Class I histone deacetylases, HDAC1, HDAC2, and HDAC3, represent potential targets for cancer treatment. However, the development of isoform-selective drugs for these enzymes remains challenging due to their high sequence and structural similarity. In the current study, we applied a computational approach to predict the selectivity profile of developed inhibitors. Molecular docking followed by MD simulation and calculation of binding free energy was performed for a dataset of 2-aminobenzamides comprising 30 previously developed inhibitors. For each HDAC isoform, a significant correlation was found between the binding free energy values and in vitro inhibitory activities. The predictive accuracy and reliability of the best preforming models were assessed on an external test set of newly designed and synthesized inhibitors. The developed binding free-energy models are cost-effective methods and help to reduce the time required to prioritize compounds for further studies.  相似文献

Synthesis of a new coumarin-triazole-based dye and its photophysical parameters such as absorption, fluorescence emission, and fluorescence quantum yield were investigated. Studies have shown that the present dye has symmetry with a mirror image, especially in the ethanol solvent, with respect to the absorption and fluorescence spectra. As a result of the UV-vis and fluorescence spectroscopy techniques used, it was determined that absorption and emission spectra were shifted to the red with increasing solvent polarity. In addition, the spectral data of the synthesized compound exhibited that the stokes shifts are small, usually less than 50 nm, and the quantum yields are significantly high. In accordance with the results obtained, it can be stated that this novel dye synthesized here can offer an insight into application in sensor applications as analytical or biosensors, optoelectronic devices, and medicine industry.  相似文献

Transition metal complexes of arginine (using Co(II), Ni(II), Cu(II) and Zn(II) cations separately) were synthesized and characterized by FTIR, TG/DTA‐DrTG, UV‐Vis spectroscopy and elemental analysis methods. Cu(II)‐Arg complex crystals was found suitable for x‐ray diffraction studies. It was contained, one mole CuII and Na+ ions, two arginate ligands, one coordinated aqua ligand and one solvent NO3? group in the asymmetric unit. The principle coordination sites of metal atom have been occupied by two N atoms of arginate ligands, two carboxylate O atoms, while the apical site was occupied by one O atom for CuII cation and two O atoms for CoII, NiII, ZnII atoms of aqua ligands. Although CuII ion adopts a square pyramidal geometry of the structure. CoII, NiII, ZnII cations have octahedral due to coordination number of these metals. Neighbouring chains were linked together to form a three‐dimensional network via hydrogen‐bonding between coordinated water molecule, amino atoms and O atoms of the bridging carboxylate groups. CuII complex was crystallized in the monoclinic space group P21, a = 8.4407(5) Å, b = 12.0976(5) Å, c = 10.2448(6) Å, V = 1041.03(10) Å3, Z = 2. Structures of the other metal complexes were similar to CuII complex, because of their spectroscopic studies have in agreement with each other. Copper complex has shown DNA like helix chain structure. Lastly, anti‐bacterial, anti‐microbial and anti‐fungal biological activities of complexes were investigated.  相似文献

A new series of benzimidazolone bridged triheterocyclic compounds bearing thiosemicarbazide, thiadiazole, triazole, moieties was synthesized and then screened for their in vitro urease, α-glucosidase, and acetylcholinesterase inhibition properties for the first time. All the synthesized compounds showed an outstanding urease inhibitory effect when compared with standards. Compounds 1 , 4 , 5b , 5d , 6b , 6d , 7b , and 7d showed significant acetylcholinesterase inhibitory activity with IC50 values between 7.32 ± 0.58 and 12.52 ± 0.13 μg/ml comparable to donepezil (15.12 ± 0.20 μg/ml). Compound 5c , having thiosemicarbazide moiety at the positions N-1 and N-3 of benzimidazolone nucleus, showed the highest α-glucosidase inhibitory activity (IC50 = 11.42 ± 0.11 μg/ml).  相似文献

In this study, nonperipherally tetra‐substituted ( 2 ), peripherally tetra‐substituted ( 3 ), and peripherally octa‐substituted ( 4 ) zinc(II) phthalocyanines were synthesized as sensitizers for dye‐sensitized solar cells (DSSCs) in which 3‐pyridin‐3‐ylpropoxy substituent acts as anchoring unit to bind TiO2 surface. The optical results indicated that there is an interaction between the dyes and the TiO2 surface. The photovoltaic performances of the DSSCs based on these dyes were found to depend on both the position and number of the substituents. Despite the more red‐shifted absorption, the DSSC based on 2 showed the conversion efficiency of 0.68%, which is lower than 1.36% and 0.92% for 3 and 4 , respectively, under one sun (AM 1.5G). The vertical orientation of the dye on TiO2 surface could be the main reason for the higher photovoltaic performance of complex 3 , which is beneficial for not only injecting the electrons into the conduction band of TiO2 but also reducing the charge recombination. Overall, these results demonstrate that the peripherally tetra‐substituted 3‐pyridin‐3‐ylpropoxy zinc(II) phthalocyanine complex ( 3 ) as a sensitizer can more efficiently utilize the photons in the red/near‐infrared region with respect to the other complexes studied.  相似文献

Thermally‐triggered shape memory polymers (SMPs) are smart materials, which are capable of changing their shapes when they are exposed a heat stimulant. Blending semi‐crystalline and elastomeric polymers is an easy and low‐cost way to obtain thermo‐responsive SMPs. In this work, novel poly(ethylene‐co‐1‐octene) (PEO) and poly(styrene‐b‐isoprene‐b‐styrene) (SIS) thermoplastic elastomer blends were prepared via melt blending method. The morphological, mechanical, rheological properties and shape memory behaviours of the blends were investigated in detail. In morphological analysis, co‐continuous morphology was found for 50 wt% PEO/50 wt% SIS and 60 wt% PEO/40 wt% SIS (60PEO/40SIS) blends. The shape memory analysis performing by dynamic mechanical analyzer showed that the 60PEO/40SIS blend also exhibited the optimum shape memory performance with 95.74% shape fixing and 98.98% shape recovery. Qualitatively shape memory analysis in hot‐water pointed out that the amount of semi‐crystalline PEO promotes shape fixing ability of the blends whereas SIS content enhances shape recovery capability. Although the SIS and PEO are immiscible polymers, the blends of them were exhibited good elastomeric properties with regard to tensile strength, toughness, and elongation at break.  相似文献

We consider the dynamic lot-sizing problem with finite capacity and possible lost sales for a process that could be kept warm at a unit variable cost for the next period t + 1 only if more than a threshold value Qt has been produced and would be cold, otherwise. Production with a cold process incurs a fixed positive setup cost, Kt and setup time, St, which may be positive. Setup costs and times for a warm process are negligible. We develop a dynamic programming formulation of the problem, establish theoretical results on the structure of the optimal production plan in the presence of zero and positive setup times with Wagner–Whitin-type cost structures. We also show that the solution to the dynamic lot-sizing problem with lost sales are generated from the full commitment production series improved via lost sales decisions in the presence of a warm/cold process.  相似文献

Suzuki coupling is one of the most powerful methods for the synthesis of biaryls. We have prepared and characterized a series of unsymmetrical sulfonated water-soluble Pd(II)-pyridyl imine complexes and investigated them as catalysts for the Suzuki cross-coupling reaction in water under microwave irradiation.
