Pouteria lucuma Ruiz and Pav., known as the ‘Gold of the Incas’ or ‘lucuma’, is a subtropical fruit belonging to the Sapotaceae family, with a very sweet flavor, used to prepare cakes, ice creams as well as in the baking and dairy industries. To date, the content of primary metabolites is known, but little information is reported about the composition in specialized metabolites. Moreover, no study is reported on skin which represent an important agricultural waste due to the high demand for lucuma. In order to have a preliminary metabolite profile of Pouteria lucuma, the extracts of pulp and skin have been analyzed by LC-ESI/LTQOrbitrap/MS/MS in negative ion mode. The careful analysis of the accurate masses, of the molecular formulas and of the ESI/MS spectra allowed to identify specialized metabolites belonging to phenolic, flavonoid and polar lipid classes. The LC-MS/MS analysis guided the isolation of compounds occurring in the pulp extract whose structures have been characterized by spectroscopic methods including 1D- and 2D-NMR experiments and ESI-MS analysis. Furthermore, the phenolic content of the extracts along with the antioxidant activity of extracts and isolated compounds was evaluated.  相似文献

The interaction with native DNA of a 2,7‐diazapyrenium‐based ligand 1 and its PtII rectangular metallacycle 2 is explored through circular and linear dichroism and fluorescence spectroscopies. The metal‐free ligand 1 binds through intercalation, with a binding constant of approximately 5×105 M ?1, whereas the metallacycle 2 binds and bends the DNA with a binding constant of 7×106 M ?1. PCR assays show that metallo‐supramolecular box 2 interferes with DNA transactions in vitro whereas the intercalator 1 does not. The metallacycle is active against four human cancer cell lines, with IC50 values ranging between 3.1 and 19.2 μM and shows similar levels of efficacy, but a different spectrum of activity, to cisplatin.  相似文献

Hyperpolarization and Cyclic Nucleotide (HCN) -gated channels represent the molecular correlates of the "funny" pacemaker current (I(f)), a current activated by hyperpolarization and considered able to influence the sinus node function in generating cardiac impulses. HCN channels are a family of six transmembrane domain, single pore-loop, hyperpolarization activated, non-selective cation channels. This channel family comprises four members: HCN1-4, but there is a general agreement to consider HCN4 as the main isoform able to control heart rate. This review aims to summarize advanced insights into the structure, function and cellular regulation of HCN channels in order to better understand the role of such channels in regulating heart rate and heart function in normal and pathological conditions. Therefore, we evaluated the possible therapeutic application of the selective HCN channels blockers in heart rate control.  相似文献

In this paper thermal and thermo-oxidative stability of nylon 6 based nanocomposites containing up to 5% by weight of fatty acid coated calcium carbonate (CaCO3) nanoparticles is studied. Thermal stability of compression-moulded samples was evaluated by thermogravimetric analysis (TG) under air and nitrogen atmosphere. Kinetic analysis of TG data was performed by using the Flynn-Wall-Ozawa method. Results show that the presence of coated nanoparticles adversely affects the thermal and thermo-oxidative stability of nylon 6. Kinetic analysis shows that a complex, multi-step decomposition process occurs. Moreover, the presence of nanoparticles do not affect the rate limiting step of nylon 6 decomposition in air, while under nitrogen atmosphere the decomposition process occurs through a diffusion-driven regime in presence of high amounts of CaCO3.  相似文献

The key structural factors underlying the unique black chromophore of eumelanin biopolymers have so far defied elucidation. Capitalizing on the ability of 1% polyvinylalcohol (PVA) to prevent pigment precipitation during melanogenesis in vitro, we have investigated the visible chromophore properties of soluble eumelanin‐like polymers produced by biomimetic oxidation of 5,6‐dihydroxyindole (DHI) and 5,6‐dihydroxyindole‐2‐carboxylic (DHICA) in 1% PVA‐containing buffer at pH 7. Upon dilution DHI‐eumelanin solutions exhibited almost linear visible absorbance changes, whereas DHICA‐eumelanin displayed a remarkable deviation from linearity in simple buffer, but not in PVA‐containing buffer. It is suggested that in DHICA polymers, exhibiting repeated interruptions of interring conjugation due to lack of planar conformations, the black chromophore is not due to an overlap of static entities defined intrinsically by the conjugation length across the carbon frame, but results largely from aggregation‐related intermolecular perturbations of the π‐electron systems which are extrinsic in character.  相似文献

A rapid analytical approach suitable to achieve a comprehensive characterization of the compounds present in the infusion prepared from the leaves of Byrsonima fagifolia Niedenzu (Malpighiaceae), a Brazilian plant used as an infusion to treat gastric disorders, was developed. The method was based on high-performance liquid chromatography coupled to electrospray negative ionisation multistage ion trap mass spectrometry (HPLC/ESI-ITMSn).The main ions in the ESI-ITMS spectra were attributed to a quinic acid core containing from one to five galloyl units. Quercetin derivatives containing one and two sugar moieties as well as galloyl esterification were also detected. These results indicated that HPLC/ESI-ITMSn is easily applicable to infusions of this plant and allows the rapid and direct identification of these compounds in crude plant extracts.  相似文献

A fast and reliable method, based on high-performance liquid chromatography coupled to electrospray ionization ion trap tandem mass spectrometry (HPLC/ESI-ITMS), was developed to investigate the infusion prepared from the leaves of Byrsonima crassa Niedenzu (Malpighiaceae), a native plant used in Brazil against gastric disorders. The use of on-line reverse-phase HPLC/ESI-ITMS allowed separation of three major classes of compounds and identification of over 20 very polar compounds characterized as galloylquinic acids, proanthocyanidins, and flavonoid glycosides, as well as the dimeric flavonoid amentoflavone and minor amounts of galloyl hexose and galloyl saccharose. This approach provided data that will allow establishment of a method for a future standardization of the infusion.  相似文献

We report on the experimental demonstration of a fast reconstruction method for multiview fluorescence diffuse optical tomography by using a wavelet-based data compression. We experimentally demonstrate that the use of data compression combined with the multiview approach makes it possible to perform a fast reconstruction of high quality. A structured illumination approach, guided by the compression scheme, has been adopted to further reduce the acquisition time. The reconstruction algorithm is based on the finite element method, and hence is suitable for samples of any arbitrary shape.  相似文献

The observed 511 keV line from the Galactic Bulge is a real challenge for theoretical astrophysics: despite a lot of suggested mechanisms, there is still no convincing explanation and the origin of the annihilated positrons remains unknown. Here we discuss the possibility that a population of slowly evaporating primordial black holes with the mass around 1016–1017 g ejects (among other particles) low-energy positrons into the Galaxy. In addition to positrons, we have also calculated the spectrum and number density of photons and neutrinos produced by such black holes and found that the photons are potentially observable in the near future, while the neutrino flux is too weak and below the terrestrial and extra-terrestrial backgrounds. Depending on their mass distribution, such black holes could make a small fraction or the whole cosmological dark matter.  相似文献
