DNA biosensors (or genosensors) are analytical devices that result from the integration of a sequence-specific probe and a signal transducer. Among other techniques, electrochemical and piezoelectric methods have recently emerged as the most attractive due to their simplicity, low instrumentation costs, possibility for real-time and label-free detection and generally high sensitivity.Focusing on the most recent activity of worldwide researchers, the aim of the present review is to give the readers a critical overview of some important aspects that contribute in creating successful genosensing devices. Advantages and disadvantages of different sensing materials, probe immobilisation chemistries, hybridisation conditions, transducing principles and amplification strategies will be discussed in detail. Dedicated sections will also address the issues of probe design and real samples pre-treatment. Special emphasis will be finally given to those protocols that, being implemented into an array format, are already penetrating the molecular diagnostics market.  相似文献

We present a combined computational and experimental study of the interaction of the Box A of the HMGB1 protein and carbenoxolone, an inhibitor of its pro-inflammatory activity. The computational approach consists of classical molecular dynamics (MD) simulations based on the GROMOS force field with quantum-refined (QRFF) atomic charges for the ligand. Experimental data consist of fluorescence intensities, chemical shift displacements, saturation transfer differences and intermolecular Nuclear Overhauser Enhancement signals. Good agreement is found between observations and the conformation of the ligand–protein complex resulting from QRFF–MD. In contrast, simple docking procedures and MD based on the unrefined force field provide models inconsistent with experiment. The ligand–protein binding is dominated by non-directional interactions.  相似文献

The reduction of [Ru(COD)(2-methylallyl) 2] (COD = 1,5-cyclooctadiene) dispersed in various room-temperature ionic liquids (ILs), namely, 1- n-butyl-3-methylimidazolium (BMI) and 1- n-decyl-3-methylimidazolium (DMI), associated with the N-bis(trifluoromethanesulfonyl)imidates (NTf 2) and the corresponding tetrafluoroborates (BF 4) with hydrogen gas (4 bar) at 50 degrees C leads to well-dispersed immobilized nanoparticles. Transmission electron microscopy (TEM) analysis of the particles dispersed in the ionic liquid shows the presence of [Ru(0)] n nanoparticles (Ru-NPs) of 2.1-3.5 nm in diameter. Nanoparticles with a smaller mean diameter were obtained in the ILs containing the less coordinating anion (NTf 2) than that in the tetrafluoroborate analogues. The ruthenium nanoparticles in ionic liquids were used for liquid-liquid biphasic hydrogenation of arenes under mild reaction conditions (50-90 degrees C and 4 bar). The apparent activation energy of E A = 42.0 kJ mol (-1) was estimated for the hydrogenation of toluene in the biphasic liquid-liquid system with Ru-NPs/BMI.NTf 2. TEM analysis of the ionic liquid material after the hydrogenation reactions shows no significant agglomeration of the [Ru(0)] n nanoparticles. The catalyst ionic liquid phase can be reused several times without a significant loss in catalytic activity.  相似文献

Treating wound infections is a difficult task ever since pathogenic bacteria started to develop resistance to common antibiotics. The present study develops hybrid hydrogels based on the formation of a polyelectrolyte complex between the anionic charges of dopamine-functionalized Gellan Gum (GG-DA) and the cationic moieties of the TMP-G2-alanine dendrimer. The hydrogels thus obtained can be doubly crosslinked with CaCl2, obtaining solid hydrogels. Or, by oxidizing dopamine to GG-DA, possibly causing further interactions such as Schiff Base and Michael addition to take place, hydrogels called injectables can be obtained. The latter have shear-thinning and self-healing properties (efficiency up to 100%). Human dermal fibroblasts (HDF), human epidermal keratinocytes (HaCaT), and mouse monocyte cells (RAW 264.7), after incubation with hydrogels, in most cases show cell viability up to 100%. Hydrogels exhibit adhesive behavior on various substrates, including porcine skin. At the same time, the dendrimer serves to crosslink the hydrogels and endows them with excellent broad-spectrum microbial eradication activity within four hours, evaluated using Staphylococcus aureus 2569 and Escherichia coli 178. Using the same GG-DA/TMP-G2-alanine ratios hybrid hydrogels with tunable properties and potential for wound dressing applications can be produced.  相似文献

Results are presented that were obtained on the geographic traceability of the white truffle Tuber magnatum Pico. Solid-phase microextraction coupled to gas chromatography/mass spectrometry (SPME-GC/MS) was employed to characterize the volatile profile of T. magnatum white truffle produced in seven geographical areas of Italy. The main components of the volatile fraction were identified using SPME-GC/MS. Significant differences in the proportion of volatile constituents from truffles of different geographical areas were detected. The results suggest that, besides genetic factors, environmental conditions influence the formation of volatile organic compounds. The mass spectra of the volatile fraction of the samples were used as fingerprints to characterize the geographical origin. Next, stepwise factorial discriminant analysis afforded a limited number of characteristic fragment ions that allowed a geographical classification of the truffles studied.  相似文献

Gold nanoparticles (AuNPs) coated with the diacetylene henicosa-10,12-diyn-1-yl (DS9) disulfide were successfully prepared by direct synthesis in toluene solutions. The average size of the nanohybrid metal core was finely adjusted by manipulation of the preparative conditions in the diameter range from 1.6 to 7.5 nm, as determined by TEM characterization. The topochemical polymerization of DS9 chemisorbed onto the gold nanoclusters of different size was carried out in colloidal suspensions by exposure to UV radiation and the process was monitored by UV-Vis and Raman spectroscopies. The results showed that in these assemblies the monomer undergoes an intra-particle polymerization and that the dominant polydiacetylene phase present is ruled by the core size. The deposition of the photoirradiated colloids onto different substrates was found to leave the polydiacetylene conjugation unaltered.  相似文献

This paper presents a study of thin films of the commercial ketone resins Laropal K80, Keton N and MS2A, attempting to reproduce the pictorial layers and protective finishes commonly present in contemporary paintings. Chemical and morphological changes due to the degradation effect of environmental agents have been specially considered. For this purpose, three different accelerated ageing processes were applied to a series of specimens prepared from the studied commercial products: thermal, UV light and ageing in an SO(2)-polluted chamber. Spectroscopic techniques such as FTIR spectroscopy and UV-Vis spectrophotometry were applied in combination with microscopic examination techniques, namely, AFM and scanning electron microscopy energy dispersive X-ray microanalysis (SEM-EDX). Chemical changes due to UV light and thermal ageing are in good agreement with those previously reported in the literature. Bleaching exhibited by the three commercial products after exposure to a SO(2)-saturated atmosphere has been related to the diffusion of SO(2)-rich water vapour into the film. This effect was particularly strong in the MS2A resin due to the higher content of hydroxyl groups in this product.  相似文献
