Glycation (or non-enzymatic glycosylation) is a common non-enzymatic covalent modification of human proteins. Glucose, the highest concentrated monosaccharide in blood, can reversibly react with amino groups of proteins to form Schiff bases that can rearrange to form relatively stable Amadori products. These can be further oxidized to advanced glycation end products (AGEs). Here, we analyzed the glycation patterns of human serum albumin (HSA) in plasma samples obtained from five patients with type 2 diabetes mellitus. Therefore, glycated peptides from a tryptic digest of plasma were enriched with m-aminophenylboronic acid (mAPBA) affinity chromatography. The glycated peptides were then further separated in the second dimension by RP-HPLC coupled on-line to an electrospray ionization (ESI) tandem mass spectrometer (MS/MS). Altogether, 18 Amadori peptides, encompassing 40% of the HSA sequence, were identified. The majority of the peptides were detected and relatively quantified in all five samples with a high reproducibility among the replicas. Eleven Lys-residues were glycated at similar quantities in all samples, with glycation site Lys549 (KAm(Glc)QTALVELVK) being the most abundant. In conclusion, the established mAPBA/nanoRP-HPLC-ESI-MS/MS approach could reproducibly identify and quantify glycation sites in plasma samples, potentially useful in diagnosis and therapeutic control. 相似文献

Composite silica particles were synthesized by a two-step (acid-base) process in an aqueous solution with a mixture of organoalkoxysilane monomers. The two-step process separates the hydrolysis and condensation procedures to easily control condensation rate. In this study, the silane monomers used were phenyltrimethoxysilane (PTMS), vinyltrimethoxysilane (VTMS), methyltrimethoxysilane (MTMS), and tetraethyl-orthosilicate (TEOS). The physical properties of the resultant composite particles were investigated with the change in the molar ratio of monomers. Reaction of (triphenylmethyl)silanetriol (1) with cyclopentadienyltitanium trichloride (CpTiCl3) in the presence of triethylamine as a HCl scavenger gave both compounds of a partial open-cage type {[Ph3CSiO(OH)]3(Ph3CSiO3/2)(CpTiO3/2)4} (2) and cube type (Ph3CSiO3/2CpTiO3/2)4 (3). The 1:1 reaction of 1 and CpTiCl3 in toluene solvent at reflux temperature for 3 d afforded compounds 2 (22%) and 3 (36%). When 1 is reacted with a 1.5 fold excess of CpTiCl3 under the same conditions, compound 3 was obtained in high yield (81%) along with 2 in trace quantities. Compounds 2 and 3 were fully characterized by the analyses of 1H, 13C, 29Si NMR, IR, and FAB MS data. The solid-state structure of 3 was determined by a single crystal X-ray diffraction study. Compound 3 had shown to have catalytic activity for the oxidation of alkenes such as 1-octene, cyclooctene, and norbornene with t-butyl hydrogen peroxide. Circulating tumor cells (CTCs) have gained increasing attention as physicians and scientists learn more about the role these extraordinarily rare cells play in metastatic cancer. In developing CTC technology, the critical criteria are high recovery rates and high purity. Current isolation methods suffer from an inherent trade-off between these two goals. Moreover, ensuring minimal cell stress and robust reproducibility is also important for the clinical application of CTCs. In this paper, we introduce a novel CTC isolation technology using selective size amplification (SSA) for target cells and a multi-obstacle architecture (MOA) filter to overcome this trade-off, improving both recovery rate and purity. We also demonstrate SSA-MOA's advantages in minimizing cell deformation during filter transit, resulting in more stable and robust CTC isolation. In this technique, polymer microbeads conjugated with anti-epithelial cell adhesion molecules (anti-EpCAM) were used to selectively size-amplify MCF-7 breast cancer cells, definitively differentiating from the white blood cells (WBCs) by avoiding the size overlap that compromises other size selection methods. 3 μm was determined to be the optimal microbead diameter, not only for size discrimination but also in maximizing CTC surface coverage. A multi-obstacle architecture filter was fabricated using silicon-on-glass (SOG) technology-a first such application of this fabrication technique-to create a precise microfilter structure with a high aspect ratio. The filter was designed to minimize cell deformation as simulation results predicted that cells captured via this MOA filter would experience 22% less moving force than with a single-obstacle architecture. This was verified by experiments, as we observed reliable cell capture and reduced cell deformation, with a 92% average recovery rate and 351 peripheral blood leukocytes (PBL) per millilitre (average). We expect the SSA-MOA platform to optimize CTC recovery rates, purity, and stability, increasing the sensitivity and reliability of such tests, thereby potentially expanding the utilization of CTC technologies in the clinic. 相似文献

