The results of spectroscopic structural and trace elemental analyses of mineral crystals produced by the soil nitrogen-fixing bacterium Azospirillum brasilense cultivated in a synthetic medium are presented and discussed. The mineral formed is shown to have a structure close to struvite (MgNH(4)PO(4) x 6H(2)O; ASTM file No. 15-762) with some differences which may be attributed to the presence of isomorphic admixtures of other cations (struvite is known to have a variety of forms). AAS/AES and ion chromatography analyses for a number of biologically important microelements and their role in the formation of the crystal structure, as well as some questions related to biomineralization are also discussed.  相似文献

Polyvinyl chloride samples of various tacticity have been prepared and photochlorinated in CCl4 for 4 hr. The chlorination degree correlates directly with the degree of syndiotacticity. Initial PVC with syndiotacticity about 56 per cent, which can be regarded as alternate sequences of syndiotactic and isotactic diads, is quickly chlorinated. The mechanism of chlorine attack on the macromolecula is directly connected with the stereoregularity.  相似文献

The changes in the layered structure of Mg-Al hydrotalcite (Mg/ Al = 2) during heat treatment have been investigated by using in situ XAFS simultaneously at the Mg and Al K-edges. The development of unique in situ instrumentation allowed the coordination environments at both the Mg and Al centers to be monitored as a function of the temperature and heat treatment. The results of this study show that the hydrotalcite structure is highly flexible, and should lead to the further development of hydrotalcites as new solid basic catalysts. Moreover, the Mg and Al cations in the cation layers show different behavior as a function of temperature. The coordination of some octahedral Al ions decreases already at a temperature of 425 K, whereas the coordination about Mg does not show any modification at this temperature. However, hydrotalcite treated at 425 K, followed by cooling down to room temperature resulted in a complete reversal to the original octahedral Al coordination. It is proposed that Al-OH bond breakage occurs at 425 K, without the evolution of H2O. This bond is restored after cooling to room temperature. The actual dehydroxylation of hydrotalcite commences between 425 and 475 K, as indicated by a change in coordination of both the Mg and Al centers. This is accompanied by the evolution of H2O molecules and the changes are hence irreversible without the presence of excess water. Heat treatment at 725 K leads to the development of an MgO-like phase (octahedral Mg) and a mixed octahedral/tetrahedral Al phase. A subsequent rehydration at room temperature entirely restores the original coordination about the Al and Mg centers of hydrotalcite to a distance of 15 A, to which XAFS spectroscopy is sensitive.  相似文献

ADH from human liver forms binary complexes with NADH, associated with a blue shift of the peak of the fluorescence emission of NADH. The wavelength shift is the same for all isoenzymes but the accompanying intensification of the fluorescence is different. The fluorescence is further increased by the formation of the very tight ternary enzyme-NADH-isobutyramide complexes. These properties are similar to those for the horse liver ADH, as well as the molecular weight of E=40 000 per active site of the dimer molecule (EE). "Stopped-flow" determined velocity constants (ER in equilibrium E+R) were found to be in good agreement with ethanol activity constants previously determined by activity measurement, confirming the validity of the ordered ternary complex mechanism also for the human ADH. No single isoenzyme activity as high as that reported by Mourad and Woronick or Drum has been found.  相似文献

Direct gas chromatographic methods to analyse phenylurea pesticides are discouraged by the thermal instability of these compounds, that in conventional hot splitless inlet systems leads to extensive and irreproducible formation of isocyanates and amines. However a careful control of the operating conditions, like the inlet temperature, the pressure and the presence of suitable chemical additives (as acetic acid, low-molecular-mass amines, organic anhydrides) can either: (i) minimise the thermal decomposition enabling the direct GC-MS analysis of phenylureas, or (ii) lead to reproducible conversion to isocyanates. Experimental design was employed to study the effect of the experimental variables on the thermal transformation of phenylurea pesticides in splitless inlet system. Two strategies were alternatively optimised: (i) the minimisation of degradation reactions to increase the signal of phenylureas; (ii) the maximisation of the degradation to isocyanates that are in turn determined. The maximal yields in isocyanate were obtained with high inlet temperatures, low carrier flows in the injection phase and the presence of acetic anhydride. By contrast, the use of relatively low inlet temperatures, high carrier flows during the injection and the presence of an amine maximise the response of the parent compounds.  相似文献

Automatic alignment (matching) of two-dimensional gel electrophoresis images is of primary interest in the evolving field of proteomics. In the present study, feature-based matching techniques, in their classical and robust versions, are described, and an automatic method of fuzzy alignment (FA) is introduced. This method allows automatic matching of two gel images with different numbers of features with unknown correspondence. Performance of FA is tested on simulated and real data sets.  相似文献

A method for the quantitative preconcentration of lead based on an existing batch process was developed for implementation in a flow system including a flame AAS detector. Lead can be quantitatively preconcentrated as pyrrolidinedithiocarbamate or dithizonate on an activated carbon minicolumn. The chelates are eluted in methyl isobutyl ketone and introduced directly into the nebuliser-burner. An enrichment factor of 50 is typically obtained for a preconcentration time of 2 min (lead can be determined at concentrations between 15 and 400 ng/ml), which results in a throughput of ca. 25 samples per hr. The sensitivity achieved with the two reagents is similar, but the selectivity provided by APDC exceeds that of dithizone. Based on the results obtained in the determination of lead in reference materials (minerals and skim milk), the proposed APDC method is applicable to real samples.  相似文献

In applying non-destructive neutron-activation analysis for gold and uranium in spiked low-grade ore samples, the following extrapolated unspiked concentrations were measured: 0.278 ± 0.015 ppm gold (chemical assay: 0.202 ± 0.020 ppm gold); 25.1 ± 1.1 ppm uranium (chemical assay value: 19.5 ±2.0 ppm uranium). Different approaches to the fitting of results, and the influence of spiking non-uniformity, are discussed.  相似文献

Summary This paper reports excess molar enthalpies of the binary systems hexane+ethyl benzene, hexane+o-xylene, hexane+m-xylene and hexane+p-xylene at 298.15 K and atmospheric pressure, over the whole composition range. The data was measured directly using a Calvet microcalorimeter. The excess magnitude was correlated to a Redlich-Kister type equation for each mixture. Also, we will discuss the results for the four mixtures studied here and by comparison with the same binary systems but containing propyl propanoate as first component. Finally, we will correlate our results with the Nitta-Chao and the three UNIFAC theoretical approximations.  相似文献

An indirect microdetermination of phosphate via EDTA titration is described, which can be applied to minerals, soils, fertilizers, biological samples, drugs and organo-phosphorus compounds. The method is based on the precipitation of phosphate as the very insoluble silver orthophosphate (Ksp = 1.3 × 10?20), dissolution of this precipitate in a solution of potassium cyanonickelate and titration of the nickel displaced by silver. The phosphate content is obtained indirectly by multiplying the number of ml of the titrant by a factor. The method takes about an hour after the phosphate is brought into solution. The accuracy is about 1% for samples containing 5–50 mg PO43? and about 3–5% for samples with 100 to 5000 μg PO43?.  相似文献
