The first total synthesis of (+/-)-ingenol has been achieved. The key features of the synthesis include the use of a highly diastereoselective Michael reaction to fix the C-11 methyl stereochemistry and the incorporation of the dimethylcyclopropane via diastereoselective carbene addition to the Delta13,14 olefin. The intramolecular dioxenone photoaddition-fragmentation sequence leads to the establishment of the critical C-8/C-10 trans intrabridgehead stereochemistry, a central challenge in the synthesis of ingenanes. The completion of the synthesis proceeds using the C-6alpha hydroxymethyl group as the sole handle for oxidation of seven contiguous carbon centers. 相似文献

There is a great demand for development of catalyst systems that are not only efficient and highly enantioselective but are also environmentally benign. Herein we report investigations into the catalytic asymmetric addition of alkyl and functionalized alkyl groups to ketones under highly concentrated and solvent-free conditions. In comparison with standard reaction conditions employing toluene and hexanes, the solvent-free and highly concentrated conditions permit reduction in catalyst loading by a factor of 2- to 40-fold. These new conditions are general and applicable to a variety of ketones and dialkylzinc reagents to provide diverse tertiary alcohols with high enantioselectivities. Using cyclic conjugated enones, we have performed a tandem asymmetric addition/diastereoselective epoxidation using the solvent-free addition conditions followed by introduction of a 5.5 M decane solution of tert-butyl hydroperoxide (TBHP) to generate epoxy alcohols. This one-pot procedure allows access to syn epoxy alcohols with three contiguous stereocenters with excellent enantio- and diastereoselectivities and high yields. Both the solvent-free asymmetric additions and asymmetric addition/diastereoselective epoxidation reactions have been conducted on larger scale (5 g substrate) with 0.5 mol % catalyst loadings. In these procedures, enantioselectivities equal to or better than 92% were obtained with isolated yields of 90%. The solvent-free and highly concentrated conditions are a significant improvement over previous solvent-based protocols. Further, this chemistry represents a rare example of a catalytic asymmetric reaction that is highly enantioselective under more environmentally friendly solvent-free conditions. 相似文献

Magnesium isotopes effects were investigated by chemical ion exchange using synthesized 2-aminomethyl-18-crown-6 (AM18C6) bonded Merrifield peptide resin. It was found that separation factors larger those reported previously were obtained, and the hydration and isotope mass effects are more significant than that of the complexation. The capacity of the crown ion exchanger was 2.3 meq/g dry resin. The adsorption capacity of the resin for magnesium ion was 26.8 mg/g dry resin at pH 7. The heavier isotopes of magnesium were enriched in the solution phase, while the lighter isotopes were enriched in the resin phase. The separation factors of (24)Mg-(25)Mg, (24)Mg-(26)Mg, and (25)Mg-(26)Mg were 1.0085, 1.0162, and 1.0081, respectively. 相似文献

An ongoing pandemic of coronavirus disease 2019 (COVID-19) is now the greatest threat to global public health. Herbal medicines and their derived natural products have drawn much attention in the treatment of COVID-19, but the detailed mechanisms by which natural products inhibit SARS-CoV-2 have not been elucidated. Here, we show that platycodin D (PD), a triterpenoid saponin abundant in Platycodon grandiflorum (PG), a dietary and medicinal herb commonly used in East Asia, effectively blocks the two main SARS-CoV-2 infection routes via lysosome- and transmembrane protease serine 2 (TMPRSS2)-driven entry. Mechanistically, PD prevents host entry of SARS-CoV-2 by redistributing membrane cholesterol to prevent membrane fusion, which can be reinstated by treatment with a PD-encapsulating agent. Furthermore, the inhibitory effects of PD are recapitulated by the pharmacological inhibition or gene silencing of NPC1, which is mutated in patients with Niemann–Pick type C (NPC) displaying disrupted membrane cholesterol distribution. Finally, readily available local foods or herbal medicines containing PG root show similar inhibitory effects against SARS-CoV-2 infection. Our study proposes that PD is a potent natural product for preventing or treating COVID-19 and that briefly disrupting the distribution of membrane cholesterol is a potential novel therapeutic strategy for SARS-CoV-2 infection.Subject terms: Viral infection, Lipid signalling 相似文献

The magnesium isotope effects were investigated by chemical ion exchange with a hydrous manganese(IV) oxide. The capacity of manganese(IV) oxide was 0.5 meq g(-1). The distribution coefficient of magnesium ions on the MnO(2) was determined by a batch method. The heavier isotopes of magnesium were enriched in the solution phase, while the lighter isotopes were enriched in the hydrous MnO(2) phase. The separation factor was determined according to the method of Glueckauf from the elution curve and isotopic assays. The separation factors of (24)Mg(2+)-(25)Mg(2+), (24)Mg(2+)-(26)Mg(2+), and (25)Mg(2+)-(26)Mg(2+) isotope pair fractionations were 1.011, 1.021, and 1.011, respectively. 相似文献

Linear and hyperbranched poly(ether‐ketone)s (PEKs) containing flexible oxyethylene spacers grafted multiwalled carbon nanotube (PEK‐g‐MWNT) nanocomposites were prepared by direct Friedel‐Crafts acylation as the polymer forming and grafting reaction. To achieve the composites, in situ polycondensations of AB monomers 3‐(2‐phenoxyethoxy)benzoic acid (3‐PEBA) and 4‐(2‐phenoxyethoxy)benzoic acid (4‐PEBA), and AB2 monomer 3,5‐bis(2‐phenoxyethoxy)benzoic acid (3,5‐BPEBA) were carried out in the presence of multiwalled carbon nanotubes (MWNTs). The reaction conditions, polyphosphoric acid (PPA) with additional phosphorous phentoxide (P2O5) in the temperature range of 110–120 °C, were previously optimized. The conditions were used as the polymerization and grafting medium that were indeed benign not to damage MWNTs but strong enough to promote the covalent attachment of PEKs onto the surface of the electron‐deficient MWNTs. From scanning electron microscopy (SEM) and transmission electron microscopy studies, the polymers were uniformly grafted onto the MWNTs. The resultant nanocomposites are soluble in most strong acids such as trifluoroacetic acid, methanesulfonic acid, and sulfuric acid. Both isothermal and dynamic TGA studies in air showed that nanocomposites displayed improved thermo‐oxidative stability when compared with those of corresponding PEK homopolymers. © 2008 Wiley Periodicals, Inc.
