Using experiments involving cyclic voltammetry and stationary potential measurements, it was shown that siloxane polymers with covalently attached ferrocene and 1,1'-dimethylferrocene relays efficiently mediate electron transfer from reduced glycolate oxidase to a conventional carbon-paste electrode. Sensors containing these polymeric relay systems and glycolate oxidase respond rapidly to low (< 0.1 mM) glycolate concentrations, with steady-state current responses achieved in less than 1 min. The dependence of the sensor response on the nature of the siloxane polymer backbone, the type of polymer-bound redox mediator used and the amount of redox mediator present is discussed. From these considerations, sensors have been designed which can operate efficiently at low applied potential and can avoid decreased current response due to dissolved oxygen.  相似文献

Enzymatic degradation of p-chlorophenol was carried out in a two-phase flow in a microchannel (100 microm width, 25 microm depth) fabricated on a glass plate (70 mm x 38 mm). This is the first report on the enzymatic reaction in a two-phase flow on a microfluidic device. The surface of the microchannel was partially modified with octadecylsilane groups to be hydrophobic, thus allowing clear phase separation at the end-junction of the microchannel. The enzyme (laccase), which is surface active, was solubilized in a succinic aqueous buffer and the substrate (p-chlorophenol) was in isooctane. The degradation of p-chlorophenol occurred mainly at the aqueous-organic interface in the microchannel. We investigated the effects of flow velocity and microchannel shape on the enzymatic degradation of p-chlorophenol. Assuming that diffusion of the substrate (p-chlorophenol) is the rate-limiting step in the enzymatic degradation of p-chlorophenol in the microchannel, we proposed a simple theoretical model for the degradation in the microchannel. The calculated degradation values agreed well with the experimental data.  相似文献

Polymerization of methyl acrylate (MA) with benzyl N, N-diethyldithiocarbamate (BDC) was carried out in bulk or in benzene under UV irradiation. The polymerization proceeded to give a high yield of polymer, however it was found that BDC did not function as an iniferter in the MA polymerization. Bimolecular termination leading to deactivation of the iniferter site was observed during the polymerization in preference to chain transfer and primary radical termination which reproduce the iniferter site. The deactivation was successfully prevented by combining BDC and tetraethylthiuram disulfide (TD) as a two-component iniferter. This two-component iniferter system was also applied to synthesis of a star polymer with 1,2,4,5-tetrakis (N,N-diethyldithocarbamylmethyl) benzene (DDC) as a tetrafunctional photoiniferter. © 1994 John Wiley & Sons, Inc.  相似文献

Reaction of [Cp*IrCl2]2 (1) with dpmp in the presence of KPF6 afforded a binuclear complex [Cp*IrCl(dpmp-P1,P2;P3)IrCl2Cp*](PF6) (2) (dpmp =(Ph2PCH2)2PPh). The mononuclear complex [Cp*IrCl(dpmp-P1,P2)](PF6) (4) was generated by the reaction of [Cp*IrCl2(BDMPP)](BDMPP =PPh[2,6-(MeO)2C6H3]2) with dpmp in the presence of KPF6. These mono- and binuclear complexes have four-membered ring structures with a terminal and a central P atom of the dpmp ligand coordinated to an iridium atom as a bidentate ligand. Since there are two chiral centers at the Ir atom and a central P2 atom, there are two diastereomers that were characterized by spectrometry. Complexes anti-4 and syn-4 reacted with [Cp*RhCl2]2 or [(C6Me6)RuCl2]2, giving the corresponding mixed-metal complexes, anti- and syn- [Cp*IrCl(dppm-P1,P2;P3)MCl2L](PF6) (6: M = Rh, L = Cp*; 7: M = Ru, L = C6Me6). Treatment with AuCl(SC4H8) gave tetranuclear complexes, anti- and syn-8 [[Cp*IrCl(dppm-P1,P2;P3)AuCl]2](PF6)2 bearing an Au-Au bond. Reaction of anti- with PtCl2(cod) generated the trinuclear complex anti-9, anti-[[Cp*IrCl(dppm-P1,P2;P3)]2PtCl2](PF6)2. These reactions proceeded stereospecifically. The P,O-chelated complex syn-[Cp*IrCl(BDMPP-P,O)] (syn-10)(BDMPP-P,O = PPh[2,6-(MeO)2C6H3][2-O-6-(MeO)C6H3]2) reacted with dpmp in the presence of KPF6, generating the corresponding anti-complex as a main product as well as a small amount of syn-complex, [Cp*Ir(BDMPP-P,O)(dppm-P1)](PF6) (11). The reaction proceeded preferentially with inversion. The reaction processes were investigated by PM3 calculation. anti- was treated with MCl2(cod), giving anti-[Cp*Ir(BDMPP-P,O)(dppm-P1;P2,P3)MCl2](PF6)(14: M = Pt; 15: M = Pd), in which the MCl2 moiety coordinated to the two free P atoms of anti-11. The X-ray analyses of syn-2, anti-2, anti-4, anti-8 and anti-11 were performed.  相似文献

