经过数次技术研究和超常创新战略的大发展,生物催化逐渐达到工业化水平,从而受到人们特别的关注.基于酶值,通过生物途径生产高附加值化合物和精细工业化学品成为人们最感兴趣的领域之一.更广泛的众多生物化学路线可由酶催化来实现,其中还有一些酶尚未被人们发现.另一方面,由于非同源底物和某些化学过程所必需的苛刻条件,导致酶催化过程的效率低、稳定性差,因而限制了生物催化的应用.因此,开发具有多催化特征、更高效率和稳定性的绿色催化剂,成为生物催化的重中之重.计算科学、代谢工程、合成生物,以及机器学习路线的运用为新催化剂的工程化提供了新方法.本文重点介绍了合成生物学和代谢工程在催化中的作用,讨论了用于催化的机器学习算法和如何选择一种预测蛋白质-配体相互作用的算法;为了预测键合和催化功能,综述了分子对接的重要性;最后给出了结束语、未来挑战和前景展望.  相似文献

Launch power is limited to the milliwatt level by stimulated Brillouin scattering (SBS) in a single-channel, coherent fiber-optic network. Increasing the number of frequency-division multiplexed channels causes the power limit to decrease quickly to submilliwatt levels due to three-wave intermodulation to keep the signal-to-noise ratio from deteriorating significantly. As the number of channels increases, stimulated Raman scattering (SRS) begins to dominate. If a maximum of 0.5-dB depletion in the highest-frequency channel is allowed, SRS dominates when the number of multiplexed channels reaches about 300. Generally, the launch-power-limiting phenomenon is dependent on the number of channels being multiplexed.  相似文献

The petroleum ether extract of dried ground seeds of Piper nigrum Linn. and some column fractions of this extract were subjected to GC and GC-MS analysis, resulting in the identification of fourteen compounds (1-14) by using NIST Mass spectral search program 1998 and the Kovat's retention indices. Ten of the compounds (1, 2, 4-12) are reported for the first time from this plant. All the fractions showed insecticidal activity against the fourth instar larvae of Aedes aegypti and against the fourth instar larvae of Anopheles stephensi Liston, determined by the WHO method.  相似文献

Cancer remains a major global malaise requiring the advent of new, efficient and low‐cost treatments. Photodynamic therapy, which combines a photosensitizer and photons to produce cytotoxic reactive oxygen species, has been established as an effective cancer treatment but has yet to become mainstream. One of the main limitations has been the paucity of photosensitizers that are effective over a wide range of wavelengths, can exert their cytotoxic effects in hypoxia, are easily synthesized and produce few if any side effects. To address these shortfalls, three new osmium‐based photosensitizers (TLD1822, TLD1824 and TLD1829) were synthesized and their photophysical and photobiological attributes determined. These photosensitizers are panchromatic (i.e. black absorbers), activatable from 200 to 900 nm and have strong resistance to photobleaching. In vitro studies show photodynamic therapy efficacy with both red and near‐infrared light in normoxic and hypoxic conditions, which translated to good in vivo efficacy of TLD1829 in a subcutaneous murine colon cancer model.  相似文献

Analytical solutions for the displacement and stress fields due to a misfit particle in a host matrix can be difficult to obtain, especially for complex particle geometries. In this work, we present a numerical method for finding such fields in the case of infinitely-long particles. The method is based on discretizing the continuous misfit region between the particle and matrix into local misfit regions consisting of interstitial dislocation loops. The results presented here indicate very good agreement with analytical solutions and better convergence with increasing loop density.  相似文献

A phase inversion process was used to co-extrude cerium–gadolinium oxide (Ce0.9Gd0.1O1.95)/NiO–CGO dual-layer hollow fibres (HF), which were then sintered to form, respectively, the electrolyte and high porosity anode precursor of a solid oxide fuel cell (SOFC) with anode inner diameter of 0.8 mm. Graded CGO–lanthanum strontium cobalt ferrite (La0.6Sr0.4Fe0.8Co0.2O3) cathode layers were then painted onto the CGO electrolyte to form a micro-tubular HF-SOFC. With a carefully designed anode current collector, this produced maximum power densities of 1186–5864 W m? 2 at 450–570 °C. High magnification imaging analysis revealed large three-phase boundary regions within the anode, a dense electrolyte layer and clearly highlighted the multiple CGO–LSCF cermet and pure LSCF cathode layers. The performance of the HF-SOFC with a twenty millimetre active length showed no degradation after four thermal cycles between 300 °C and 570 °C.  相似文献

One way to profile complex mixtures for receptor affinity is to couple liquid chromatography (LC) on-line to biochemical detection (BCD). A drawback of this hyphenated screening approach is the relatively high consumption of sample, receptor protein and (fluorescently labeled) tracer ligand. Here, we worked toward minimization of sample and reagent consumption, by coupling nano-LC on-line to a light-emitting diode (LED) based capillary confocal fluorescence detection system capable of on-line BCD with low-flow rates. In this fluorescence detection system, a capillary with an extended light path (bubble cell) was used as a detection cell in order to enhance sensitivity. The technology was applied to a fluorescent enhancement bioassay for the acetylcholine binding protein, a structural analog of the extracellular ligand-binding domain of neuronal nicotinic acetylcholine receptors. In the miniaturized setup, the sensitive and low void volume LED-induced confocal fluorescence detection system operated in flow injection analysis mode allowing the measurement of IC50 values, which were comparable with those measured by a conventional plate reader bioassay. The current setup uses 50 nL as injection volume with a carrier flow rate of 400 nL/min. Finally, coupling of the detection system to gradient reversed-phase nano-LC allowed analysis of mixtures in order to identify the bioactive compounds present by injecting 10 nL of each mixture.  相似文献

This paper presents a novel approach for the automatic localization of pupil and iris. Pupil and iris are nearly circular regions, which are surrounded by sclera, eyelids and eyelashes. The localization of both pupil and iris is extremely important in any iris recognition system. In the proposed algorithm pupil is localized using Eccentricity based Bisection method which looks for the region that has the highest probability of having pupil. While iris localization is carried out in two steps. In the first step, iris image is directionally segmented and a noise free region (region of interest) is extracted. In the second step, angular lines in the region of interest are extracted and the edge points of iris outer boundary are found through the gradient of these lines. The proposed method is tested on CASIA ver 1.0 and MMU Iris databases. Experimental results show that this method is comparatively accurate.  相似文献
