The highly ordered Si nanotube (SiNT) arrays have been fabricated and demonstrated to be an attractive matrix to accommodate macromolecules, such as proteins, and exhibit facile direct electrochemistry of cytochrome c without any pretreatment. A pair of well-defined redox waves can be observed in the aqueous solution containing cytochrome c on this SiNT electrode using cyclic voltammetry. The peak separation (DeltaEp) between the anodic and the cathodic peaks is 63 mV at the sweep rate of 10 mV/s, and the peak current is proportional to the square root of the sweep rates, which indicates that the redox process is a quasi-reversible process. The experimental results show that the good biocompatibility and nanoscale hollow structure of the SiNT arrays can be applicable to preparing Si-based biosensors and protein characterizations. 相似文献

通过紫外-可见光谱、荧光光谱、同步荧光光谱、圆二色谱、衰减全反射红外光谱、负染-透射电镜、等温滴定微量热等实验方法系统地探讨了咪唑型离子液体与牛血清蛋白(BSA)的缔合特性.结果发现,离子液体[Bmim]Cl的加入使得BSA的紫外吸收强度增加,同时也会导致其荧光猝灭,并且这种猝灭是静态猝灭.同步荧光的研究结果表明,[Bmim]Cl分子可与蛋白质中接近色氨酸残基的区域发生相互作用,使蛋白质的构象和内部的疏水结构发生改变;负染色法透射电镜直观地显示了加入离子液体后形成的蛋白质-离子液体复合物结构逐渐变大;圆二色谱和衰减全反射红外光谱表明:在离子液体与BSA缔合过程中,离子液体的加入使得BSA二级结构中的α-螺旋和β-折叠的含量降低,从而引起蛋白质二级结构的变化;表面张力法和等温滴定微量热法进一步证实上述缔合作用为静电作用和疏水作用共同作用的结果,但离子液体的烷基链与BSA疏水内腔之间的疏水作用是离子液体与BSA缔合的主要驱动力. 相似文献

The separation and determination of four parabens (methyl, ethyl, propyl, and butyl p-hydroxybenzoate) which are commonly used as preservatives in cosmetic products, by micellar electrokinetic capillary chromatography (MEKC) with and without large-volume sample stacking (LVSS) technique were compared. As an effective on-line concentration technique, LVSS was successfully combined with MEKC to determine neutral parabens in an acidic media. The effects of some typical parameters such as sample volume, buffer pH, temperature, and concentration of surfactant were examined. The detection limits for this LVSS-MEKC method were found to be 3.0 × 10−7 M for each of the parabens based on the signal-to-noise ratio of 3, which were around 300 times lower than normal MEKC technique. The curves of peak response versus concentration were linear from 1.0 × 10−6 to 5.0 × 10−5 M with regression coefficients of 0.9987, 0.9960, 0.9925 and 0.9864, respectively. A simple and easy-manipulative sample preparation method was developed and validated by analyzing commercially available cosmetic samples. It was found that with current sample preparation process and instrumentation system, 0.5 g of sample is enough for the analysis of parabens preservatives in cosmetic product with satisfactory results. 相似文献

A new type of turn-on fluorescent probe CF-AC for the detection of Cys was firstly reported. The probe exhibited an excellent response to Cys with high selectively and sensitivity. In the presence of Cys, two fluorescence emission peaks at 525 nm and 650 nm appeared accompanied by the fluorescence color change from blue to red. 飞机燃油系统是一个由许多相互联系的子系统构成的复杂总体,因而易于发生各类故障,当故障发生时会造成严重影响,为此,设计了一种基于禁忌神经网络和DS证据的飞机燃油系统故障诊断方法;首先,建立了飞机燃油系统的故障诊断模型,然后,建立了3层的BP神经网络故障诊断模型,并采用禁忌优化算法对BP神经网络进行参数优化,得到多个并行诊断的禁忌神经网络,输入样本数据对其训练并利用BP反向传播算法再次调优;最后将测试样本数据输入各禁忌神经网络,并将诊断结果作为证据采用DS证据理论进行融合,得到最终的故障诊断结果;实验结果表明:引入DS证据理论的故障诊断方法能有效克服单一故障诊断方法无法精确诊断故障的不足,诊断精度高,具有较大的优越性。 相似文献

As an important microbial resource, Actinomycetes, especially Streptomyces, have important application values in medicine and biotechnology. Streptomyces fungicidicus SYH3 was isolated from soil samples in tomato-growing areas and showed good inhibitory effects on Alternaria solani in tomato. To obtain pure active compounds, SYH3 fermentation broth was subjected to XAD-16 macroporous resin and silica gel column chromatography. Combined with the repeated preparation and separation of preparative high-performance liquid chromatography (HPLC), a total of four monomer compounds were obtained after activity tracking. Compound 4 was identified as a new six-membered lactone ring compound named 6-(5-hydroxy-6-methylheptyl)-5,6-dihydro-2H-pyran-2-one by 1D and 2D nuclear magnetic resonance (NMR) data and mass spectrometry (MS). The other three active compounds belong to the cyclodipeptide, and their half maximal inhibitory concentration (IC50) values against A. solani were 43.4, 42.9, and 30.6 μg/mL, respectively. Compound 4 significantly inhibited the spore germination and induced swollen and deformed local hyphae of A. solani with an IC50 value of 24.9 μg/mL. Compound 4 also had broad-spectrum antifungal activity and had a good antifungal effect on the tested plant-pathogenic fungi. The modes of action of new compound (4) still require further investigation, representing a novel and effective anti-fungal agent for future application. 相似文献
