In this paper, the relationship between the reversible redox properties and the second-order nonlinear optical (NLO) responses for the title series of complexes has been systematically investigated by using the time-dependent density functional theory (TDDFT) method combined with the sum-over-states (SOS) formalism. The results reveal that the successive reduction processes of five PW11ReN redox states should be PW11ReVII (1) --> PW11ReVI (2) --> PW11ReV (3) --> PW11ReV1e ( 4) --> PW 11ReV2e (5). Furthermore, their electrochemical properties have been reproduced successfully. It is noteworthy that the second-order NLO behaviors can be switched by reversible redox for the present studied complexes. Full oxidation constitutes a convenient way to switch off the second-order polarizability (system 1). The incorporation of extra electrons causes significant enhancement in the second-order NLO activity, especially for the third reduced state (system 4), whose static second-order polarizability (betavec) is about 144 times larger than that of fully oxidized 1. The characteristic of the charge-transfer transition corresponding to the dominant contributions to the betavec values indicates that metal-centered redox processes influence the intramolecular donor or acceptor character. Therefore, these kinds of complexes with the facile and reversible redox states could become excellent switchable NLO materials.  相似文献

Salvianolic acid A (SAA), a major effective constituent of Salvia miltiorrhizas, is widely used in traditional Chinese medicine. A sensitive rapid analytical method was established and validated for SAA in rat plasma, which was further applied to assess the pharmacokinetics of SAA in rats receiving a single oral dose of SAA. The method used liquid chromatography tandem mass spectrometry in multiple reaction monitoring mode with chloramphenicol as the internal standard. A simple liquid-liquid extraction based on ethyl acetate was employed. The combination of a simple sample cleanup and short chromatographic run time (3 min) increased the throughput of the method substantially. The method was validated over the range 1.4-1000 ng/mL with a correlation coefficient >0.99. The lower limit of quantification was 1.4 ng/mL for SAA in plasma. Intra- and inter-day accuracies for SAA were 95-113 and 98-107%, and the inter-day precision was less than 12%. This method is more sensitive and faster than previous methods. After a single oral dose of 100 mg/kg of SAA, the mean peak plasma concentration (Cmax) of SAA was 318 ng/mL at 0.5 h, the area under the plasma concentration-time curve (AUC0-12 h) was 698 +/- 129 ng.h/mL, and the elimination half-life (T1/2) was 3.29 h.  相似文献

In this study, paclitaxel loaded poly( L-lactic acid) (PTX-PLLA) microparticles were prepared using solution enhanced dispersion by supercritical CO2(SEDS) technique. This supercritical antisolvent technique offers the advantage of negligible organic solvent residua in the drug loaded microparticles. Scanning electron microscopy (SEM) showed that microparticles exhibited rather spherical shape and small particle size with narrow particle size distribution. X-ray diffraction (XRD) and differential scanning calorimeter (DSC) indicated that PTX was amorphously dispersed in the PLLA matrix. The drug loading and encapsulation efficiency of PTX-PLLA microparticles were 14.33% and 62.68%, respectively. In vitro cytotoxicity evaluation of PTX-PLLA microparticles against nonsmall-cell lung cancer A549 and ovarian cancer SKOV3 cell lines indicated that PTX-PLLA had superior antiproliferation activity against the A549 and SKOV3 cell lines, compared with free PTX formulations. The cellular internalization of fluorescent microparticles was evidenced by fluorescence microscope and further confirmed by transmission electron microscopy (TEM). This was attributed to the efficient intracellular accumulation of PTX via cell phagocytosis and sustained release of PTX from PLLA matrix. The anticancer activity of PTX-PLLA was associated with PTX-induced cell apoptosis such as nuclear aberrations, condensation of chromatin and swelling damage in mitochondria. The cell apoptosis index detected by flow cytometry was higher in PTX-PLLA group than in free PTX. The PTX-PLLA formulation, which was obtained through micronization of PTX and encapsulation of micronized PTX into PLLA simultaneously in the SEDS process, significantly potentiated the anticancer activity of PTX.  相似文献

Electropolymerization, morphology characterization, and ion transport of poly(3,4-ethylenedioxythiophene) (PEDOT) films doped with different counterions (chloride, ferrocyanide (FCN), and poly(p-styrenesulfonate) (PSS-)) on a platinum electrode were investigated using scanning electrochemical microscopy (SECM) during both potential step (chronocoulometric) and cyclic voltammetric scans. An ultramicroelectrode (UME) tip was positioned close to the surface of a PEDOT-modified substrate electrode, and the responses of both electrodes to a substrate potential step or linear sweep were monitored simultaneously. Chloride or ferrocyanide (FCN) ejection during PEDOT reduction was shown to be a function of the reduction potential. The nature of the cation in the bulk solution was not found to be important in the kinetics of ion transport in PEDOT+/FCN- films. Direct evidence for the incorporation of cations of Ru(NH3)6(3+/2+) in a PEDOT film during its reduction was also obtained by SECM measurements. The adsorption of Ru(NH3)6(3+) in fully oxidized PEDOT+/PSS- films was observed and attributed to ion exchange between the Na+ co-ion of PSS- and Ru(NH3)6(3+) in the bulk solution.  相似文献

