The pH-static enzyme sensor : An ISFET-based enzyme sensor,insensitive to the buffer capacity of the sample

An ISFET-based urea sensor is combined with a noble-metal electrode which provides continuous coulometric titration of the products of the enzymatic reaction. The sensor thus becomes independent of the buffer capacity of the sample; and because the enzyme is operating at a constant pH, the linear response range is expanded.     

The kalimantacin/batumin biosynthesis operon encodes a self-resistance isoform of the FabI bacterial target

BatG is a trans-2-enoyl-ACP reductase, encoded in?the kalimantacin/batumin (kal/bat) biosynthesis operon. It is not essential for the production of the kal/bat secondary metabolite. Instead, BatG is an isoform of FabI, conferring full resistance to target bacteria. It also complements FabI in its role in fatty acid biosynthesis. The identification of FabI as the antibacterial target is important to assess clinical potential of the kalimantacin/batumin antibiotics against Staphylococcus aureus.     

Clean-up of some organochlorine and pyrethroid insecticides by automated solid-phase extraction cartridges coupled to capillary GC-ECD

The ASPEC (Automatic Sample Preparation with Extraction Columns) system has been coupled on-line to capillary GC-ECD by means of a loop-type interface equipped with a solvent vapour exit. Both ASPEC and GC conditions have been optimized leading to an effective clean-up of the extracts analyzed. By means of solid-phase extraction cartridges filled with silica, it has been possible to analyze concentrated surface water extracts for a group of 18 electron-capturing compounds present in the water at ppt levels. ASPEC-GC has also been applied to the determination of synthetic pyrethroids at ppt levels in surface water. The complete analytical procedure is greatly facilitated by automation and miniaturization. Miniaturization results in a considerable decrease in the sample volume required. The potential of the method for the analysis of other pesticides is estimated.     

Isoguanine and 5‐Methyl‐Isocytosine Bases,In Vitro and In Vivo

The synthesis, base‐pairing properties and in vitro and in vivo characteristics of 5‐methyl‐isocytosine (isoC^Me) and isoguanine (isoG) nucleosides, incorporated in an HNA(h) (hexitol nucleic acid)–DNA(d) mosaic backbone, are described. The required h‐isoG phosphoramidite was prepared by a selective deamination as a key step. As demonstrated by T_m measurements the hexitol sugar showed slightly better mismatch discrimination against dT. The d‐isoG base mispairing follows the order T>G>C while the h‐isoG base mispairing follows the order G>C>T. The h‐ and d‐isoC^Me bases mainly mispair with G. Enzymatic incorporation experiments show that the hexitol backbone has a variable effect on selectivity. In the enzymatic assays, isoG misincorporates mainly with T, and isoC^Me misincorporates mainly with A. Further analysis in vivo confirmed the patterns of base‐pair interpretation for the deoxyribose and hexitol isoC^Me/isoG bases in a cellular context, through incorporation of the bases into plasmidic DNA. Results in vivo demonstrated that mispairing and misincorporation was dependent on the backbone scaffold of the base, which indicates rational advances towards orthogonality.     

Stimuli-responsive electrospun fibers and their applications

Stimuli-responsive electrospun nanofibers are gaining considerable attention as highly versatile tools which offer great potential in the biomedical field. In this critical review, an overview is given on recent advances made in the development and application of stimuli-responsive fibers. The specific features of these electrospun fibers are highlighted and discussed in view of the properties required for the diverse applications. Furthermore, several novel biomedical applications are discussed and the respective advantages and shortcomings inherent to stimuli-responsive electrospun fibers are addressed (136 references).     

Monitoring of tropospheric ozone in the ambient air with passive samplers

Two sampling campaigns in suburban places in the north zone of Santa Clara city, Cuba, have been carried out on a weekly base with the use of Radiello passive diffusion tubes in order to monitor the tropospheric ozone (O₃) levels in 2010. The first campaign was scheduled from February to April (cold season) and the second one in August and October (warm season), both of them at two sampling sites, i.e., Farm and School of Art Instructors. After aqueous extraction, the samples were analyzed by UV–VIS spectrophotometry.A seasonal trend was observed with the maximum O₃ concentrations in the cold season and the minimum levels in the warm season. Samples collected during the cold season showed the highest O₃ levels. Higher levels were reached at the Farm site with average values of about 58 ± 12 μg/m³, which exceeded the limit of the Cuban Standard 99:1999. In the warm season, the O₃ concentrations were similar for both sites, but lower than those observed in the cold season. The overall, seasonal average value was found to be 24 μg/m³. Despite the higher weekly average temperatures in August, the O₃ concentrations during this month showed the lowest values of the whole sampling period, which finding is in agreement with that reported by the Meteorological Institute of Cuba.Mathematical models, based on the Cochrane-Orcutt algorithm, were fitted to the acquired data set to explain the change in the tropospheric ozone concentrations under various meteorological conditions during the two campaigns. The correlation coefficients for both the cold and the warm seasons demonstrated a strong correlation, i.e., 0.779 and 0.951, respectively. The high correlation of wind speed in the model from the first sampling campaign explains the sharp decrease in O₃ concentrations at the SAI sampling site from the sixth week of sampling.
