The dispersion of results from proficiency tests for the analysis of pesticide residues in foodstuffs suggests that improvements in the compatibility of measurement results are needed. Currently observed divergences can make the evaluation conclusion on foodstuffs compliance with certain legislation dependent on the consulted laboratory. This work discusses the origin and impact of this lack of compatibility, following the metrological concepts presented at the latest version of the “International Vocabulary of Metrology” (VIM3), thus allowing for a clear diagnostic of the problem. The reporting of results from different measurement methods uncorrected for the observed analyte recovery makes them traceable to different “operationally defined measurement procedures” (VIM3) and, therefore, not comparable. When results from different measurement methods are reported corrected for analyte recovery, R, and R is different for spiked and incurred residues, measurement results may be not compatible if this effect is not considered on the uncertainty budget. This discussion is illustrated with metrological models for any possible combination of “measurement performance” and “maximum residue level”. These models are complemented with experimental data of the analysis of pesticide residues in a sample of ginseng powder from a proficiency test. The adopted experimental design allowed the identification of additional threats to metrological compatibility in this field. Solutions to the faced problem are discussed for practicability and impact on regulatory issues. The use of a universal “reference measurement procedure” proves to be the most feasible way of ensuring comparability of measurements in this field.  相似文献

Background

Cigarette smoking enhances the risk of stroke. However, the underlying molecular mechanisms are largely unknown. The present study established an in vivo rat secondhand cigarette smoking (SHS) model and examined the hypothesis that SHS upregulates endothelin receptors with increased cerebrovascular contraction via the Raf/extracellular signal-regulated kinase (ERK)/mitogen-activated protein kinases (MAPK) pathway.

Results

Rats were exposed to SHS for up to 8 weeks. The cerebral artery vasoconstriction was recorded by a sensitive myograph. The mRNA and protein expressions for endothelin receptors in cerebral arteries were studied by real-time PCR and Western blot. Compared to fresh air exposed rats, cerebral arteries from SHS rats exhibited stronger contractile responses (P < 0.05) mediated by endothelin type A (ETA) receptors. The expressions of mRNA and protein for ETA receptors in the cerebral arteries from SHS rats were higher (P < 0.05) than that in control. SHS did not affect endothelin type B (ETB) receptor-mediated contractions, mRNA or protein levels. The results suggest that SHS upregulates ETA, but not ETB receptors in vivo. After SHS exposure, the mRNA levels of Raf-1 and ERK1/2, the protein expression of phosphorylated (p)-Raf-1 and p-ERK1/2 were increased (P < 0.05). Raf-1 inhibitor, GW5074 suppressed the enhanced ETA receptor-mediated contraction, mRNA and protein levels induced by SHS. In addition, GW5074 inhibited the SHS-caused increased mRNA and phosphorylated protein levels of Raf-1 and ERK1/2, suggesting that SHS induces activation of the Raf/ERK/MAPK pathway.

Conclusions

SHS upregulates cerebrovascular ETA receptors via the Raf/ERK/MAPK pathway, which provides novel understanding of mechanisms involved in SHS-associated stroke.  相似文献

New broadside-coupled coplanar waveguides (CPW) having a completely asymmetrical structure, suitable for hybrid and monolithic microwave and millimeter-wave integrated circuits requiring broad bandwidths as well as loose and tight couplings, is proposed and examined for the first time. The results of the inivestigation, performed based on the spectral-domain analysis, demonstrate the versatility of the asymmetrical broadside-coupled CPW as compared to its symmetrical counterpart.  相似文献

An experimental study of heat transfer during quenching of a cylindrical stainless steel test specimen has been performed. A subcooled water jet is directed onto the upward facing flat face of the cylinder. The test specimen is heated to an initial temperature slightly above 900 °C and then quenched. The resulting boiling curves and heat transfer distributions are presented for impingement velocities of 2.85 and 6.4 m/s (Re = 7900 and 18,900). High-speed imaging shows that three distinct regions on the quenched surface can be identified: an expanding circular wetted region surrounding the impinging point, annular transition zone just outside the wetting front, and a unwetted region outside this zone. The free-surface of the liquid in the wetted region is smooth in the nucleate and transition boiling regimes. The annular transition zone or the wetting front region outside the wetted region is characterized by a highly disturbed liquid-gas interface, which can be attributed to intense vapor generation. At the outer edge of the transition zone, the liquid is deflected away from the surface. The velocity of the wetting front significantly increases with the jet impact velocity, which indicates that the wetting front position is governed by the ability of the flowing liquid to transport the bubbles radially outwards from the wetted region.  相似文献

The S-H bond dissociation enthalpies [BDE(S-H)] of a set of 5-X- and 6-X-3-pyridinethiols (X = F, Cl, CH3, OCH3, NH2, N(CH3)2, CF3, CN, and NO2) have been computed using the density functional theory based (RO)B3LYP procedure with 6-311++G(2df,2p) basis set. The effects of substituents on the BDE(S-H), proton affinity of the pyridinethiol anion [PA(S-)] and ionization energy (IE) are analyzed and their correlations with Hammett's substituent constants are examined. Subsequently, a series of 6-substituted 3-pyridinethiols have been explored to find out their antioxidant potentials. Finally, a number of 3-pyridinethiol based compounds are theoretically proposed as novel antioxidants.  相似文献

Surface‐enhanced Raman scattering (SERS) is a technique capable of identifying each component in a mixture because of its intrinsically narrow spectral bands. In a clinical setting, the identification of bacteria from its initial culture by collecting the colonies on the culture plate significantly decreases the analysis time and the cost. The identification of bacteria from their mixtures is attempted using SERS. A simple mixing procedure of bacterial samples and concentrated colloidal suspension is proven to be mostly satisfactory for the generation of the reproducible SERS spectra that can be used for bacterial identification. The mixture of three different but related bacterial species Shigella sonnei, Proteus vulgaris, and Erwinia amylovara and three Escherichia coli strains (BFK13, BHK7, DH5 α) are used as model systems to test the feasibility of the approach. The results indicate that it is possible to identify the composition of a bacterial mixture. Ion chromatography (IC) presents new possibilities for assessing information about environmental samples, namely waters of various compositions, ranging from high-purity water to highly saline ones. Constant proportion between major ions present in seawater, has been assumed in the past, from which the first practical equation relating chlorinity and salinity has been developed, being later substituted by a practical salinity scale, derived from conductivity measurements relative to a standard seawater, according to internationally accepted recommended procedures. Seawaters are characterized by salinity values around 35 while derived saline solutions may present considerable changes in ionic composition, conductivity, hence on salinity. Natural and anthropogenic phenomena may introduce new issues requiring clarification for which qualitative and quantitative information from additional sources is useful, e.g. ionic composition from IC. The different ranges of concentration of major and minor species present in seawater and derived saline solutions are a challenge for the optimization of a practical methodology for composition assessment in two single IC runs, one for anions and another one for cations, which has been attained in this work. Composition of saline solutions determined by IC was critically assessed in terms of anion-cation balance and further related to conductivity and salinity measurements aiming to evaluate the quality/completeness of ion chromatographic analyses performed at preselected conditions and to search for other meaningful relations for efficient recognition/distinction between saline solutions of different types.  相似文献
