Liquid-liquid phase asymmetric Darzens condensations were promoted by d-glucose- and d-mannose-based crown ethers. The corresponding aromatic and heteroaromatic α,β-epoxyketones were obtained with moderate to high enantioselectivities (up to 96%) as well as diastereoselectivities (up to 98:2) under mild reaction conditions. The absolute configurations of several of the epoxyketones were determined by single crystal X-ray analysis. The Michael additions of diethyl acetylaminomalonate to trans-β-nitroalkenes were carried out in a solid-liquid two-phase system in the presence of a d-glucose-based crown catalyst with up to 99% ee.  相似文献

Three different fluorescent tags have been compared for the quantitative analysis of aspartate and glutamate in brain microdialysate samples. Separation conditions have been optimized to achieve short analysis time using reversed polarity separation in coated capillary. Method validation has revealed similar quantification limit of 0.1?μM of analytes using either of the labels, although LOD values were different: 7.8-9.8?nM for 4-fluoro-7-nitro-2,1,3-benzoxadiazole, 3.5?nM for fluorescein-5-isothiocyanate and 1.3-1.5?nM for carboxyfluorescein succinimidyl ester derivatives. The almost two orders of magnitude difference between LOD and LOQ values is likely due to the unreliable derivatization reaction at low sample concentration. Based on the superior stability, FITC derivatization was used for the analysis of biological samples. The applicability of the method has been demonstrated by analyzing basal and potassium evoked amino acid concentrations in individual brain microdialysate samples.  相似文献

Lipofuscin is a fluorescent material with significant phototoxic potential that accumulates with age in the retinal pigment epithelium (RPE) of the eye. It is thought to be a factor in retinal degeneration diseases. The most extensively characterized lipofuscin component, N‐retinylidene‐N‐retinylethanolamine (A2E), has been proposed to be a byproduct of reactions involving the visual pigment chromophore. To examine the impact of the visual pigment and photoreceptor cell type on lipofuscin accumulation, we analyzed the RPE from Nrl?/? mice of various ages for lipofuscin fluorescence and A2E levels. The photoreceptor cells of the Nrl?/? retina contain only cone‐like pigments, and produce cone‐like responses to photostimulation. The cone‐like nature of these cells was confirmed by the presence of RPE65. Lipofuscin was measured with fluorescence imaging, whereas A2E was quantified by UV/VIS absorbance spectroscopy coupled to HPLC. The identity of A2E was corroborated with tandem mass spectrometry. Lipofuscin and A2E accumulated with age, albeit to lower levels compared with wild type mice. The emission spectra of RPE lipofuscin granules from Nrl?/? mice were similar to those from wild type mice, with λmaxca 610 nm. These results demonstrate that cone visual pigments can contribute to the production of lipofuscin and A2E.  相似文献

Purpose

Our aim was to characterize bi-exponential diffusion signal changes in normal appearing white matter of multiple sclerosis (MS) patients.

Methods

Diffusion parameters were measured using mono-exponential (0–1000 s/mm2) and bi-exponential (0–5000 s/mm2) approaches from 14 relapsing-remitting subtype of MS patients and 14 age- and sex-matched controls after acquiring diffusion-weighted images on a 3T MRI system. The results were analyzed using parametric or nonparametric tests and multiple linear regression models.

Results

Mono-exponential apparent diffusion coefficient (ADC) slightly increased in controls (P=.09), but decreased significantly in MS as a function of age, nonetheless an elevated ADC was observed with increasing lesion number in patients. Bi-exponential analyses showed that the increased ADC is the result of decreased relative volume fraction of slow diffusing component (fs). However, the fast and slow diffusion components (ADCf, ADCs) did not change as a function of either age in controls or lesion number and age in MS patients.

