采用毛细管电泳-电化学检测法(CE-ED)同时分离测定了中药杠板归中的阿魏酸、香草酸、槲皮素、咖啡酸、原儿茶酸等主要生物活性成分的含量。考察了工作电极电位、运行缓冲液的pH值和浓度、分离电压和进样时间等实验参数对实验结果的影响。在优化的实验条件下,以直径300 μm的碳圆盘电极为工作电极,检测电位为+0.95V(vs. SCE),在10mmol/L磷酸盐(pH 9.2)的运行缓冲溶液中,五个分析物能够在17min内实现很好的基线分离,被测物浓度与峰电流在3个数量级呈良好的线性,检测限(S/N=3)范围从7.1×10-8 到 9.3×10-8g mL-1。该方法已应用于实际样品的分析,样品处理简单,获得了令人满意的结果。  相似文献

Photochromic behavior of Li-doped MoO3 sol–gels prepared by the peroxo sol–gel method was studied in details using various experimental techniques, including UV–vis, XRD, Raman, EPR, and XPS. The lithium doping has drastically enhanced the stability of the MoO3 sol–gels. Upon UV-light irradiation from a high-pressure mercury lamp, the Li-doped MoO3 sol turned from yellowish into deep blue, and the sol in ethanol exhibited much more intense color change than the sol in water. The UV-light exposure has the effect of re-arranging the structural units of MoO3 and building up short-range order inside the solid. The formation of a blue colored bronze in the photo-irradiated sols because of the reduction of Mo6+ to Mo5+ is also evidenced by the experimental data. Based on these data, a mechanism for the photochromic behavior is proposed.  相似文献

本文对草酸修饰电极的制作、痕量铅在草酸修饰电极上的反应机理和电化学特性作了研究。采用阳极溶出伏安法,测定铅的灵敏度比玻碳电极提高7.5倍,在1.0~15.0ng·/mL铅浓度范围内溶出峰高与铅浓度呈良好的线性关系。连续测定12次变异系数为3.0%。利用草酸修饰电极对人尿中痕量铅进行了测定,结果良好。  相似文献

A series of novel cationic functional hexaalkylguanidinium ionic liquids and anionic functional tetraalkylguanidinium ionic liquids have been synthesized, and then magnetic chitosan graphene oxide (MCGO) composite has been prepared and coated with these functional guanidinium ionic liquids to extract protein by magnetic solid-phase extraction. MCGO-functional guanidinium ionic liquid has been characterized by vibrating sample magnetometer, field emission scanning electron microscopy, X-ray diffraction spectrometer and Fourier transform infrared spectrometer. After extraction, the concentrations of protein were determined by measuring the absorbance at 278 nm using an ultra violet visible spectrophotometer. The advantages of MCGO-functional guanidinium ionic liquid in protein extraction were compared with magnetic chitosan, graphene oxide, MCGO and MCGO-ordinary imidazolium ionic liquid. The proposed method has been applied to extract trypsin, lysozyme, ovalbumin and bovine serum albumin. A comprehensive study of the adsorption conditions such as the concentration of protein, the amount of MCGO-functional guanidinium ionic liquid, the pH, the temperature and the extraction time were also presented. Moreover, the MCGO-functional guanidinium ionic liquid can be easily regenerated, and the extraction capacity was about 94% of the initial one after being used three times.  相似文献

研究了新显色剂4,4’-二[(8-羟基喹啉)偶氮]-3,3’-二甲基联苯(D-HQADMB)与Cd(Ⅱ)的显色反应条件。在吐温-80存在下的pH7.8~9.0的缓冲介质中,Cd(Ⅱ)与D-HQADMB形成稳定的红色配合物,组成比为Cd(Ⅱ)+D-HQADMB=1+2,λmax=530nm,表观摩尔吸光系数为7.20×104L/mol·cm,Cd量在0~1.3μg/mL范围内符合比耳定律。用制定的方法测定矿样中的微量镉,结果满意。  相似文献

