A highly specific polyclonal antiserum to the environmental contaminant 1,1,1-trichloro-2,2-bis-(4-chlorophenyl)-ethane (p,p′-DDT)

A very selective polyclonal antiserum against 1,1,1-trichloro-2,2-bis-(4-chlorophenyl)-ethane (p,p′-DDT) was obtained by a careful choice of the haptenic structure (2,2-bis(4-chlorophenyl)-ethanol hemisuccinate). This hapten was conjugated to BSA to prepare the immunogen. The effects of different types of solid phases on the equilibrium reaction between the hapten on solid phase and the polyclonal antiserum were evaluated to obtain a fine tuning of the antiserum performances in terms of specificity for p,p′-DDT and sensitivity to low levels of this pesticide. The calibration curves obtained show that it is possible to set up a sensitive assay for p,p′-DDT, employing a p,p′-dichlorodiphenylacetic acid-based solid phase, with a detection limit of 0.12 ng/mL and a working range of about 0.21–40 ng/mL. Selectivity towards several p,p′-DDT-related substances was good (o,p-DDT 17%, p,p′-DDD 1.2% o,p-DDD 6.3%, p,p′-DDE 6.7%).

Cooperative effects in phospholipid monolayers induced by a peptide from HIV-1 capsid protein

The study of interactions between biological molecules and model membranes is essential for the understanding of a number of physiological mechanisms involved in viral infections and dissemination. In this paper, the analysis of the interaction between a peptide from the p24 protein of Human Immunodeficiency Virus type 1 (HIV-1) and a phospholipid monolayer has pointed to a cooperative response in which very small amounts of peptide p24-1 (e.g. 0.05 mol%) can lead to measurable effects. Monolayer surface pressure and surface potential isotherms were affected for peptide concentrations as low as 0.05 mol%, with saturation at 0.5 mol%. The expansion effect from p24-1 is confirmed by changes in morphology of the monolayers using Brewster angle microscopy. Even though p24-1 is disordered in aqueous solutions, the interaction with dipalmitoyl phosphatidylcholine (DPPC) causes it to adopt an alpha-helix structure, as shown by circular dichroism (CD) data for multilamellar vesicles (MLV). The expansion of the phospholipid monolayer in a cooperative way may imply that p24-1 has potential antiviral activity, by participating in the cell rupture, with no need of specific receptors in the membrane.     

Simultaneous determination of two components by spectrofluorimetric techniques

A procedure is described for the resolution of two-component mixtures by use of data from variable-angle fluorescence spectrometry. The experimental data are treated by multilinear regression. The method is applied to the resolution of mixtures of fluorescein and dichlorofluorescein at the ng ml^?1 level; mean errors are < 4%.     

Syntheses and Structures of Heterometallic Fe–Ta Chalcogenido Clusters

Two Fe–Ta containing sulfido complexes were prepared by the reaction of the metal halide salts with bis-trimethylsilylsulfide in the presence of PMe₃. The complexes demonstrate that coordination chemistry with iron sulfides can give access to a range of heterometallic complexes. In [Cl(Me₃P)Ta( ₂-S)₂( ₃-S)Fe(PMe₃)₂]₂ the two [Cl(Me₃P)Ta] units are arranged around one central Fe₂( ₂-S)₂ unit. In [(Me₃P)₄(MeCN)₂Fe^II]²⁺[(Me₃P)₂Ta^IVFe^II ₃( ₃-S)₄Br₄]^2– a [TaFe₃S₄]²⁺ cuboidal arrangement was observed. The complex salt forms a polymeric structure in the solid-state with weak H-bonds between the ions. The [(Me₃P)₂Ta^IVFe^II ₃( ₃-S)₄Br₄]^2– ion was characterised by magnetic measurements showing strong antiferromagnetic interactions between the metal centres.     

Synthesis of the ABC ring system of manzamine A

A synthesis of the core ABC ring system of the manzamine alkaloids is described, starting from arecoline. The key steps involve a Claisen rearrangement to set up a 4-substituted-3-methylenepiperidine and a stereoselective azomethine ylide dipolar cycloaddition reaction. Condensation of the aldehyde 6 and sarcosine ethyl ester hydrochloride salt gives an intermediate azomethine ylide, which undergoes an intramolecular cycloaddition reaction to set up two new rings and three new chiral centers stereoselectively. The aldehyde 6 was not a suitable substrate for related azomethine ylide cycloaddition reactions with other amines. However, the related dimethyl acetal 26 could be condensed with a variety of amines to give the desired tricyclic products. The cycloaddition reaction with N-methyl or N-allyl glycine ethyl ester gave almost exclusively the exo adduct, whereas cycloaddition with glycine ethyl ester gave the endo adduct.     

Zum Nachweis der Oxyde von Molybdän,Wolfram und Vanadin

Zusammenfassung Es wird eine sehr empfindliche Reaktion zum Nachweis der Oxyde des Molybdäns, Wolframs und Vanadins angegeben. Sie beruht auf der Abscheidung von braunem Thallium(III)-oxydhydrat, aus ammoniakalischer Lösung von Thallium(I)-salzen auf Zusatz von Wasserstoffsuperoxyd und den oben genannten Oxyden, die hiebei als Katalysatoren wirken.Herrn Prof. Dr.Emil Abel, dem österreichischen Altmeister der Katalysenforschung, zum 80. Geburtstag.
