The cluster compounds [Pt3(μ-CO)3(PR3)3] act as Lewis bases towards the metal halides of Group XI, MX, Group XII, MX2, and Group XIII, MX3, to form cluster compounds of the composition [{MXn}{Pt3(μ-CO)3(PR3)3}]. The X-ray crystal structure, NMR and IR data are given for the compound [{ZnI}{Pt3(μ-CO)3(PPh(i-Pr)2)3}].  相似文献

The photolability of the antitumor antibiotic hedamycin ( 1 ) was investigated by irradiation in different solvents in the presence or in the absence of oxygen. The products formed were separated chromatographically and their structures determined by NMR spectroscopy. Photolysis of 1 in the presence of oxygen gave only one isolable product, photohedamycin A ( 3 ), where ring E of hedamycin had been transformed into an enol ether. The reaction in the absence of oxygen yielded the photohedamycins B, C, and D ( 5, 6 , and 7 , respectively). In these compounds, one of the epoxides of hedamycin had been opened reductively, and in photohedamycin D ( 7 ) the substituent at C(8) - originally ring E of hedamycin - was now acyclic. In addition to these compounds, the photolyses yielded a large number of unstable minor products, which could not be isolated.  相似文献

The oligosaccharide antibiotic avilamycin A is composed of a polyketide-derived dichloroisoeverninic acid moiety attached to a heptasaccharide chain consisting of six hexoses and one unusual pentose moiety. We describe the generation of mutant strains of the avilamycin producer defective in different sugar biosynthetic genes. Inactivation of two genes (aviD and aviE2) resulted in the breakdown of the avilamycin biosynthesis. In contrast, avilamycin production was not influenced in an aviP mutant. Inactivation of aviGT4 resulted in a mutant that accumulated a novel avilamycin derivative lacking the terminal eurekanate residue. Finally, AviE2 was expressed in Escherichia coli and the gene product was characterized biochemically. AviE2 was shown to convert UDP-D-glucuronic acid to UDP-D-xylose, indicating that the pentose residue of avilamycin A is derived from D-glucose and not from D-ribose. Here we report a UDP-D-glucuronic acid decarboxylase in actinomycetes.  相似文献

Two different extraction procedures to determine alachlor (2-chloro-2,6-diethyl-N-methoxymethyl-acetanilide) and its metabolites 2,6-diethylaniline (DEA) and 2-chloro-2,6-diethylacetanilide (CDEA) at very low concentration levels in groundwater are compared. Both methods were performed during a field leaching study and laboratory soil column experiments. Solid phase extraction (SPE) is characterized by the enrichment of 11 water samples at 1 g RP-C18 material, while micro liquid/liquid extraction (MLLE) means that 400 ml water samples saturated with sodium chloride were extracted once with 500 l of toluene. Both methods differ in their concentration factors, i.e. 5000 for SPE and 800 for MLLE. The determination limits for alachlor determined as the lowest spiked concentration analyzed are 10 ng/l with SPE and 25 ng/l with MLLE. Even though the determination limits of the two methods are similar, the volatile metabolite 2,6-diethylaniline was not sufficiently recovered by SPE. Furthermore, SPE is more expensive and time consuming than MLLE. The latter is simple in use, rapidly performed and needs no evaporation step and clean-up before GC analysis. This is of special advantage for the analysis of 2,6-diethylaniline. The determination limits of the metabolites CDEA and DEA analyzed by MLLE are 25 ng/l. The extracts from both methods were directly analyzed by gas chromatography using electron capture and nitrogen-phosphorus detection.  相似文献

