The interaction of human galectin-1 with a variety of oligosaccharides, from di-(N-acetyllactosamine) to tetra-saccharides (blood B type-II antigen) has been scrutinized by using a combined approach of different NMR experiments, molecular dynamics (MD) simulations, and isothermal titration calorimetry. Ligand- and receptor-based NMR experiments assisted by computational methods allowed proposing three-dimensional structures for the different complexes, which explained the lack of enthalpy gain when increasing the chemical complexity of the glycan. Interestingly, and independently of the glycan ligand, the entropy term does not oppose the binding event, a rather unusual feature for protein-sugar interactions. CLEANEX-PM and relaxation dispersion experiments revealed that sugar binding affected residues far from the binding site and described significant changes in the dynamics of the protein. In particular, motions in the microsecond-millisecond timescale in residues at the protein dimer interface were identified in the presence of high affinity ligands. The dynamic process was further explored by extensive MD simulations, which provided additional support for the existence of allostery in glycan recognition by human galectin-1.  相似文献

We present a microeconomic model of social stratification, which includes an endogenous fertility component. In the model, egalitarian and stratified societies coexist. The latter are divided into 2 hereditary classes: a warrior elite and a productive class. The model entails that the extra cost warriors must incur to train and equip their children for war determines the relative sizes of both classes and the degree of economic inequality. Higher costs of warrior children imply a greater economic advantage for warriors and a smaller ratio of warriors to producers. These results are consistent with the historical evidence. Finally, we explore conditions under which the social contributions of the warrior elite could discourage a revolution.  相似文献

The use of intact cells in medical research offers a number of advantages over employing cell-free systems. In diagnostics, cells isolated from liquid biopsies can be directly used, speeding up the time of analysis and diminishing the risk of protein degradation by sample manipulation. In drug discovery, studies in live cells take into account aspects neglected in cell-free systems, such as uptake, metabolization, and subcellular concentration by compartmentalization of potential drug candidates. Therefore, probes for studies in cellulo are of paramount importance. Acid ceramidase (AC) is a lysosomal enzyme that hydrolyses ceramides into sphingoid bases and fatty acids. The essential role of this enzyme in the outburst and progress of several diseases, some of them still incurable, is well sustained. Despite the great clinical relevance of AC as a biomarker and therapeutic target, the specific monitoring of AC activity in live cells has remained elusive due to the concomitant existence of neutral and alkaline ceramidases. In this work, we report that 1-deoxydihydroceramides are exclusively hydrolysed by AC. Using N-octanoyl-18-azidodeoxysphinganine as a probe and a BODIPY-substituted bicyclononyne, we show the click-reliant predominant staining of lysosomes, with extra-lysosomal labeling also occurring in some cells. Importantly, using pharmacological and genetic tools together with high resolution mass spectrometry, we demonstrate that both lysosomal and extra-lysosomal staining are AC-dependent. These findings are translated into the specific flow cytometry monitoring of AC activity in intact cells, which fills an important gap in the field of diseases linked to altered AC activity.

