A carbonyl ylide cycloaddition approach to the squalene synthase inhibitors zaragozic acids A and C is described. The carbonyl ylide precursor 8 was synthesized starting from di-tert-butyl D-tartrate (47) via an eleven-step sequence involving the regioselective reduction of the mono-MPM (MPM=4-methoxybenzyl) ether 48 with LiBH4 and the diastereoselective addition of sodium tert-butyl diazoacetate to alpha-keto ester 10. The reaction of alpha-diazo ester 8 with 3-butyn-2-one (40) in the presence of a catalytic amount of [Rh2(OAc)4] gave the desired cycloadduct 59 as a single diastereomer. The dihydroxylation of enone 59 followed by sequential transformations permitted the construction of the fully functionalized 2,8-dioxabicyclo[3.2.1]octane core 5. Alkene 79 derived from 5 serves as a common precursor to zaragozic acids A (1) and C (2), since the elongation of the C1 alkyl side chain can be attained by olefin cross-metathesis, especially under the influence of Blechert's catalyst (85).  相似文献

A simple and efficient one-pot procedure for the synthesis of α-diketones from aldehydes via benzoin condensation under the influence of a catalytic amount of azolium salt combined with DBU has been developed. Thus, aldehyde was allowed to react with the azolium salt/DBU catalytic system at room temperature, and then the reaction mixture was heated to 70 °C under air atmosphere to afford the corresponding 1,2-diketone in good yield. This would be an efficient alternative method of synthesizing α-diketones from aldehydes under metal-free conditions.  相似文献

Scattering of the light beam propagating through a dielectric-slab optical waveguide occurs due to refractive index inhomogeneities of the thin film region and due to boundary irregularities of the filmsubstrate and the film-air interfaces. The quantity and direction of the scattered light are evaluated by means of a perturbation method together with the use of a stationary phase method, for a variety of correlation lengths and variances and the thin film thicknesses. The results show that for a slab waveguide of t/=10.0,t and being the thickness of thin film and the wavelength of light, the effect of refractive index inhomogeneities is pronounced in comparison with that of waveguide wall irregularities. In this case, therefore, the scattering pattern is determined mainly by the correlation length of refractive index inhomogeneities.  相似文献

The bitterness of 11 commercial medicines was evaluated both by a multichannel taste sensor and in human gustatory sensation tests with 15 volunteers. For basic drugs with amino groups in the molecule, such as quinine, there was a comparatively strong relative response electric potential (mV) of channels 1 or 2, those containing negatively charged membranes and the bitterness determined by human gustatory sensation tests. The suppression of the bitterness of quinine by sucrose and aspartame could be quantified using the artificial taste sensor and the results concurred with those from gustatory sensation tests. The usefulness of the sensor was thus confirmed for this type of compound. Anionic drugs, such as diclofenac sodium or salicylic acid gave rise in a negative response electric potential in channels 5 or 6, those containing positively charged membrane, seemed to be useful information even though their tastes are being sour rather than bitter. For drugs with both an amino (cationic) group and carboxylic acid (anionic) group in the molecule, such as theophylline, caffeine, and metronidazole, the relative response electric potential (mV) of channels containing negatively charged membranes was not increased, even though bitterness was observed in human gustatory sensation tests. Therefore, a different design of membrane component is required for more general evaluation of the bitterness of various medicines.  相似文献

A newly designed anthryl dendron having perfluoroalkyl chains at terminals showed thermotropic liquid crystallinity, which was characterized using polarizing optical microscopy, differential scanning calorimetry, and X-ray diffraction. The anthryl dendron forms SB phase at room temperature below 53 °C, at which temperature a phase transition to Colrd takes place. In a fluorous solvent, regioselective photodimerization of the anthryl dendron occurred to give head-to-head photodimers, although photodimerization in the SB phase and chloroform gave both head-to-head and head-to-tail photodimers.  相似文献

The electron beam ion source (Kobe EBIS) has been developed to perform modification of surfaces using highly charged ions (HCIs) at the Kobe University, Japan. Recent study revealed that periodic intermission of electron beam improves charge state distribution of extracted ions. The period of intermission is in the order of 100 ms, and the width of beam-off time is 1 ms or less. This operational mode (pulse mode) makes it possible to produce Ar15+ to Ar17+ effectively, whereas the charge is limited less than 14+ under the ordinary operational mode with direct current (DC) electron beam. A spike of HCIs with a peak current in the order of nA is also observed at each moment of electron beam off. The measurement of the time evolution of Ar16+ intensity around the timing of mode change revealed that the intensity of extracted Ar16+ changes slowly after mode change with a time constant of few seconds.  相似文献

A series of tin‐doped hydroxypropyl cellulose (HPC) lyotropic liquid crystal (LC) was synthesized using a simple process and their properties were characterized using selective reflection, wide‐angle X‐ray diffraction (WAXD), and the band texture observed under polarized optical microscope. The present preparation is applicable for mass production using large substrate with low cost HPC. A cholesteric lyotropic LC phase was observed for the hybrid solution with higher than 40 wt % HPC. After sol–gel condensation, the HPC‐Sn hybrid LC films were calcined at 400 °C and the as‐prepared product was determined to obtain tin dioxide (SnO2) which was characterized using WAXD. The iridescent color and ~2 nm structure seen after the condensation disappeared in the as‐prepared SnO2. Scanning electronic microscope images of the SnO2 showed that the HPC content in the HPC‐Sn hybrid played an important role in controlling the SnO2 morphology. A spectrum of relatively monochromatic extreme ultraviolet (13.5 nm) emission was measured in the as‐prepared SnO2 in comparison with bulk tin and inverse opal SnO2. © 2009 Wiley Periodicals, Inc. We report the first observations of anomalous infrared and visible light absorption of spherical gold nanoparticles dispersed in a comb copolymer. The dependence of the infrared absorption of the C-O stretch band of the comb copolymer on the volume fraction and size of the gold nanoparticles is measured. Absorption intensities increase when the volume fraction is increased from 0.05 to 0.57 or when the particle size is reduced from 40 to 15 nm. We also show that the experimental transmittances of gold nanoparticles in the visible region are not consistent with a theory of infrared absorption enhancement based on collective electron resonance.
