On the basis of a biogenetic proposal for explaining the biogenesis of manzamine A, the cycloaddition of dihydropyridinium salt 26 with diene derivative 5 leads to adducts 27. These adducts, as well as their related and previously described analogues 9, are now shown to be precursors of diene derivatives such as 10, 13, and 28. Treatment of diene 32 with sodium azide resulted in a one-step formation of the tricyclic imino derivative 34. This key intermediate was further transformed into tricyclic derivative 40, which possesses the essential features of the ABC ring of manzamine A.  相似文献

This work presents a method for extracellular and intracellular pH measurements in live cells based on a combination of the bead injection (BI) technique and fluorescence microscopy. For extracellular pH measurement, cells are grown on fluorescent beads, packed into a small column by a sequential injection instrument, and fluorescence intensity from the beads stained by the indicator is recorded by a fluorescence microscope. The method is applied to quantifying carbachol stimulation of Chinese hamster ovary (CHO) cells transfected with the m1 muscarinic receptor and is verified by a glucose depletion experiment. The results yield an EC(50) value of 1 muM for carbachol, which is in reasonable agreement with the literature value 3 muM determined by an existing potentiometric technique for measuring acid release. The intracellular measurement utilizes CHO M1 cells growing on non-fluorescent beads. For this method the cells rather than the beads are stained by incubating them in a solution of the fluorescent pH indicator BCECF. The cells are also stimulated with carbachol and the intracellular pH dependent fluorescence from the cells is recorded. The results show dependence between intracellular pH changes and carbachol concentration and yield an EC(50) value of 4 muM.  相似文献

Stable trichloro-octadecyl silane (ODS) derivatives of a 5 nm γ-alumina ceramic membrane were prepared. Gas permeabilities of the untreated membrane did not show Knudsen diffusion at 20°C. Gas permeabilities of the ODS membrane were three orders of magnitude lower; He, Ne, Ar, CO2, C3H8 have near constant permeabilities 360 mol s−1 m−2 bar−1, except methane which has the highest permeability of the group, 481 mol s−1 m−2 bar−1. The mechanism of diffusion is solution/diffusion. Remarkably, permeabilities of ODS-alumina membrane were reduced by 5 X after exposure to a pressure difference of 1 atm (active layer side) against vacuum for only 10 min. The effect was metastable but could be reversed on standing for several hours, reversal of pressure difference or after washing with (hydrocarbon solvent) toluene. The mechanism was presumed to be due to movement of the octadecyl-hydrocarbon chains of the silane monolayer causing a partially blocked pore structure; perhaps a unique example of self-fouling.  相似文献

The electron impact ionization mass spectra of 2,4,5,5-tetrasubstituted 1,2,4-triazolidine-3-thiones studied confirmed that the substituent at position 4 has the most dramatic influence on the fragmentation pattern. When the substituent is a methylallyl group the molecular ions exhibit four main routes of fragmentation, but when it is an ethoxycarbonyl/acetyl or a methyl group these direct decompositions of the molecular ion become less abundant. Interestingly all 4-ethoxycarbonyl derivatives and the 4-acetyl derivative exhibited the ions [M-R4-COOC2H4]+ and [M-R4-COCH2]+, respectively, with the same composition.  相似文献

Free radicals are generated in liquid solutions by harmonically modulated photolysis of suitable substrates. Harmonic analysis of the absorbance as functions of wavelength and modulation frequency yields the optical spectra and the decay kinetics of the transient species. The experimental technique and the analysis are described in detail. Results on t-butyl, 2-propyl and benzyl radicals generated by photolysis of the corresponding dialkyl resp. dibenzyl ketones are reported. They confirm previous spectral assignments and show that the termination reactions are diffusion controlled.  相似文献

A unique process of chemical oxidation polymerization of aniline using manganese dioxide (MnO2) as the oxidizing agent in an aqueous medium is described. The reaction between aniline and MnO2 follows a mechanism by which the organic monomer is oxidized while the metal oxide undergoes reductive dissolution. The effects of the amount of oxidizing agent and aniline, pH and temperature of the reactive system, type of acid on the yield and conductivity of polyaniline are discussed. The resulting polyaniline was characterized by [R and UV-Vis spectrometry. Polyaniline with a conductivity of 12.5 S/cm was obtained using 0.033 tool of aniline oxidized by 0.023 tool MnO2 in the presence of 100 mL of 2.7 mol/L HCI at 25℃ for 4 h.  相似文献

Gaseous protonated aziridine ions are produced at the threshold from β-phenoxyethylamine molecular ions. The evidence for this is collisional activation spectra, using various precursors (including labelled analogues) under electron impact and field ionization conditions. Partial conversion to the acyclic \documentclass{article}\pagestyle{empty}\begin{document}$ {\rm CH}_{\rm 3} {\rm CH = }\mathop {\rm N}\limits^ + {\rm H}_{\rm 2} $\end{document} isomer occurs at higher electron energies and is rationalized by means of a potential energy surface constructed from energetic data.  相似文献

The low-energy collision-activated dissociation of symmetrical n-butyl-substituted and of allyl-substituted onium cations has been recorded using fast atom bombardment ionization and a tandem mass spectrometry quadrupole mass spectrometer. Structure of the fragments and decomposition pathways have been ascertained using a multi-quadrupole MS/MS/MS triple analyser instrument. Whereas most sulphonium cations exhibit only heterolytic cleavages, fragmentation of ammonium and phosphonium is mainly homolytic. Allylic sulphonium, and to a lesser extent allylic ammonium, easily undergo propene elimination. This reaction, which does not occur in 1-propenyl oniums, might involve a McLafferty type rearrangement.  相似文献

A method is presented for constructing diabatic potential energy matrices from ab initio quantum chemistry data. The method is similar to that reported previously for single adiabatic potential energy surfaces, but correctly accounts for the nuclear permutation symmetry of diabatic potential energy matrices and other complications that arise from the derivative coupling of electronic states. The method is tested by comparison with an analytic model for the two lowest energy states of H(3).  相似文献

(S)-5,5,5,5′,5′,5′-Hexafluoroleucine ((S)- 13 ) of 81 % ee is prepared from hexafluoroacetone ( l ) and ethyl bromopyruvate (= ethyl 2-oxopropanoate) in 7 steps with an overall yield of 18% (Schemes 1 and 2). Key step in this sequence is the highly enantioselective reduction of the carbonyl group in α-keto ester 4 either by bakers' yeast (91 % ee) or by ‘catecholborane’ 6 utilizing an oxazaborolidine catalyst, yielding hydroxy ester (R)- 5 with 99% ee. The absolute configuration was determined by X-ray analysis of the HCl adduct (S,R)- 9b of (2S)-N-[(R)- l-phenylethyl]-5,5,5,5′,5′,5′-hexafluoroleucine ethyl ester.  相似文献
