Time-resolved transient absorption spectroscopy with sub-9 fs ultrashort laser pulses in the deep-ultraviolet (DUV) region is reported for the first time. Single 8.7 fs DUV pulses with a spectral range of 255-290 nm are generated by a chirped-pulse four-wave mixing technique for use as pump and probe pulses. Electronic excited state and vibrational dynamics are simultaneously observed for an aqueous solution of thymine over the full spectral range using a 128-channel lock-in detector. Vibrational modes of the electronic ground state and excited states can be observed as well as the decay dynamics of the electronic excited state. Information on the initial phase of the vibrational modes is extracted from the measured difference absorbance trace, which contains oscillatory structures arising from the vibrational modes of the molecule. Along with other techniques such as time-resolved infrared spectroscopy, spectroscopy with sub-9 fs DUV pulses is expected to contribute to a detailed understanding of the photochemical dynamics of biologically significant molecules that absorb in the DUV region such as DNA and amino acids.  相似文献

Histone deacetylases (HDACs) are key enzymatic regulators of many cellular processes such as gene expression, cell cycle, and tumorigenesis. These enzymes are attractive targets for drug development. However, very few simple methods for monitoring HDAC activity have been reported. Here, we have developed a fluorogenic probe, K4(Ac)-CCB, which consists of the histone H3 peptide containing acetyl-Lys and a coumarin fluorophore with a carbonate ester. By the simple addition of the probe to a HDAC solution, enzyme activity was clearly detected through spontaneous intramolecular transesterification, which renders the probe fluorescent. In addition, K4(Ac)-CCB can be applied to the evaluation of HDAC inhibitor activity. This is the first report to demonstrate the monitoring of HDAC activity by using a one-step procedure. Thus, our novel fluorogenic probe will provide a powerful tool for epigenetic research and the discovery of HDAC-targeted drugs.  相似文献

The phase-transfer behavior of poly(acrylic acid) (PAA) particles from the hydrophobic ionic liquid N,N-diethyl-N-methyl-N-(2-methoxyethyl)ammonium bis(trifluoromethanesulfonyl)amide phase to the water phase in the particle state, which we reported previously, was examined in more detail. PAA particles were prepared in the ionic liquid 1-butyl-3-methylimidazolium bis(trifluoromethanesulfonyl)amide ([Bmim][TFSA]) and the organic solvent chloroform and were extracted. The transfer of PAA particles to water in the particle state was also observed in [Bmim][TFSA] systems. In contrast, the transfer phenomenon was not observed in the chloroform system. It was clarified that water/oil interfacial tension γ(wo) is an important parameter in the extraction of PAA in the particle state from the viewpoint of free energy. When the cationic surfactant tetradecyltrimethylammonium bromide, aqueous solution was used as the extraction medium, the PAA particles were extracted in the particle state from chloroform to water, in which γ(wo) became as low as that of the ionic liquid. This suggests that the phase-transfer phenomenon of PAA particles in the particle state was induced by the ionic liquid's unique property of low interfacial tension with water despite its high hydrophobic character.  相似文献

We have designed and synthesized a reversible near-infrared (NIR) fluorescence probe, 2-Me TeR, for reactive oxygen species (ROS), utilizing the redox properties of the tellurium (Te) atom. 2-Me TeR is oxidized to fluorescent 2-Me TeOR by various ROS, while the generated 2-Me TeOR is quickly reduced in the presence of glutathione to regenerate 2-Me TeR. This redox-induced reversible NIR-fluorescence response of 2-Me TeR allowed us to detect the endogenous production of ROS and subsequent homeostatic recovery of the intracellular reductive environment in hydrogen peroxide-stimulated HL-60 cells. This probe is expected to be useful for monitoring the dynamics of ROS production continuously in vivo.  相似文献

Reactions of 10-benzo[h]quinolyllithium with a series of organosilanes led to the formation of neutral pentacoordinate complexes. The diethynyl-substituted complex was able to be converted to di(arylethynyl)-substituted ones by the Sonogashira-Hagihara coupling reaction. Electronegative substituents shortened N-Si distances and enhanced fluorescence intensity from the complexes.  相似文献

Micron‐sized monodisperse poly(ionic liquid) (PIL) particles, poly([2‐(methacryloyloxy)ethyl]trimethylammonium bis(trifluoromethanesulfonyl)amide), were prepared by dispersion polymerization at 70 °C in methanol with poly(vinylpyrrolidone) as a stabilizer. The obtained particle size could be controlled by addition of ethanol to the methanol medium while maintaining narrow monodispersity. The PIL particles exhibit unique properties; they can be observed by scanning electron microscopy without platinum coating, which is generally used to avoid an electron charge. Moreover, the solubility of the PIL particles can be easily changed by changing the counter anion, similar to the process for ionic liquids.  相似文献

Highly proton-conductive elastic composites have been successfully prepared from H3PO4-doped silica gel and a styrene-ethylene-butylene-styrene (SEBS) block elastic copolymer. Ionic conductivities of the composites depended on the concentration of H3PO4 and the heat treatment temperature of the H3PO4-doped silica gel. It was found that H3PO4 added is present mainly as free orthophosphoric acid in the silica gel. The composite composed of H3PO4-doped silica gel with a molar ratio of H3PO4/SiO2 = 0.5 heat-treated at temperatures below 200°C and SEBS elastomer in 5 mass% showed a high conductivity of 10–5 S cm–1 at 25°C in an dry N2 atmosphere. The water adsorption during a storage in 25% relative humidity at room temperature for 1 day enhanced the ionic conductivities of composites by about one order of magnitude. Lower conductivities obtained in the composite with the H3PO4-doped silica gel heat-treated at 250°C for 1 h were due to the formation of crystalline Si3(PO4)4. The temperature dependence of conductivity of the composites was the Vogel-Tamman-Fulcher type, indicating that proton was transferred through a liquidlike phase formed in micropores of the H3PO4-doped silica gels. The temperature dependence of the modulus of the composite was similar to that of the SEBS elastomer. The thermoplastically deforming temperature of the composite was around 100°C, which was higher by 30°C than that of the SEBS elastomer.  相似文献

Silica gels doped with several protonic acids such as HClO4, H2SO4 and H3PO4 have been prepared by the sol-gel method and totally solid electric double-layer capacitors have been successfully fabricated using the highly proton-conductive silica gels as an electrolyte and activated carbon powder (ACP) hybridized with the silica gels as a polarizable electrode. It was found that the addition of HClO4, which had the highest value of acid dissociation constant among these three acids, most effectively increased the proton conductivity of the resultant acid-doped silica gels. Tablets of the HClO4-doped silica gels exhibited conductivities as high as 10–5–10–2 S cm–1 at room temperature in dry N2 atmosphere. One of the capacitors fabricated using the protonic acid-doped silica gels had a capacitance of 44 F/(gram of total ACP in the capacitor), which was comparable to those of conventional capacitors using liquid electrolytes.  相似文献
