carlo conformational analysis of 2-4 based on NOE-derived distances indicated that the amino-sugar moiety of 2 comes close to C41 because of the short bridge, whereas those of 3 and 4 are pointing outward. The mutual orientation of the amino-sugar moiety and macrolide ring is so rigid in derivatives 2 and 3 that these conformations should be unchanged upon complex formation in lipid membranes. These results strongly suggest that the large difference in sterol preference between derivatives 2 and 3 is ascribed to the different orientation of amino-sugar moieties. These findings allowed us to propose a simple model accounting for AmB-sterol interactions, in which hydrogen bonding between 2'-OH of AmB and 3beta-OH of ergosterol plays an important role. 相似文献

Stabilities and structural fluctuations of both neutral and charged Au55 clusters are examined and discussed in relation to recent experimental observations of small gold particles with an electron microscope. Transition probabilities between the icosahedral and cuboctahedral structures are calculated according to the transition state theory using a model potential consisting of attractive many-body, repulsive pairwise and Coulomb parts. It is shown that for a neutral cluster the cuboctahedral structure has too short life time to be observed around room temperature and that, on the other hand, for more than 6-fold multiply charged clusters, both structures have life times of the order of 0.1 s around room temperature and, therefore, the transition between them can be observed. 相似文献

In this study, we aimed to isolate bacteria capable of degrading the polysaccharide ulvan from the green algae Ulva sp. (Chlorophyta, Ulvales, Ulvaceae) in marine environments. We isolated 13 ulvan-degrading bacteria and observed high diversity at the genus level. Further, the genera Paraglaciecola, Vibrio, Echinicola, and Algibacter, which can degrade ulvan, were successfully isolated for the first time from marine environments. Among the 13 isolates, only one isolate (Echinicola sp.) showed the ability not only to produce externally expressed ulvan lyase, but also to be periplasmic or on the cell surface. From the results of the full-genome analysis, lyase was presumed to be a member of the PL25 (BNR4) family of ulvan lyases, and the bacterium also contained the sequence for glycoside hydrolase (GH43, GH78 and GH88), which is characteristic of other ulvan-degrading bacteria. A simple Raman probe was realized using a single flexible hollow waveguide (HW). A HW coated with a silver film, which had reasonable transmission and little optical background noise, was used as a bidirectional transmission fiber for both the excitation and collection of Raman scattered light. The HW itself generated no Raman scattering or fluorescence noise during transmission. A complex filtering system at the end of the waveguide was thus unnecessary. In addition, the measured Raman spectra showed better signal-to-noise ratios than a conventional Raman fiber probe. The HW's suitability as a Raman fiber probe was also demonstrated. 相似文献

Ordering transitions are observed in azo‐dendrimer‐dissolved nematic liquid crystal (NLC) droplets dispersed in a glycerol matrix. The dendrimer molecules are spontaneously attached at the interface between NLC and glycerol, so that the nematic directors orient perpendicular to the interface, in the radial configuration. Photoisomerization makes the directors be tangential to the interface, in a photoinduced ordering transition from radial to bipolar structure. Similar experiments are conducted both in cholesteric (Ch) and smectic‐A (SmA) LC droplets. Complicated photoinduced ordering transitions are also observed in Ch and SmA droplets, and the associated molecular orientation changes are discussed. Photoisomerizable azo‐dendrimer molecules provide a possible way to control orientation, even in microdroplet systems in which the interface cannot be treated by conventional surface agents. 相似文献

Abstract Continuous flames have been observed in Supercritical water oxidation (scWO) of isopropyl alcohol (IPA), using a vertical continuous reactor with sapphire windows and a mixing nozzle. Two types of continuous flame were confirmed: the one was long pale blue colored and the other was red short cone shaped, changing blue to red at around air ratio 2.0. The flame was strongly influenced by IPA concentration, air ratio and design of the mixing nozzle. Results for decomposition of PA are presented for IPA concentrations ranging from 600 up to 28260 ppm as TOC and initial reactor temperatures, were mostly around 490°C, at 25 MPa. Decomposition rate at steady state was over 99.9%. Experimentally measured CO2 and O2 concentrations at the flue gas were in good agreement with theoretically calculated values. Even for low air ratio as 1.1, high decomposition rate without CO, NO, NO2 was achieved. 相似文献

This paper describes a curved field-sequential-color matrix display using fast-response ferroelectric liquid crystal. Black
matrix and transparent electrode patterns were formed on a thin plastic substrate by a transfer method from a glass substrate.
While a composite film of liquid crystal and micro-polymers of walls and fibers was formed between the flexible substrates
by printing, laminating and curing processes of a solution of monomers and liquid crystal, the mechanical stability was enhanced
by use of multi-functional monomers to form large display panels. The image pixels of the matrix panel were driven by an active
matrix scheme using an external switch transistor array at a frequency of 180 Hz for intermittent three-primary-color backlight
illumination. The flexible A4-paper-sized color display with 24 × 16 pixels and 60 Hz field frequency was demonstrated by
illuminating it with sequential three-primary-color lights from light-emitting diodes of the backlight. Our display system

We present the first study of the gluonic excitation in the three-quark (3Q) system in SU(3) lattice QCD with beta=5.8 and 16(3) x 32 at the quenched level. For the spatially fixed 3Q system, we measure the gluonic excited-state potential, which is responsible for the properties of hybrid baryons. The lowest gluonic-excitation energy in the 3Q system is found to be about 1 GeV in the hadronic scale. This large gluonic-excitation energy is expected to bring about the success of the simple quark model without gluonic modes. 相似文献