In the presence of 2.5 mol % of [Pd(2)(dba)(3)] (dba=dibenzylideneacetone) and 5 mol % of PPh(3), nearly equimolar amounts of dimethyl nona-2,7-diyne-1,9-dioate derivatives (diyne diesters) and dialkyl acetylenedicarboxylates were allowed to react in toluene at 110 degrees C to afford [2+2+2] cycloadducts in moderate-to-good yields. Similarly, dimethyl trideca-2,7,12-triyne-1,13-dioate derivatives (triyne diesters) were catalytically transformed into phthalic acid ester analogues in excellent yields. To gain insight into the mechanism of these intramolecular alkyne cyclotrimerizations, stoichiometric reactions of [Pd(2)(dba)(3)] with a diyne diester and a triyne diester bearing ether tethers were conducted in acetone at room temperature to furnish an oligomeric bicyclopalladacyclopentadiene and a Pd(0) triyne complex, respectively. The structures of these novel complexes were unequivocally determined by Xray structure analysis. The isolated triyne complex was heated at 50 degrees C or treated with PPh(3) in acetone at room temperature to afford the arene product. We developed a metabolically engineered Saccharomyces cerevisiae, which produces optically pure l-lactic acid efficiently using cane juice-based medium. In this recombinant, the coding region of pyruvate decarboxylase (PDC)1 was completely deleted, and six copies of the bovine l-lactate dehydrogenase (l-LDH) genes were introduced on the genome under the control of the PDC1 promoter. To confirm optically pure lactate production in lowcost medium, cane juice-based medium was used in fermentation with neutralizing conditions. l-lactate production reached 122 g/L, with 61% of sugar being transformed into l-lactate finally. The optical purity of this l-lactate, that affects the physical characteristics of poly-l-lactic acid, was extremely high, 99.9% or over. These two authors contributed equally to this work.  相似文献

Reactions of Fe[N(SiMe(3))(2)](2) with 1 and 2 equiv of Ph(3)SiSH in hexane afforded dinuclear silanethiolato complexes, [Fe(N(SiMe(3))(2))(mu-SSiPh(3))](2) (1) and [Fe(SSiPh(3))(mu-SSiPh(3))](2) (2), respectively. Various Lewis bases were readily added to 2, generating mononuclear adducts, Fe(SSiPh(3))(2)(L)(2) [L = CH(3)CN (3a), 4-(t)BuC(5)H(4)N (3b), PEt(3) (3c), (LL) = tmeda (3d)]. From the analogous reactions of M[N(SiMe(3))(2)](2) (M = Mn, Co) and [Ni(NPh(2))(2)](2) with Ph(3)SiSH in the presence of TMEDA, the corresponding silanethiolato complexes, M(SSiPh(3))(2)(tmeda) [M = Mn (4), Co (5), Ni (6)], were isolated. Treatment of 3a with (PPh(4))(2)[MoS(4)] or (NEt(4))(2)[FeCl(4)] resulted in formation of a linear trinuclear Fe-Mo-Fe cluster (PPh(4))(2)[MoS(4)(Fe(SSiPh(3))(2))(2)] (7) or a dinuclear complex (NEt(4))(2)[Fe(2)(SSiPh(3))(2)Cl(4)] (8). On the other hand, the reaction of 3a with [Cu(CH(3)CN)(4)](PF(6)) gave a cyclic tetranuclear copper cluster Cu(4)(SSiPh(3))(4) (9), where silanethiolato ligands were transferred from iron to copper. Silicon-sulfur bond cleavage was found to occur when the cobalt complex 5 was treated with (NBu(4))F in THF, and a cobalt-sulfido cluster Co(6)(mu(3)-S)(8)(PPh(3))(6) (10) was isolated upon addition of PPh(3) to the reaction system. The silanethiolato complexes reported here are expected to serve as convenient precursors for sulfido cluster synthesis.  相似文献

Amphotericin B-sterol conjugates were synthesized and examined for their membrane permeabilizing activity. Ergosterol and cholesterol, each connected with amphotericin B via an ethylenecarbamate or hexamethylenecarbamate linker, were examined by K(+) flux assays using liposomes and by single-channel recording across phospholipid membrane. Among four conjugates tested, AmB-ergosterol bearing an ethylenecarbamate linker exhibited the most powerful activity, which substantially exceeded that of the cholesterol homolog. Single-channel recording clearly exhibited that the ergosterol conjugate elicited channel current with the conductance of 28 pS, which was comparable with those by AmB, and revealed a higher channel open probability than the cholesterol conjugate. These results imply that direct interaction between amphotericin B and ergosterol is reproduced by their conjugate, which may serve as a model compound for understanding the drug's selective toxicity.  相似文献

Treatment of 1-silylcyclopropylmethanols with TsOH in methanol gives different homoallyl ethers depending upon the configuration of substituents on cyclopropane ring and the kinds of substituents on carbinyl carbon. Especially, the reaction of cyclopropylmethanols having no substituents on the same side with silyl group on cyclopropane ring proceeds to give the corresponding E-homoallyl ethers with high stereoselectivity. The enantioselective reaction of alpha-seleno carbanions derived from bis(phenylseleno)acetal and bis(2-pyridylseleno)acetal in the presence of bis(oxazoline)s with various electrophiles gave products with high enantioselectivity. The enantioselective reaction of alpha-lithio benzyl 2-pyridyl selenide gave the products with stereochemistry reverse to that obtained in the reaction of alpha-lithio benzyl phenyl selenide. Mechanistic investigation suggests the enantiodetermination of these reactions at -78 degrees C depends on dynamic thermodynamic resolution. The enantioselective reaction was applied to the preparation of enantioenriched olefins and epoxide.  相似文献
