Abstract— Chloroaluminum phthalocyanine (CAPC) is an efficient photosensitizer for the inactivation of Chinese hamster V79 cells. In order to investigate possible molecular mechanisms in the photo-dynamic action of CAPC and visible light, the induction and repair rate of two classes of DNA lesions have been determined, i.e. DNA single-strand breaks and DNA-protein cross-links. In cells pretreated with 1 μ.M CAPC, a fluence of 12 kJ/m2 of red light (>600 nm) kills approximately 50% of the cells and induces 3 to 3.5 Gy-equivalents of single-strand breaks. The repair of these breaks was slower than the repair of single-strand breaks induced by -irradiation. The photodynamic action of CAPC also induces a large number of DNA-protein cross-links which, in contrast to -radiation-induced DNA-protein cross-links, do not appear to be repaired during 4 h of post-treatment incubation in fresh medium. These studies suggest that DNA may be an important target for the cytotoxicity of CAPC + red light.  相似文献

A number of 2-X-3-nitro-1,8-naphthyridines (X= H,D,OH,Cl,NH2, OEt) react with the anion of chloromethyl phenyl sulfone exclusively into 2-X-3-nitro-4-(phenylsulfonylmethyl)-1,8-naphthyridines in high yield. The reaction is found by quantum chemical calculations to be controlled by the interactions of the HOMO of the nucleophile with the LUMO of the substrate, and not by charge.  相似文献

The electronic dipole transition moment functions of the A 2Π-X 2Σ+, B 2Σ+-X 2Σ+ and B 2Σ+-A 2Π transitions and the dipole moment function of the X 2Σ+ state of CO+ have been calculated using large contracted CI wavefunctions. The computed transition moment functions together with experimental potential energy curves were used to obtain radiative lifetimes of the excited electronic states B 2Σ+ and A 2Π. Radiative lifetimes of vibrational levels of the X 2Σ+ state were derived from the calculated dipole moment function. The high-frequency deflection technique was used to obtain radiative lifetimes of the ν′ = 0, 1,2 and 3 vibrational levels of the B 2Σ+ state and also radiative lifetimes of individual rotational levels of ν′ =0. The calculated radiative lifetimes are shorter than the measured ones by about 10%. The experimental ν′ dependence is reproduced by theoretical calculation. The calculated radiative lifetimes for the A 2Π state are in excellent agreement with lifetimes measured with an ion trap technique.  相似文献

Summary.  In this investigation the crystallization of PbTiO3 upon annealing of pure nanopowders and PbTiO3–SiO2 (1:1 v/v) nanocomposite powders prepared by the sol-gel technique was studied. Using X-ray diffraction phase analysis, the start of PbTiO3 crystallization in pure PbTiO3 powders was detected at 400°C. Distinct crystallization of PbTiO3 in PbTiO3–SiO2 nanocomposites starts at 700°C, whereas SiO2 remains amorphous. There are indications that an interface interaction between the PbTiO3 and the SiO2 phase plays an important role in hindering the crystallization of PbTiO3. The particle size (size of coherently scattering regions) was estimated from the broadening of the X-ray diffraction line profiles. The average size of PbTiO3 nanocrystallites increases with temperature and time of annealing, the influence of temperature being more significant than that of the annealing time. Differential scanning calorimetry confirmed the results of the X-ray diffraction with respect to the start of the crystallization. Laser beam scattering and scanning electron microscopy provided the statistical distribution of the grain size and the morphology of the powder grains, showing that each grain of the powders contains several nanocrystallites (coherently scattering regions). Consumers are avoiding the consumption of highly processed foods, aware of the negative effects of the additives or high temperatures used on the biological value of the food. This causes an interest in ways of minimal processing or low-temperature cooking procedures. However, to achieve the desired organoleptic quality, it is necessary to know the relationship between the parameters of the treatments and the type of raw material. The purpose of this study was to investigate the complex effects of traditional cooking and sous vide heat treatment, cold storage time and muscle on the physicochemical and sensory properties of beef. The study material consisted of samples of musculus longissimus thoracis and musculus semitendinosus obtained from beef half-carcasses. The muscles were subjected to traditional cooking in water at 95 °C until the temperature inside the piece reached 65 °C and sous vide treatment at 65 °C for 2 h. The study was performed after 2 and 21 days of cold storage. Instrumental evaluation of texture parameters, color and sensory evaluation of meat was carried out. Meat stored for 21 days was characterized by more favorable TPA test (Texture Profile Analysis) results compared to meat evaluated 48 h post mortem. The study also showed positive effects of sous vide heat treatment on texture parameters and sensory properties (especially on tenderness and palatability), as well as differences in the formation of quality traits between muscles. Given the trends associated with energy-saving technologies, it is desirable to seek the optimal combination of temperature and time of fixation treatments at an acceptable level of quality. The use of low-temperature cooking for as little as 2 h, yields positive results in sensory evaluation of juiciness, tenderness, or palatability.  相似文献