Tetrakis(trifluoromethyl) cyclotetraarsane (F3CAs)4 ( 2 ) was used to repeat the UV initiated [4+2]‐cycloaddition reaction of the diarsene F3CAs=AsCF3 ( 1 ) with cyclohexa‐1,3‐diene (CHD) and to isolate single crystals of the cycloadduct 4 for a X‐ray diffraction analysis. 4 crystallizes in the space group and contains the diarsene group in its E‐configuration. 2 was also applied for [2+2]‐cycloaddition reactions of 1 with tBuC≡P and MeC≡CNiPr2, but in contrast to positive results with (F3CP)4 the products were too labile for isolation. However, 2 was successfully used at room temperature as precursor for coordinating 1 as π‐donor ligand to the Pd(PPh3)2 complex fragment yielding η2‐bis(trifluoromethyl)diarsene‐bis(triphenylphosphane)‐palladium(0) 5 , which was characterized by X‐ray diffraction of single crystals and by spectroscopic investigations (NMR, IR, MS). Attempts to prove the existence of the diarsene 1 , generated by different methods, by spectroscopic studies very probably failed due to its extreme reactivity, not allowing the necessary concentrations for detection. Quantum chemical calculations of the stability of 1 with respect to dimerization, the stability of the [2+2]‐cycloadduct with 1‐di(isopropyl)aminopropyne and the energy difference between 4 and the 2,3‐dimethyl‐1,3‐butadiene cycloadduct of 1 were performed to understand the considerable differences between 1 and the related diphosphene F3CP=PCF3.  相似文献

For the first time several T(h)-symmetrical hexakisadducts of C(60) bearing up to six electro- and photoactive o-phenylene diamine or 9,10-dialkoxyanthracene moieties were synthesized and subjected to photoinduced electron/energy-transfer studies. Both donors form a densely packed pi-donor shell surrounding the fullerene core. In these novel core-shell ensembles (7 and 19), either an efficient energy transfer from the dialkoxyanthracene periphery, or an electron transfer from the o-phenylene diamine periphery transduces the flow of excited-state energy or electrons, respectively, to the fullerene moiety, which resides in the central core. Due to the relatively high reduction potential of the fullerene core, which is anodically shifted by approximately equal to 0.7 V, compared with that of pristine C(60), the outcome of these intramolecular reactions depends mainly on the donor ability of the peripheral system. Interestingly, the charge-separated state in the o-phenylene diamine heptad (7; tau=2380 ns in benzonitrile) is stabilized by a factor of 20 relative to the corresponding o-phenylene diamine dyad (6; tau=120 ns in benzonitrile), an effect that points unequivocally to the optimized storage of charges in this highly functionalized fullerene ensemble.  相似文献

Two macrocyclic aminoglycosides were prepared from a 1,4-butanediol linked 2-deoxy-L-rhamnal which was O-allylated at the 4- and 4'-positions via the precursor allyl 3,4-di-O-acetyl-2,6-dideoxy-alpha-L-arabino-hexoside employing olefin metathesis and ring closing metathesis in a sequential manner. The macrocycles were 15N-labelled at all four amino groups in order to study interactions with regulatory RNA structures in solution by NMR spectroscopy. A key step for the introduction of the 15N-label was a reductive amination step using commercially available 15NH4OAc. The reductive amination proceeds with excellent stereocontrol. As a by-product the unusual acyclic amino nitrile was isolated which originated from intramolecular imine formation followed by cyanide addition to the intermediate C=N double bond.  相似文献

The measurement of intracellular calcium fluxes in real time is widely applied within the pharmaceutical industry to measure the activation of G-protein coupled receptors (GPCRhyp;s), either for pharmacological characterisation or to screen for new surrogate ligands. Initially restricted to G(q) coupled GPCRs, the introduction of promiscuous and chimeric G-proteins has further widened the application of these assays. The development of new calcium sensitive dyes and assays has provided sensitive, homogeneous assays which can be readily applied to high throughput screening (HTS). In this paper we describe the full automation of this assay type using a fluorometric imaging plate reader (FLIPR ) integrated into a Beckman/Sagian system to establish a simple robotic system that is well suited for the current medium throughput screening in this area of lead discovery. Using a recently completed HTS we discuss important determinants for FLIPR based screening, highlight some limitations of the current approach, and look at the requirements for future automated systems capable of keeping up with expanding compound files.  相似文献

Hybrid processes for enantioseparations have a considerable potential for reducing investment and operational costs. An example is the combination of simulated moving bed (SMB) chromatography and selective crystallisation. However, the design of integrated processes is a difficult task. A shortcut method is presented that can serve as a tool for design and estimation of the potential of such processes. The approach requires only limited experimental data and thus allows for systematic parameter studies. The method is based on the determination of the purity-performance characteristic of the SMB process and rigorous application of mass balances. The use of relative mass fluxes allows derivation of simple algebraic expressions for essential process parameters. The significant potential of combining SMB and crystallisation is demonstrated for the example of the separation of mandelic acid enantiomers.  相似文献