A new and sensitive electrochemical DNA hybridization detection assay, using tris(2,2′-bipyridyl)cobalt(III) [Co(bpy)33+]-doped silica nanoparticles as the oligonucleotide (ODN) labeling tag, and based on voltammetric detection of Co(bpy)33+ inside silica nanoparticles, is described. Electro-active Co(bpy)33+ is not possible for directly linking with DNA, it is doped into the silica nanoparticles in the process of nanoparticles synthesis for DNA labeling with trimethoxysilylpropydiethylenetriamine (DETA) and glutaraldehyde as linking agents. The Co(bpy)33+ labeled DNA probe is used to hybridize with target DNA immobilized on the surface of glassy carbon electrode. Only the complementary sequence DNA (cDNA) could form a double-stranded DNA (dsDNA) with the DNA probe labeled with Co(bpy)33+ and give an obvious electrochemical response. A three-base mismatch sequence and non-complementary sequence had negligible response. Due to the large number of Co(bpy)33+ molecules inside silica nanoparticles linked to oligonucleotide DNA probe, the assay showed a high sensitivity. It allows the detection at levels as low as 2.0×10−10 mol l−1 of the target oligonucleotides.  相似文献

以一种基于目标蛋白凝血酶的取代反应来研究核酸适体与互补核酸和核酸适体与目标蛋白之间竞争结合的热力学特性, 研究了核酸适体-互补核酸的解离反应和在目标蛋白存在下取代反应的平衡常数、标准焓变和标准熵变等热力学参数, 结果表明该取代反应是一个熵驱动的自发过程, 熵驱动从双链的核酸转变为核酸适体-目标蛋白的复合物. 该热力学研究会对核酸适体-互补核酸和核酸适体-凝血酶之间的结合过程的机理有一个更深的理解, 将有助于进一步揭示核酸与蛋白这两种生命中最关键物质之间的相互作用和关系, 为更好地理解基本的生物过程和预测设计适体生物传感器, 发展用于疾病诊断的方法有着重要的意义.  相似文献

By constructing a genomic library, an endoglucanase gene (cel9P) was cloned from Paenibacillus sp. BME-14 which was isolated from the sea. It had an open-reading frame of 1,629 bp, encoding a peptide of 542-amino acid residue with a calculated molecular mass of 60 kDa. The enzyme showed the highest amino acid identity of 52% with other known endoglucanases and had a C-terminal catalytic domain belonging to the glycosyl hydrolases family 9. The optimum pH and temperature for enzymatic activity was pH 6.5 and 35 °C. The metal ions of Ca2+, Mg2+, and Mn2+ had a positive effect on the activity while Hg2+, Cu2+, and EDTA had a negative effect. Notably, Cel9P had 65% of the maximal activity at 5 °C. Based on the special characteristic of Cel9P, it had a potential significance for study of cold-active mechanism and industry applications.  相似文献

本采用四硫富瓦烯(TTF)作为葡萄糖氧化酶与玻碳电极之间的电子媒介体,把葡萄糖氧化酶因定在Nafion-TTF修饰电极上,制成葡萄糖生物传感器。用这种传感器测定人体血清中葡萄糖的含量,其线性范围在4.0×10^-5~10^-3mol/L之间,响应时间为20s。该传感器具有选择性、重现性好,灵敏度高等优点。  相似文献

Granular, needle-, rod-, or whisker-shaped gamma-Fe2O3 nanocrystals have been prepared via controlling the chemical microenvironment of the reaction system under gamma-irradiation. A novel extension-curl-effect model was proposed to explain the effect of the chemical microenvironment on the morphology of gamma-Fe2O3 nanocrystals. The as-prepared gamma-Fe2O3 nanocrystals were characterized by powder X-ray diffraction and transmission electron microscopy. Their magnetic properties were also studied by zero-field M?ssbauer spectroscopy, which indicated that different shaped gamma-Fe2O3 nanocrystals could exhibit interesting magnetic behavior.  相似文献