Quantum chemical studies employing combined density functional and multireference configuration interaction methods suggest five excited electronic states to be involved in the prompt and delayed fluorescence emission of TpAT-tFFO. Three of them, a pair of singlet and triplet charge transfer (CT) states (S1 and T1) and a locally excited (LE) triplet state (T3), can be associated with the (Me → N) conformer, the other two CT-type states (S2 and T2) form the lowest excited singlet and triplet states of the (Me → Ph) conformer. The two conformers, which differ in essence by the shearing angle of the face-to-face aligned donor and acceptor moieties, are easily interconverted in the electronic ground state whereas the reorganization energy is substantial in the excited singlet state, thus explaining the two experimentally observed time constants of prompt fluorescence emission. Forward and reverse intersystem crossing between the singlet and triplet CT states is mediated by vibronic spin–orbit interactions involving the LE T3 state. Low-frequency vibrational modes altering the distance and alignment of the donor and acceptor π-systems tune the S1 and T3 states (likewise S2 and T3) into and out of resonance. The enhancement of intersystem crossing due to the interplay of vibronic and spin–orbit coupling is considered a general feature of organic through-space charge-transfer thermally activated delayed fluorescence emitters.

The flexibility of dose and dosage forms makes 3D printing a very interesting tool for personalized medicine, with fused deposition modeling being the most promising and intensively developed method. In our research, we analyzed how various types of disintegrants and drug loading in poly(vinyl alcohol)-based filaments affect their mechanical properties and printability. We also assessed the effect of drug dosage and tablet spatial structure on the dissolution profiles. Given that the development of a method that allows the production of dosage forms with different properties from a single drug-loaded filament is desirable, we developed a method of printing ketoprofen tablets with different dose and dissolution profiles from a single feedstock filament. We optimized the filament preparation by hot-melt extrusion and characterized them. Then, we printed single, bi-, and tri-layer tablets varying with dose, infill density, internal structure, and composition. We analyzed the reproducibility of a spatial structure, phase, and degree of molecular order of ketoprofen in the tablets, and the dissolution profiles. We have printed tablets with immediate- and sustained-release characteristics using one drug-loaded filament, which demonstrates that a single filament can serve as a versatile source for the manufacturing of tablets exhibiting various release characteristics.  相似文献

