建立了同位素稀释质谱测定猪肉中氯霉素残留的方法,对影响溶剂提取、萃取净化、液相色谱-质谱检测方法准确度的因素进行了量化与优化。猪肉样品经液氮冷冻粉碎,加入氯霉素-D5同位素内标和0.1 mol/L醋酸钠缓冲溶液(pH 5),用乙酸乙酯提取4次,正己烷脱脂,HLB固相萃取小柱净化后采用负离子多反应监测模式下的液相色谱-质谱技术检测。方法的线性范围为0.01~0.5 ng/g,相关系数r2大于0.999;检出限和定量限分别为0.004 ng/g和0.02 ng/g;在0.02 ng/g和1.0 ng/g加标水平的回收率(n=3)分别为95.2%~109.1%和99.7%~102.5%;日内和日间相对标准偏差均小于2%。研究了氯霉素及氯霉素-D5的基体效应,测得不同流动相和稀释溶剂组成时基体效应因子k的范围在0.950~1.015之间。研究结果对同位素稀释质谱痕量残留检测结果准确度的控制、方法的建立与验证具有参考意义。 相似文献

The reaction of amino acids and phosphorus trichloride in THF was studied by 31P NMR tracing and ESI‐MS/MS. A series of hydridophoranes and cyclic dipeptides were obtained. The reaction presented interesting diversity and the reaction mechanism was proposed. The mechanism suggests that phosphorus plays an important role in the synthesis of amino acid hydridophorane and cyclic dipeptides. The results also show that 31P NMR and ESI‐MS/MS are useful tools for the investigation of reaction mechanism. 相似文献

A detailed characterization of intermediates in water oxidation catalyzed by a mononuclear Ru polypyridyl complex [Ru(II)-OH(2)](2+) (Ru = Ru complex with one 4-t-butyl-2,6-di-(1',8'-naphthyrid-2'-yl)-pyridine ligand and two 4-picoline ligands) has been carried out using electrochemistry, UV-vis and resonance Raman spectroscopy, pulse radiolysis, stopped flow, and electrospray ionization mass spectrometry (ESI-MS) with H(2)(18)O labeling experiments and theoretical calculations. The results reveal a number of intriguing properties of intermediates such as [Ru(IV)═O](2+) and [Ru(IV)-OO](2+). At pH > 2.9, two consecutive proton-coupled one-electron steps take place at the potential of the [Ru(III)-OH](2+)/[Ru(II)-OH(2)](2+) couple, which is equal to or higher than the potential of the [Ru(IV)═O](2+)/[Ru(III)-OH](2+) couple (i.e., the observation of a two-electron oxidation in cyclic voltammetry). At pH 1, the rate constant of the first one-electron oxidation by Ce(IV) is k(1) = 2 × 10(4) M(-1) s(-1). While pH-independent oxidation of [Ru(IV)═O](2+) takes place at 1420 mV vs NHE, bulk electrolysis of [Ru(II)-OH(2)](2+) at 1260 mV vs NHE at pH 1 (0.1 M triflic acid) and 1150 mV at pH 6 (10 mM sodium phosphate) yielded a red colored solution with a Coulomb count corresponding to a net four-electron oxidation. ESI-MS with labeling experiments clearly indicates that this species has an O-O bond. This species required an additional oxidation to liberate an oxygen molecule, and without any additional oxidant it completely decomposed slowly to form [Ru(II)-OOH](+) over 2 weeks. While there remains some conflicting evidence, we have assigned this species as (1)[Ru(IV)-η(2)-OO](2+) based on our electrochemical, spectroscopic, and theoretical observations alongside a previously reported analysis by T. J. Meyer's group (J. Am. Chem. Soc. 2010, 132, 1545-1557). 相似文献

