Subcultivation of Vero cells grown in a proprietary animal component-free medium named IPT-AFM, on microcarriers, was studied. TrypLE Select, a non-animal-derived protease, was used as an alternative to trypsin for cell passaging. We first studied the effect of increasing concentrations of TrypLE Select toward cell growth and then studied the inactivation of the protease using either soybean trypsin inhibitor (STI) or the soy hydrolysate Hypep 1510, in six-well plates. Data showed that cell growth was impaired by residual level of TrypLE Select; STI was identified as an efficient agent to neutralize this effect. To restore cell growth and inactivate TrypLE Select, STI should be added to the medium at least at 0.2 g L?1. Cells were also grown in spinner flask on 2 g L?1 Cytodex1 in IPT-AFM. In these conditions, the cell detachment yield was equal to 78?±?8 %. Furthermore, cells exhibited a typical growth profile when using the dislodged cells to seed a new culture. A cell detachment yield of 70?±?19 % was also achieved when the cells were grown in a 2-L stirred bioreactor in IPT-AFM, on 3 g L?1 Cytodex1. This protocol can be of great interest to scale-up the process of Vero cells cultivation in IPT-AFM on Cytodex1 from one stirred bioreactor culture to another.  相似文献

A number of molecular diagnostic methods have been developed for the detection and identification of mutations in tumor samples, which are important for the choice of treatment in the context of personalized medicine. For the treatment of metastatic melanoma, Vemurafenib is recommended for patients with BRAF V600 activating mutations. However, the different assays developed to date for the detection of these mutations lack sensitivity or specificity or do not allow a sequencing-based identification or validation of the mutation. Recently, enhanced improved and complete enrichment co-amplification at lower denaturation temperature-polymerase chain reaction (E-ice-COLD-PCR) has been developed as a sensitive method for the detection and identification of mutations in KRAS codons 12/13. Here, we present the first E-ice-COLD-PCR assay for the detection and identification of BRAF codon 600 mutations, which has a large dynamic range, as 25 pg to 25 ng can be used as DNA input without any reduction in mutation enrichment efficiency, and which can detect down to 0.01 % of mutated alleles in a wild-type background. The assay has been validated on fresh frozen, formalin-fixed paraffin-embedded (FFPE), and plasma samples of melanoma patients and has allowed the detection and identification of BRAF mutations present in samples appearing as wild type using standard pyrosequencing, endpoint genotyping, or Sanger sequencing. Thus, the BRAF V600 E-ice-COLD-PCR assay is currently one of the most powerful molecular diagnostic tools for the ultrasensitive detection and identification of BRAF codon 600 mutations.  相似文献

A highly efficient drug vector for photodynamic therapy (PDT) drug delivery was developed by synthesizing PEGylated gold nanoparticle conjugates, which act as a water-soluble and biocompatible "cage" that allows delivery of a hydrophobic drug to its site of PDT action. The dynamics of drug release in vitro in a two-phase solution system and in vivo in cancer-bearing mice indicates that the process of drug delivery is highly efficient, and passive targeting prefers the tumor site. With the Au NP-Pc 4 conjugates, the drug delivery time required for PDT has been greatly reduced to less than 2 h, compared to 2 days for the free drug.  相似文献

A self-setting cellulose scaffold, used as a component of an injectable bone substitute, was labeled with ruthenium tris-bipyridine complexes functionalized by triethoxysilane groups. The labeling yield was found to vary according to the number and location of Si(OR)3 groups present on the ruthenium complex backbone. For label loadings, less than 0.1 wt%, no significant modification of the rheological properties of the gel was observed.  相似文献

A new polyphenolic natural substance: 2,5-dihydroxy-benzoic acid 3'-formyl-5'-hydroxy-phenyl ester (1), Anamighrinal and 3-(O-alpha-L-rhamnosyl) quercetin (2) have been isolated from the methanolic aerial part extract of the plant Anagallis monelli by chromatographic separation. Their structures have been deduced essentially by one and two NMR spectroscopic procedures and mass spectrometry. Antioxidant, mutagenic, antimutagenic activities, of the natural products were realised and positive results were recorded.  相似文献

A method using safe and mild conditions to prepare water-core colloidosomes of a few micrometers in size is examined. Using poly(styrene-co-butylacrylate) colloidal particles of 180 nm in size, with a low-temperature glass transition of 30 or 42 degrees C, the self-assembly into micrometer-sized water-core polymer shell particles is demonstrated. The effect of oil phase type, surfactant and cosolvent, sintering time, and the method of transfer into an aqueous phase are all examined. The work demonstrates the production of water-core rigid-shell colloidosomes at low temperatures, between 35 and 65 degrees C.  相似文献

Ferrocenyl-chalcones and their bichalcone analogues were characterized by IR and NMR spectroscopy, as well as electrochemically. Their UV–visible spectra were recorded, and the electronic transitions were assigned by time-dependent DFT calculations.
