Globular proteins are an important component of many food products. Heat-induced aggregation of globular proteins gives them new properties that can be useful in food products. In order to optimize functionality, the aggregation process needs to be controlled, which in turn requires good understanding of the mechanism. Heating aqueous solutions of globular proteins leads to the formation of aggregates with one of four distinctly different morphologies: spherical particles, flexible strands, semi-flexible fibrils, and fractal clusters. We review recent research in this area focusing on the parameters that control the morphology including the influence of hydrolysis. The aggregation mechanism and the effect of the morphology on the functionality will be addressed. A distinction is made between primary aggregation leading to roughly spherical particles or more or less flexible strands and secondary aggregation leading to fractal clusters, gels or precipitates. We will discuss how the formation of aggregates with different morphologies is related to the formation of either particulate or fine stranded gels.  相似文献

The modification of silica nanoparticles with hexafluoropropylene oxide (HFPO) oligomers has been investigated. HFPO oligomers with two different average degrees of polymerization (DPn = 8 and 15) were first prepared by anionic ring-opening polymerization, deactivated by methanol, and in some cases postfunctionalized by aminopropyl(tri)ethoxysilane or allylamine. The "grafting onto" reactions of these oligomers were then carried out either on bare silica (reaction between a silanol surface and ethoxy-silanized HFPO) or on silica functionalized by amino groups (in an amidation reaction with methyl ester-ended HFPO) or mercapto groups (via the radical addition of allyl-functionalized HFPO). Hybrid nanoparticles thus obtained were characterized by solid-state (29)Si NMR and FTIR spectroscopies as well as elemental and thermogravimetric analyses. The results assessed a significant yield of covalent grafting of HFPO oligomers when performing the hydrolysis-condensation of ethoxylated HFPO on the bare silica surface, compared to the other two methods that merely led to physically adsorbed HFPO chains. Chemically grafted nanohybrids showed a high thermal stability (up to 400 °C) as well as a very low surface tension (typically 5 mN/m) compared to physisorbed complexes.  相似文献

The development of personalized and non‐invasive cancer therapies based on new targets combined with nanodevices is a major challenge in nanomedicine. In this work, the over‐expression of a membrane lectin, the cation‐independent mannose 6‐phosphate receptor (M6PR), was specifically demonstrated in prostate cancer cell lines and tissues. To efficiently target this lectin a mannose‐6‐phosphate analogue was synthesized in six steps and grafted onto the surface of functionalized mesoporous silica nanoparticles (MSNs). These MSNs were used for in vitro and ex vivo photodynamic therapy to treat prostate cancer cell lines and primary cell cultures prepared from patient biopsies. The results demonstrated the efficiency of M6PR targeting for prostate cancer theranostic.  相似文献

A previously developed approach to the synthesis of the phomoidrides has been modified to incorporate all necessary carbon atoms prior to the key tandem carbonylation/Cope rearrangement reaction. This modification necessitated the synthesis of a challenging all-carbon quaternary stereocenter, which in turn rendered ineffective several reactions from the original synthesis. An oxidative radical cleavage of a spirocyclopropane ring system was developed that accomplishes the synthesis of the quaternary center, and a regioselective double hydroboration reaction was devised that provides an alternate approach to a key sequence of functional group interconversions, where the originally developed route was found to be ineffective.  相似文献

This paper reports the synthesis and testing of a molecularly imprinted polymer membrane for digoxin analysis. Digoxin-specific bulk polymer was obtained by the UV initiated co-polymerisation of methacrylic acid and ethylene glycol dimethacrylate in acetonitrile as porogen. After extracting the template analyte, the ground polymer particles were mixed with plasticizer polyvinyl chloride to form a MIP membrane. A reference polymer membrane was prepared from the same mixture of monomers but with no template. The resultant membrane morphologies were examined by scanning electron microscopy. The imprinted membrane was tested as the recognition element in a digoxin-sensitive fluorescence sensor; sensor response was measured using standard solutions of digoxin at concentrations of up to 4 × 10−3 mg L−1. The detection limit was 3.17 × 10−5 mg L−1. Within- and between-day relative standard deviations RSD (n = 5) were in the range 4.5-5.5% and 5.5-6.5% respectively for 0 and 1 × 10−3 mg L−1 digoxin concentrations. A selectivity study showed that compounds of similar structure to digoxin did not significantly interfere with detection for interferent concentrations at 10, 30 and 100 times higher than the digoxin concentration. This simply manufactured MIP membrane showed good recognition characteristics, a high affinity for digoxin, and provided satisfactory results in analyses of this analyte in human serum.  相似文献