The adsorption and thermal stability of 2-octylthieno[3,4-b]thiophene (OTTP) on the Au(111) surfaces have been studied using scanning tunneling microscopy (STM), temperature programmed desorption (TPD), and X-ray photoelectron spectroscopy (XPS). UHV-STM studies revealed that the vapor-deposited OTTP on Au(111) generated disordered adlayers with monolayer thickness even at saturation coverage. XPS and TPD studies indicated that OTTP molecules on Au(111) are stable up to 450K and further heating of the sample resulted in thermal decomposition to produce H(2) and H(2)S via C-S bond scission in the thieno-thiophene rings. Dehydrogenation continues to occur above 600K and the molecules were ultimately transformed to carbon clusters at 900K. Highly resolved air-STM images showed that OTTP adlayers on Au(111) prepared from solution are composed of a well-ordered and low-coverage phase where the molecules lie flat on the surface, which can be assigned as a (9×2√33)R5° structure. Finally, based on analysis of STM, TPD, and XPS results, we propose a thermal decomposition mechanism of OTTP on Au(111) as a function of annealing temperature. 相似文献

The analysis of mineral contents in space foods is needed to obtain an information on a comprehensive elemental composition as well as the investigation on the effects of human nutrition and health based on the dietary intake of mineral elements. Recently, six items of new Korean space foods (KSFs) such as kimchi, bibimbap, bulgogi, a ramen, a mulberry beverage and a fruit punch which was developed by the KAERI, and the contents of more than 15 elements in the samples were examined by using instrumental neutron activation analysis (INAA). Five biological certified reference materials, NIST SRM were used for analytical quality control. Chlorination reaction behavior of Zircaloy-4 (Zry-4) cladding hulls was demonstrated by using a quartz reactor system. By reacting at 380 °C for 3 h, mass of the Zry-4 hulls decreased by 65.8 wt% with Cl2 utilization of 87.1 mol%. Composition of collected product was analyzed and it was revealed that concentration of Zr was higher than 99.97 wt%. The purity of Zr in the experimental result was higher than expectation when considering Sn (1.31 wt%) and Fe (0.25 wt%) contents which can produce gaseous SnCl4 and FeCl3 at the experimental condition. Theoretical calculations were performed to clarify the high purity of Zr by using the HSC code. The simulation results revealed that formation of ZrCl4 is more preferred than SnCl4, FeCl3, and CrCl3. The preference of chloride formation was confirmed by the theoretical calculation, and it was suggested that the major constituents of Zry-4 might react with Cl2 to produce chlorides in an order of ZrCl4 > CrCl3 > SnCl4 > FeCl3. It was also suggested that continuous removal of ZrCl4 and sufficient supply of Zr source during the chlorination reaction might have contributed to the high purity of Zr. 相似文献

Present article deals with functionality of silver nanostructured surfaces prepared by potentiostatic electrochemical deposition on the paraffin impregnated graphite electrode as template‐free substrates. The effect of the electrodeposition conditions on two silver surface functions: analytical signal enhancement in Surface‐enhanced Raman spectroscopy and pre‐ionization function, applied in secondary ion mass spectrometry (SIMS) is reported. Functional silver nanostructured substrate was prepared at a potential ?850 mV with a deposition duration of 20 min. Analytical signal enhancement factors of 3.2 ×105 for Raman peak at 649 cm?1, 3.0×105 for peak at 810 cm?1 and 2.7×105 for peak at 1539 cm?1 were determined for Rhodamine 6G at deposited surface. Slight pre‐ionization effect has been observed in SIMS, and 1.2×105 fold signal enhancement was established for fragment of Rhodamine 6G with m/z 429 (M‐CH3‐Cl).
