We report a novel method of one-step direct amination on polycrystalline diamond to produce functionalized surfaces for DNA micropatterning by photolithography. Polycrystalline diamond was exposed to UV irradiation in ammonia gas to generate amine groups directly. After patterning, optical microscopy confirmed that micropatterns covered with an Au mask were regular in size and shape. The regions outside the micropatterns were passivated with fluorine termination by C3F8 plasma, and the chemical changes on the two different surfaces--the amine groups inside the patterned regions by one-step direct amination and fluorine termination outside the patterned regions--were characterized by spatially resolved X-ray photoelectron spectroscopy (XPS). The patterned areas terminated with active amine groups were then immobilized with probe DNA via a bifunctional molecule. The sequence specificity was conducted by hybridizing fluorescently labeled target DNA to both complementary and noncomplementary probe DNA attached inside the micropatterns. The fluorescence micropatterns observed by epifluorescence microscopy corresponded to those imaged by optical microscopy. DNA hybridization and denaturation experiments on a DNA-modified diamond show that the diamond surfaces reveal superior stability. The influence of a different amination time on fluorescence intensity was compared. Different terminations as passivated layers were investigated, and as a result, fluorine termination points to the greatest signal-to-noise ratio.  相似文献

The infinite dilution activity coefficients of exactly athermal fluids were calculated by Monte Carlo simulation with hard-core models. The hard-core models used in this work were hard-sphere and hard-spherocylinder models. The Widom test particle method was adopted to calculate the residual chemical potentials of solutes in pure solvent and in pure solute solutions. The infinite dilution activity coefficients of solutes were obtained from the residual chemical potentials of solutes. The infinite dilution activity coefficients calculated by Monte Carlo simulation were compared with those of athermal terms in activity coefficient equations. Staverman–Guggenheim equation overestimates the activity coefficients. The deviations of activity coefficients increase with increasing the hard-core volume of solute. Flory–Huggins equation based on molar volume gives good results for the hard-spherocylinder systems. Elbro-FV equation gives good results for both the hard-sphere and hard-spherocylinder systems.  相似文献

The boron 1s photoelectron spectrum of (11)BF(3) has been measured at a photon energy of 400 eV and a resolution of about 55 meV. The pronounced vibrational structure seen in the spectrum has been analyzed to give the harmonic and anharmonic vibrational frequencies of the symmetric stretching mode, 128.1 and 0.15 meV, as well as the change in equilibrium BF bond length upon ionization, -5.83 pm. A similar change in bond length has been observed for PF(3) and SiF(4), but a much smaller change for CF(4). Theoretical calculations for BF(3) that include the effects of electron correlation give results that are in reasonable accord with the experimental values. The Lorentzian (lifetime) width of the boron 1s core hole in BF(3) is found to be 72 meV, comparable to the value of 77 meV that has been reported for CF(4).  相似文献

We synthesized Eu(III) and Sm(III) complexes with tridentate phosphine oxide ligands, Eu(hfa)(3)(TPPM) and Sm(hfa)(3)(TPPM) (hfa: hexafluoroacetylacetonato, TPPM: tris(diphenylphosphinyl)methane), and we then examined their luminescent properties. In the complexes the Eu(III) and Sm(III) centres were fully surrounded by low-vibrational frequency ligands, which led to relatively high emission quantum yields (Φ(Eu) = 30%, Φ(Sm) = 4.7%). The X-ray single crystal structures of the Eu(hfa)(3)(TPPM) revealed nona-coordinated Eu(III) complexes and C-H/O hydrogen bonding formations between the acidic hydrogen atom of the TPPM ligand and oxygen atoms of solvent molecules. The C-H/O hydrogen bonding slightly affected the coordination structure around the Eu(III) ion. Despite the seemingly small effect on the structural change, because the emission band profile of the (5)D(0)→(7)F(2) transition is sensitive to changes in the coordination environment of the Eu(III) complex, we observed a red shift in the emission spectral line.  相似文献