Differential thermal analysis coupled to temperature-controlled diffraction have given evidence of a topological metastability phenomenon in an extended compositional range of the La2−xNdxMo2O9 solid solution. A metastable-stable phase diagram is proposed for this series of LAMOX-type fast oxide-ion conductors. In the Nd range 0<x?0.35, a freezing of the oxygen/vacancy disorder of the β-phase at ambient temperature can be achieved through a splat-quenching to water-ice mixture or/and shaping/sintering into pellet. In the intermediate 0.4?x?1.2 range, the amount of β-metastable phase grows upon substitution for powders. The negative impact of β-metastable to α phase transition on conductivity tends to disappear through the partial stabilization of the β phase by shaping/sintering.  相似文献

We present a new and advanced methodology, developed for surface functionalisation of gold and to study immobilisation of an immuno-specific system of proteins. A combination of electrochemical quartz crystal microbalance and Raman spectroscopy techniques allowed a complete understanding of the system starting from surface functionalisation and progressing to the functional structure analysis of immobilised proteins. A simple electrochemical procedure was formulated to prepare sulphonyl chloride terminated gold surfaces that form a strong sulphonamide bond with the receptor protein staphylococcal protein A (SpA). On the SpA grafted surfaces, the immobilisation of a human IgG and consecutive binding of an immuno-specific anti-human IgG was observed. The surface functional groups form a strong interaction with SpA without disturbing its functional properties. The native functional structure of SpA and also the IgGs was found to be retained in their immobilised state.  相似文献

The influence of the water content in the initial composition on the size of silica particles produced using the Stöber process is well known. We have shown that there are three morphological regimes defined by compositional boundaries. At low water levels (below stoichiometric ratio of water:tetraethoxysilane), very high surface area and aggregated structures are formed; at high water content (>40?wt%) similar structures are also seen. Between these two boundary conditions, discrete particles are formed whose size are dictated by the water content. Within the compositional regime that enables the classical Stöber silica, the structural evolution shows a more rapid attainment of final particle size than the rate of formation of silica supporting the monomer addition hypothesis. The clearer understanding of the role of the initial composition on the output of this synthesis method will be of considerable use for the establishment of reliable reproducible silica production for future industrial adoption.  相似文献

Neutral polymeric surfactants were synthesized by covalent attachment of hydrophobic groups (aromatic rings) onto a polysaccharide backbone (dextran). By changing the conditions of the modification reaction, the number of grafted hydrophobic groups per 100 glucopyranose units (substitution ratio) was varied between 7 and 22. In aqueous solution, these polymers behaved like classical associative polymers as demonstrated by viscometric measurements. The associative behavior was more pronounced when the substitution ratio increased. The surface-active properties of the modified dextrans were evidenced by surface tension (air/water) and interfacial tension (dodecane/water) measurements. In each case the surface or interfacial tension leveled down above a critical polymer concentration, which was attributed to the formation of a dense polymer layer at the liquid-air or liquid-liquid interface. Dodecane-in-water emulsions were prepared using the polymeric surfactants as stabilizers, with oil volume fractions ranging between 5 and 20%. The oil droplet size (measured by dynamic light scattering) was correlated to the amount of polymer in the aqueous phase and to the volume of emulsified oil. The thickness of the adsorbed polymer layer was estimated thanks to zeta potential measurements coupled with size measurements. This thickness increased with the amount of polymer available for adsorption at the interface. The dextran-based surfactants were also applied to emulsion polymerization of styrene and stable polystyrene particles were obtained with a permanent adsorbed dextran layer at their surface. The comparison with the use of an unmodified dextran indicated that the polymeric surfactants were densely packed at the surface of the particles. The colloidal stability of the suspensions of polystyrene particles as well as their protection against protein adsorption (bovine serum albumin, BSA, used as a test protein) were also examined.  相似文献