Geometrical cis- and trans- isomers of 2-chloro-, 2-bromo- and 2-fluoro-4-methyl-1,3,2-dioxaphosphorinan-2-thiones were obtained in a diastereoselective way by (a) sulfurization of corresponding cyclic PIII-halogenides, (b) reaction of cyclic phosphorothioic acids with phosphorus pentachloride and (c) halogen–halogen exchange at PIV-halogenide. Their conformation and configuration at the C4-ring carbon and phosphorus stereocentres were studied by NMR (1H, 31P) methods, X-ray analysis and density functional (DFT) calculations. The stereochemistry of displacement reactions (alkaline hydrolysis, methanolysis, aminolysis) at phosphorus and its mechanism were shown to depend on the nature of halogen. Cyclic cis- and trans-isomers of chlorides and bromides react with nucleophiles (HO, CH3O, Me2NH) with inversion of configuration at phosphorus. DFT calculations provided evidence that alkaline hydrolysis of cyclic thiophosphoryl chlorides proceeds according to the SN2-P mechanism with a single transition state according to the potential energy surface (PES) observed. The alkaline hydrolysis reaction of cis- and trans-fluorides afforded the same mixture of the corresponding cyclic thiophosphoric acids with the thermodynamically more stable major product. Similar DFT calculations revealed that substitution at phosphorus in fluorides proceeds stepwise according to the A–E mechanism with formation of a pentacoordinate intermediate since a PES with two transition states was observed.  相似文献

Accumulation of heavy metals (HMs) by ornamental plants (OPs) from contaminated agriculture soils is a unique technique that can efficiently reduce the metal load in the food chain. Amaranthus tricolor L. has attractive characteristics acquiring a higher growth rate and large biomass when grown at heavy metal contaminated soils. Site-specific detailed information is not available on the use of A. tricolor plant in metal phytoremediation from the polluted sites. The study aimed to enhance the uptake of HMs (Pb, Zn, and Cu) via amending poultry litter extract (PLE), vinasse sugarcane (VSC), and humic acid (HA) as natural mobilized organic materials compared to ethylene diamine tetraacetic acid (EDTA), as a common mobilized chemical agent by A. tricolor plant. The studied soils collected from Helwan, El-Gabal El-Asfar (Cairo Governorate), Arab El-Madabeg (Assiut Governorate), Egypt, and study have been conducted under pot condition. Our results revealed all organic materials in all studied soils, except EDTA in EL-Gabal El-Asfar soil, significantly increased the dry weight of the A. tricolor plant compared to the control treatment. The uptake of Pb and Zn significantly (p > 0.05) increased due to applying all organic materials to the studied soils. HA application caused the highest uptake as shown in Pb concentration by more than 5 times in Helwan soil and EDTA by 65% in El-Gabal El-Asfar soil while VSC increased it by 110% in El-Madabeg soil. Also, an increase in Zn concentration due to EDTA application was 58, 42, and 56% for Helwan, El-Gabal El-Asfar, and El-Madabeg soil, respectively. In all studied soils, the application of organic materials increased the remediation factor (RF) than the control. El-Madabeg soil treated with vinasse sugarcane gave the highest RF values; 6.40, 3.26, and 4.02% for Pb, Zn, and Cu, respectively, than the control. Thus, we identified A. tricolor as a successful ornamental candidate that, along with organic mobilization amendments, most efficiently develop soil health, reduce metal toxicity, and recommend remediation of heavy metal-contaminated soils. Additionally, long-term application of organic mobilization amendments and continued growth of A. tricolor under field conditions could be recommended for future directions to confirm the results.  相似文献

Excessive UV solar radiation exposure causes human health risks; therefore, the study of multifunctional filters is important to skin UV protective ability and also to other beneficial activities to the human organism, such as reduction of reactive oxygen species (ROS) responsible for cellular damages. Potential multifunctional filters were obtained by intercalating of ferulate anions into layered simple metal hydroxides (LSH) through anion exchange and precipitation at constant pH methods. Ultrasound treatment was used in order to investigate the structural changes in LSH-ferulate materials. Structural and spectroscopic analyses show the formation of layered materials composed by a mixture of LSH intercalated with ferulate anions, where carboxylate groups of ferulate species interact with LSH layers. UV-VIS absorption spectra and in vitro SPF measurements indicate that LSH-ferulate systems have UV shielding capacity, mainly UVB protection. The results of reactive species assays show the ability of layered compounds in capture DPPH, ABTS•+, ROO, and HOCl/OCl reactive species. LSH-ferulate materials exhibit antioxidant activity and singular optical properties that enable their use as multifunctional filters.  相似文献