Electrophilic trisubstituted ethylenes, phenoxy ring-substituted methyl 2-cyano-3-phenyl-2-propenoates, RPhCH=C(CN)CO2CH3, where R is 4-(4-BrC6H5O), 2-(4-ClC6H5O), 3-(4-ClC6H5O), 4-(3-ClC6H5O), 4-(4-ClC6H5O), 4-(4-FC6H5O), 2-(3-CH3OC6H5O), 2-(4-CH3OC6H5O), 3-(4-CH3OC6H5O), 4-(4-CH3OC6H5O), 3-(4-CH3C6H5O) were prepared and copolymerized with styrene. The monomers were synthesized by the piperidine catalyzed Knoevenagel condensation of phenoxy ring-substituted benzaldehydes and methyl cyanoacetate, and characterized by CHN analysis, IR, 1H and 13C-NMR. All the ethylenes were copolymerized with styrene (M1) in solution with radical initiation (ABCN) at 70°C. The compositions of the copolymers were calculated from nitrogen analysis and the structures were analyzed by IR, 1H and 13C-NMR. The order of relative reactivity (1/r1) for the monomers is 4-(4-CH3OC6H5O) (6.07) > 3-(4-ClC6H5O) (3.38) > 3-(4-CH3OC6H5O) (2.78) > 4-(3-ClC6H5O) (2.77) > 2-(4-ClC6H5O) (2.29) > 3-(4-CH3C6H5O) (1.98) > 4-(4-FC6H5O) (1.92) > 4-(4-ClC6H5O) (1.89) > 2-(3-CH3OC6H5O) (1.39) > 2-(4-CH3OC6H5O) (0.90) > 4-(4-BrC6H5O) (0.77). Relatively high Tg of the copolymers in comparison with that of polystyrene indicates a decrease in chain mobility of the copolymer due to the high dipolar character of the trisubstituted ethylene monomer unit. Decomposition of the copolymers in nitrogen occurred in two steps, first in the 200-500°C range with residue (2.5-8.0% wt), which then decomposed in the 500-800°C range.  相似文献

Abstract

In our studies of the stability constants of metal complexes, we have investigated a number of macrocyclic ligands with pendant donor groups. The ligands are characterized by the fact that they have nitrogen donors in the macrocyclic ring and oxygen or sulfur donors in the pendant arms. These ligands represent seven different macrocycles, and by varying the pendant donor groups, ten different ligands are indicated. The affinities of these ligands for fifteen metal ions will be described. The Fe(III) complex of triazanonane with o-hydroxypyridyl or o-hydroxybenzyl pendant donor groups are the most stable ferric complexes ever reported. The In(III) complex of triazacyclononane with pendant mercaptoethyl donor groups, is exceptionally stable. Also, the Ca(II) complex of DOTA probably has the highest stability of any calcium(II) complex. These, and other comparisons will be made on the basis of the thermodynamic stability constant data for the ligands described.  相似文献

The ground state configuration of the gas phase cationic dyes pinacyanol chloride and rhodamine B are optimized with HF/6–311 + G(2d,2p) method and basis set. B3PW91/6–311 + G(2df,2p) functional and basis set is used to calculate the Mulliken atom charge distribution, total molecular energy, the dipole moment, the vertical ionization potential, the adiabatic electron affinity and the lowest excited triplet state, the last three as an energy difference between separately calculated open shell and ground states. The triplet and extra electron states are optimized to find the relaxation energy. In the ground state optimization of both dyes the chloride anion migrates to a position near the center of the chromophore. For rhodamine B the benzoidal group turns perpendicular to the chromophore plane. For both dyes, the LUMO is mostly of π character associated with the aromatic part of the molecule containing the chromophore. The highest occupied MOs consist of three almost degenerate eigenvectors involving the chloride anion coordinated with σ electrons in the molecular framework. The fourth highest MO is of π character. For both molecules in the gas phase ionization process the chloride anion loses the significant fraction of electric charge. In electron capture, the excess charge goes mainly on the dye cation.  相似文献

An experimental investigation on the electrokinetic phenomenon known as primary electroviscous effect is described for suspensions of Pyrex glass, a highly charged and well known material. By means of an automatic method, the viscosity of the suspensions is measured for different volume fractions of solids, at various electrolyte concentrations and pH values. These measurements allow the estimation of the electroviscous coefficient, p. The electrophoretic mobility was determined for the same systems and zeta potential calculated from these experimental data in order to carry out the comparison between the measured values of p and different theoretical predictions. A qualitative agreement between theory and experiment was found in many cases, but the rigorous theories seem to systemmatically underestimate the experimental p values. The reasons for this disagreement are discussed in addition to the general trends of the electrokinetic behaviour of Pyrex glass.  相似文献