A new class of label-free molecular beacon (MB) system based on DNA strands that contain abasic (AP) sites (AP-DNA) and adopt stem-loop structures, in combination with fluorescent ligands that bind these AP sites, has been developed. Unlike a conventional MB, which requires covalent labeling of the MB with a fluorophore and a quencher, the developed system (APMB) does not require covalent attachment of signal transduction units. Detailed sensing functions of a series of APMB systems were examined with the aid of the fluorescent ligand named ATMND to provide insight into the design strategy for APMB systems. The effects of the stem length and the position of the AP site in the stem moiety on the fluorescence response of the APMB system were examined. Genotyping of a G/C SNP of PCR amplification products was successfully demonstrated with the APMB system and blue-fluorescent ATMND as a ligand. The APMB system was further extended to a system that utilized green-fluorescent lumiflavin.  相似文献

Here we report a novel hard-templating strategy for the synthesis of mesoporous monocrystalline Pt nanoparticles (NPs) with uniform shapes and sizes. Mesoporous Pt NPs were successfully prepared through controlled chemical reduction using ascorbic acid by employing 3D bicontinuous mesoporous silica (KIT-6) and 2D mesoporous silica (SBA-15) as a hard template. The particle size could be controlled by changing the reduction time. Interestingly, the Pt replicas prepared from KIT-6 showed polyhedral morphology. The single crystallinity of the Pt fcc structure coherently extended over the whole particle.  相似文献

A diphenylanthracene-based diamidine (1a) fluorescent probe for the detection of dicarboxylic acids has been designed and synthesized, which has an extended π-conjugation rather than a simple anthracene ring, in order to observe highly different fluorescence wavelengths after complex formation with dicarboxylic acids. The fluorescence spectra of the mixed solution of the diamidine 1a and carboxylic acids showed two different fluorescence bands, which corresponded to the complex formation (amidinium-carboxylate formation, λem?=?450?nm, light blue color) and dissociated amidinium formation (λem?=?510?nm as a broad band, green color). The complexed and dissociated states were confirmed by DOSY NMR and TD-DFT calculations. These different fluorescence wavelengths may come from the differences in the dihedral angles between the phenyl rings at the 1,8-position and anthracene ring (difference in π-conjugation) of 1a under complex formation and dissociated amidinium formation. The proposed mechanism for the observation of the different fluorescence wavelengths (complex formation and amidinium formation) was also confirmed by the fluorescence study of diamidine 1b which causes restricted rotation of the phenyl rings by substitution of the steric methyl groups, and observed the same fluorescence spectra for the complex formation and amidinium formation (400, 420, 450?nm as a vibrational structure of anthracene ring). These fluorescence characteristics of the diamidine 1a are also applicable for the detection of α,ω?dicarboxylic acids.  相似文献

Isotactic polypropylene film was stretched in poly(ethylene glycol) at 140°C and its melting behavior was investigated by using a differential scanning calorimeter (DSC-1B). The shape of the melting curve depends largely on the stretching ratio, v. A sample stretched to moderate extension (1 < v < 3.5–4) has only a single melting peak (163°C) in the thermogram. When the sample is stretched beyond v = 3.5–4, the thermogram becomes more and more complex with increase of v, and some peaks appear when stretched to 10 < v < 13. The lowest peak which is considered to be the melting peak of the intermolecular crystals produced by the unfolding of chain molecules in the lamellae develops gradually with increase of v. In the thermogram for v = 18 the lowest temperature peak is most pronounced, in contrast to the highest temperature peak which decreases markedly in intensity. The phenomenon shows that large amounts of lamellar crystals are converted to intermolecular crystals in this region. On further stretching (v > 20) a very sharp high temperature peak appears, whose half-width is about 1°C. Qualitatively similar results were obtained for the samples stretched in poly(ethylene glycol) at 150°C and in air at 140 and 150°C.  相似文献
