Development of novel active acceptors possessing a positively charged structure for the transglycosylation reaction with Endo-M and their application to oligosaccharide analysis

With Boc-Asn-GlcNAc as a basic structure, four permanently positively charged kinds of new acceptors (GP-Boc-Asn-GlcNAc, GT-Boc-Asn-GlcNAc, HMP-Boc-Asn-GlcNAc, MPDPZ-Boc-Asn-GlcNAc) and five kinds of similar structure acceptors (2-PA-Boc-Asn-GlcNAc, 3-PA-Boc-Asn-GlcNAc, 4-PA-Boc-Asn-GlcNAc, HP-Boc-Asn-GlcNAc, PDPZ-Boc-Asn-GlcNAc) were synthesized as acceptors for the resolution of oligosaccharides in glycopeptides. The synthesized acceptors enzymatically reacted with Disialo-Asn (donor) in the presence of Endo-M. The reaction yields of each transglycosylation product were not obvious, because we do not have all the authentic Disialo-Asn-Boc-acceptors. Therefore, we used the peak area of the transglycosylation product detected by mass spectrometry and evaluated the utility of each acceptor. Among the Boc-Asn-GlcNAc acceptors, the positively charged MPDPZ derivative peak area was the highest, MPDPZ-Boc-Asn-GlcNAc with a positively charged structure showed about a 2.2 times greater sensitivity of the transglycosylation product compared to the conventional fluorescence acceptor DBD-PZ-Boc-Asn-GlcNAc. As a result, the MPDPZ-Boc-Asn-GlcNAc acceptor was suitable for the transglycosylation reaction with Endo-M. The development of a qualitative determination method for the N-linked oligosaccharides in glycoproteins was attempted by combination of the transglycosylation reaction and semi-micro high-performance liquid chromatography/electrospray ionization quadrupole time-of-flight tandem mass spectrometry (HPLC/ESI-QTOF-MS/MS). The asparaginyl-oligosaccharides in glycoproteins, liberated by treatment with Pronase E, were separated, purified and labeled with positively charged MPDPZ. The resulting derivatives were separated by a semi-micro HPLC system. The eluted N-linked oligosaccharide derivatives were then introduced into a QTOF-MS instrument and sensitively detected in the ESI(+) mode. Various fragment ions based on the carbohydrate units appeared in the MS/MS spectra. Among the peaks, m/z 782.37 corresponding to MPDPZ-Boc-Asn-GlcNAc is the most important one for identifying the asparaginyl-oligosaccharides. Disialo-Asn-Boc-MPDPZ was easily identified by the selected-ion chromatogram at m/z 782.37 by MS/MS detection. Therefore, the identification of N-linked oligosaccharides in glycoproteins seems to be possible by the proposed semi-micro HPLC separations followed by the QTOF-MS/MS detection. Furthermore, several oligosaccharides in ovalbumin and ribonuclease B were successfully identified by the proposed procedure.     

Au-ZnO hybrid nanopyramids and their photocatalytic properties

We successfully synthesized Au-ZnO hybrid nanoparticles with a novel hexagonal pyramid-like structure. The growth process of the as-prepared hybrid nanopyramids is clearly discussed. Because of their homogeneous composition and controlled morphology, the Au-ZnO hybrid nanopyramids demonstrate better photocatalytic efficiency than pure ZnO nanocrystals.     

Influence of bromoalkyloxy side chain on mesomorphic behavior in heterocyclic 7-(4-bromoalkyloxy)-3-(4′-decyloxyphenyl)-4H- 1-benzopyran-4-ones

A new homologous series of isoflavone-based ethers,7-(4-bromoalkyloxy)-3-(4’-decyloxyprienyl)-4H-1-benzopyran-4-ones were synthesized and characterized.The mesomorphic properties of all homologues were investigated by differential scanning calorimetry(DSC) and polarizing optical microscopy(POM).Enantiotropic smectic A(SmA) phase was observed for all homologues.The structure-property study was carried out by comparing the mesomorphic behavior of the homologues with those previously reported analogues.The bromine atom from the alkyloxy side chain of the benzene ring fused to the heterocyclic moiety was found to be capable of changing the mesomorphic properties.     

A spirocyclic oxindole analogue:Synthesis and antitumor activities

A new synthesis of spirocyclic oxindole analogue spiro[piperidine-4,3’-pyrrolo[2,3-b]pyridin]-2’（1’H）-one 1 is described.The key steps involve dialkylation of arylacetonitrile and cyclization of the azaoxindole ring by an intramolecular Buchwald-Hartwig amidation of carboxylic ami Je and aryl chloride.A small library was obtained by reductive amination of 1 with various aldehydes and was screened against human lung cancer cell A549,human liver cancer cell BEL7402,and human colon cancer cell HCT-8.The results show that most of the 1（?）rary compounds 2 have some inhibitory activities.2-（Trifluoromethoxy） benzylic substituted spirocyclic azaoxindole 2e was identified as a nanomolar inhibitor against human lung cancer cell A-549(IC₅₀=50 nmol/L).     

Friedel-Crafts alkylation of indoles with nitroalkenes catalyzed by Cu(Ⅱ)-imine complex

A series of new ligands L₁-L₇ were readily prepared in one step.Friedel-Crafts alkylation of indoles with nitroalkenes catalyzed by a novel Cu（Ⅱ）-L complex has been developed.The remarkable advantages of this reaction are mild reaction conditions,simple workup procedure,high yields of products and the use of ethanol as a green solvent.     

Pyridinium tribromide catalyzed condensation of indoles and aldehydes to form bisindolylalkanes

An efficient synthetic method for bis(indol-3-yl)alkane derivatives has been developed.In the presence of 5 mol%of pyridinium tribromide(PTB),the condensation of indoles and aldehydes proceeded smoothly under mild conditions,giving rise to the corresponding bis(indol-3-yl)alkanes in good to excellent yields.     

Melamine-(H_2SO_4)_3 and PVP-(H_2SO_4)_n as solid acids:Synthesis and application in the first mono-and di-nitration of bisphenol A and other phenols

Melamine and poly vinylpyrrolidone（PVP） reacted with neat sulfuric acid readily to form two new organic solid acids namely melamine-（H₂SO₄）₃ and PVP-（H₂SO₄）_n.These solid acids were used for the first nitration of bisphenol A as well as other phenols in the presence of NH₄NO₃.Mono- and di-nitro bisphenol A have been characterized with IR and ¹H NMR techniques.     

P-Dodecylbenzenesulfonic acid(DBSA),a Brфnsted acid-surfactant catalyst for Biginelli reaction in water and under solvent free conditions

We report herein the use of p-dodecylbenzenesulfonic acid(DBSA) as a catalyst for a one-pot Biginelli reaction to afford 3,4- dihydropyrimidinone derivatives in good to excellent yields.This reaction proceeds efficiently in water and under solvent free conditions.Comparisons of results indicate that although the yields are high and comparable for both methods,the reaction times are considerably shorter under solvent free conditions.