The viscoelasticity of aqueous micellar solutions of two oxyethylene/oxybutylene block copolymers (E(92)B(18) and B(20)E(510)) has been investigated using a torsional resonator operated at 26 kHz. For both systems considered, values of the dynamic viscosity (eta'(infinity)) point to partial draining of the micellar corona induced by the high-frequency oscillatory field. At low effective volume fractions, values of the elastic modulus (G'(infinity)) indicate that the repulsive interactions between micelles can be modeled by a power law function u(r) proportional to 1/r(nu) with exponents close to 13 and 6 for copolymers E(92)B(18) and B(20)E(510) respectively. At a critical copolymer concentration (c*) plots of log(G'(infinity)) against log(c) deviate from the straight lines established at low concentrations, implying that the systems undergo ergodic/nonergodic transitions.  相似文献

We report a method of coupled CE-LIF detection with flow cytometry for high-throughput determination and quantitation of fluorophores in single intact K562/S (KS) cells. The membrane properties of KS cell including fluophore transport rate and apparent permeability coefficient were further quantitatively characterized. The method has advantages for accurate quantitation and unique capacity of high-throughput analysis. The strategy will be useful for the quantitation of fluorophores in the intact cells, such as measurement of multidrug resistance, quantitation of specific protein expression, and quantitative characterization of protein and enzyme functions.  相似文献

High throughput screening (HTS) campaigns, where laboratory automation is used to expose biological targets to large numbers of materials from corporate compound collections, have become commonplace within the lead generation phase of pharmaceutical discovery. Advances in genomics and related fields have afforded a wealth of targets such that screening facilities at larger organizations routinely execute over 100 hit-finding campaigns per year. Often, 10(5) or 10(6) molecules will be tested within a campaign/cycle to locate a large number of actives requiring follow-up investigation. Due to resource constraints at every organization, traditional chemistry methods for validating hits and developing structure activity relationships (SAR) become untenable when challenged with hundreds of hits in multiple chemical families per target. To compound the issue, comparison and prioritization of hits versus multiple screens, or physical chemical property criteria, is made more complex by the informatics issues associated with handling large data sets. This article describes a collaborative research project designed to simultaneously leverage the medicinal chemistry and drug development expertise of the Novartis Institutes for Biomedical Research Inc. (NIBRI) and ArQule Inc.'s high throughput library design, synthesis and purification capabilities. The work processes developed by the team to efficiently design, prepare, purify, assess and prioritize multiple chemical classes that were identified during high throughput screening, cheminformatics and molecular modeling activities will be detailed.  相似文献

Improved binary PSO for feature selection using gene expression data   总被引:2,自引:0,他引:2  
Gene expression profiles, which represent the state of a cell at a molecular level, have great potential as a medical diagnosis tool. Compared to the number of genes involved, available training data sets generally have a fairly small sample size in cancer type classification. These training data limitations constitute a challenge to certain classification methodologies. A reliable selection method for genes relevant for sample classification is needed in order to speed up the processing rate, decrease the predictive error rate, and to avoid incomprehensibility due to the large number of genes investigated. Improved binary particle swarm optimization (IBPSO) is used in this study to implement feature selection, and the K-nearest neighbor (K-NN) method serves as an evaluator of the IBPSO for gene expression data classification problems. Experimental results show that this method effectively simplifies feature selection and reduces the total number of features needed. The classification accuracy obtained by the proposed method has the highest classification accuracy in nine of the 11 gene expression data test problems, and is comparative to the classification accuracy of the two other test problems, as compared to the best results previously published.  相似文献

The development of a new method for stereoselective glycosylation with glycosyl trichloroacetimidate donors employing cationic palladium(II), Pd(CH(3)CN)(4)(BF(4))(2), is described. This process employs Pd(CH(3)CN)(4)(BF(4))(2) as an efficient activator, providing access to a variety of disaccharides and glycopeptides. This reaction is highly stereoselective and proceeds under mild conditions with low catalyst loading. Interestingly, this palladium catalysis directs beta-glucosylations in the absence of classical neighboring group participation.  相似文献