Conclusions

These data demonstrated that the myelin content of the white matter affects diffusion in relapsing-remitting subtype of multiple sclerosis that is possibly a consequence of the shift between different water fractions.  相似文献

The $[r,s,t]$ -chromatic number $\chi _{r,s,t}(G)$ of $G$ is defined to be the minimum $k$ such that $G$ admits an $[r,s,t]$ -coloring. In this note we examine $\chi _{1,1,t}(K_p)$ for complete graphs $K_p.$ We prove, among others, that $\chi _{1,1,t}(K_p)$ is equal to $p+t-2+\min \{p,t\}$ whenever $t \ge \left\lfloor {\frac{p}{2}}\right\rfloor -1,$ but is strictly larger if $p$ is even and sufficiently large with respect to $t.$ Moreover, as $p \rightarrow \infty $ and $t=t(p),$ we asymptotically have $\chi _{1,1,t}(K_p)=p+o(p)$ if and only if $t=o(p).$   相似文献

In this paper we give a brief review on the present status of automated detection systems describe for the screening of diabetic retinopathy. We further detail an enhanced detection procedure that consists of two steps. First, a pre-screening algorithm is considered to classify the input digital fundus images based on the severity of abnormalities. If an image is found to be seriously abnormal, it will not be analysed further with robust lesion detector algorithms. As a further improvement, we introduce a novel feature extraction approach based on clinical observations. The second step of the proposed method detects regions of interest with possible lesions on the images that previously passed the pre-screening step. These regions will serve as input to the specific lesion detectors for detailed analysis. This procedure can increase the computational performance of a screening system. Experimental results show that both two steps of the proposed approach are capable to efficiently exclude a large amount of data from further processing, thus, to decrease the computational burden of the automatic screening system.  相似文献

Negative ion desorption electrospray ionization (DESI) was used for the analysis of an ex vivo tissue sample set comprising primary colorectal adenocarcinoma samples and colorectal adenocarcinoma liver metastasis samples. Frozen sections (12 μm thick) were analyzed by means of DESI imaging mass spectrometry (IMS) with spatial resolution of 100 μm using a computer-controlled DESI imaging stage mounted on a high resolution Orbitrap mass spectrometer. DESI-IMS data were found to predominantly feature complex lipids, including phosphatidyl-inositols, phophatidyl-ethanolamines, phosphatidyl-serines, phosphatidyl-ethanolamine plasmalogens, phosphatidic acids, phosphatidyl-glycerols, ceramides, sphingolipids, and sulfatides among others. Molecular constituents were identified based on their exact mass and MS/MS fragmentation spectra. An identified set of molecules was found to be in good agreement with previously reported DESI imaging data. Different histological tissue types were found to yield characteristic mass spectrometric data in each individual section. Histological features were identified by comparison to hematoxylin-eosin stained neighboring sections. Ions specific to certain histological tissue types (connective tissue, smooth muscle, healthy mucosa, healthy liver parenchyma, and adenocarcinoma) were identified by semi-automated screening of data. While each section featured a number of tissue-specific species, no potential global biomarker was found in the full sample set for any of the tissue types. As an alternative approach, data were analyzed by principal component analysis (PCA) and linear discriminant analysis (LDA) which resulted in efficient separation of data points based on their histological types. A pixel-by-pixel tissue identification method was developed, featuring the PCA/LDA analysis of authentic data set, and localization of unknowns in the resulting 60D, histologically assigned LDA space. Novel approach was found to yield results which are in 95% agreement with the results of classical histology. KRAS mutation status was determined for each sample by standard molecular biology methods and a similar PCA/LDA approach was developed to assess the feasibility of the determination of this important parameter using solely DESI imaging data. Results showed that the mutant and wild-type samples fully separated. DESI-MS and molecular biology results were in agreement in 90% of the cases.  相似文献

We study the dynamics of a spin-dependent quantum dot system, where an unsharp and a sharp detection scenario is introduced. The back-action of the unsharp detection related to the magnetization, proposed in terms of the continuous quantum measurement theory, is observed via the von Neumann measurement (sharp detection) of the electric charge current. The behavior of the average electron charge current is studied as a function of the unsharp detection strength γγ, and features of measurement back-action are discussed. The achieved equations reproduce the quantum Zeno effect. Considering magnetic leads, we demonstrate that the measurement process may freeze the system in its initial state. We show that the continuous observation may enhance the transition between spin states, in contradiction with rapidly repeated projective observations, when it slows down. Experimental issue, such as the accuracy of the electric current measurement, is analyzed.  相似文献