Protein methylation is involved in dozens of biological processes and plays an important role in adjusting protein physicochemical properties, conformation and function. However, with the rapid increase of protein sequence entering into databanks, the gap between the number of known sequence and the number of known methylation annotation is widening rapidly. Therefore, it is vitally significant to develop a computational method for quick and accurate identification of methylation sites. In this study, a novel predictor (Methy_SVMIACO) based on support vector machine (SVM) and improved ant colony optimization algorithm (IACO) is developed to identify methylation sites. The IACO is utilized to find the optimal feature subset and parameter of SVM, while SVM is employed to perform the identification of methylation sites. Comparison of the IACO with conventional ACO shows that the IACO converges quickly toward the global optimal solution and it is more useful tool for feature selection and SVM parameter optimization. The performance of Methy_SVMIACO is evaluated with a sensitivity of 85.71%, a specificity of 86.67%, an accuracy of 86.19% and a Matthew's correlation coefficient (MCC) of 0.7238 for lysine as well as a sensitivity of 89.08%, a specificity of 94.07%, an accuracy of 91.56% and a MCC of 0.8323 for arginine in 10-fold cross-validation test. It is shown through the analysis of the optimal feature subset that some upstream and downstream residues play important role in the methylation of arginine and lysine. Compared with other existing methods, the Methy_SVMIACO provides higher Acc, Sen and Spe, indicating that the current method may serve as a powerful complementary tool to other existing approaches in this area. The reaction of 2,9-di(pyrid-2'-yl)-1,10-phenanthroline (dpp) with [RuCl(3)·3H(2)O] or [Ru(DMSO)(4)Cl(2)] provides the reagent trans-[Ru(II)(dpp)Cl(2)] in yields of 98 and 89%, respectively. This reagent reacts with monodentate ligands L to replace the two axial chlorides, affording reasonable yields of a ruthenium(II) complex with dpp bound tetradentate in the equatorial plane. The photophysical and electrochemical properties of the tetradentate complexes are strongly influenced by the axial ligands with electron-donating character to stabilize the ruthenium(III) state, shifting the metal-to-ligand charge-transfer absorption to lower energy and decreasing the oxidation potential. When the precursor trans-[Ru(II)(dpp)Cl(2)] reacts with a bidentate (2,2'-bipyridine), tridentate (2,2';6,2'-terpyridine), or tetradentate (itself) ligand, a peripheral pyridine on dpp is displaced such that dpp binds as a tridentate. A new triptycene-derived macrotricyclic host containing two dibenzo-[18]-crown-6 moieties was synthesized and shown to form 1:1 complexes with paraquat derivatives in solution, in which the guests all thread the central cavity of the host. However, it was interestingly found that, depending on the paraquat derivatives with different functional groups, the host can form stable 1:1 or 1:2 complexes in different complexation modes in the solid state, which is significantly different from those of the macrotricyclic host containing two dibenzo-[24]-crown-8 moieties. The formation of the complexes was also proved by the ESI MS and electrochemical experiments. We report on a rapid method for the detection of Salmonella O8. It does not require an enrichment step but rather uses an aptamer as a probe that was selected by system evolution of ligands by exponential enrichment (SELEX) assay. Firstly, aptamer against Salmonella O8 was selected from a 78 bp random DNA library that was prepared in-vitro. The binding ability of the aptamers to target bacterium was examined by aptamer-linked immobilized sorbent assay. A high affinity aptamer was successfully selected from the initial random DNA pool, and its secondary structure was also investigated. Next, this high affinity aptamer B10 was used to recognize Salmonella O8 via fluorescence microscopy. The selected aptamer has a high specificity and high affinity against its target. We believe that the resulting fluorescence in-situ labeling assay is a potentially useful alternative in rapid screening and detection of foodborne pathogens.

A facile approach has been developed for the preparation of various morphologies of Au–Ag2Te nanomaterials (NMs) that exhibit strong photocatalytic activity. Te NMs (nanowires, nanopencils, and nanorice) were prepared from TeO2 in the presence of various concentrations (16, 8, and 4 M ) of a reducing agent (N2H4) at different temperatures (25 and 60 °C). These three Te NMs were then used to prepare Au–Ag2Te NMs by spontaneous redox reactions with Au3+ and Ag+ ions sequentially. The Au–Ag2Te nanopencils exhibit the highest activity toward degradation of methylene blue and formation of active hydroxyl radicals on solar irradiation, mainly because they absorb light in the visible region most strongly. All three differently shaped Au–Ag2Te NMs (10 μg mL?1) provide a death rate of Escherichia coli greater than 80 % within 60 min, which is higher than that of 51 % for commercial TiO2 nanoparticles (100 μg mL?1). Under light irradiation, the Au NPs in Au–Ag2Te NMs enhance the overall photo‐oxidation ability of Ag2Te NMs through faster charge separation because of good contact between Ag2Te and Au segments. With high antibacterial activity and low toxicity toward normal cells, the Au–Ag2Te NMs hold great potential for use as efficient antibacterial agents. 相似文献

The detection and identification of epidermal growth factor receptor 2 (HER2)-positive breast cancer cells is crucial for
the clinic therapy of breast cancer. For the aim of the detection, a novel surface-enhanced Raman scattering (SERS) probe
for distinguishing breast cancers at different HER2 statuses is reported in this paper. In such a probe, anti-HER2 antibody-conjugated
silver nanoparticles have been synthesized for specific targeting of HER2-positive breast cancer cells. More importantly,
different from the previously reported SERS probe for targeting cancer cells, p-mercaptobenzoic acid is utilized as both the Raman reporter and the conjugation agent for attaching antibody molecules, which
leads to a much simplified structure. For investigating the ability of such a probe to distinguish breast cancer cells, SKBR3
and MCF7 cells were chosen as two model systems, which are HER2-positive- and HER2-negative-expressing cells, respectively.
The experimental results reveal that SKBR3 cells exhibit much stronger SERS signals than MCF7 cells, indicating that the probe
could be utilized to distinguish breast cancer cells at different HER2 statuses. This kind of SERS probe holds a potential
for a direct detection of living breast cancer cells with the advantages of easy fabrication, high SERS sensitivity, and biocompatibility. 相似文献
