In this study the dose-depth distribution pattern of proton beams was investigated by inactivation of human cells exposed to high-LET (linear energy transfer) protons. The proton beams accelerated up to 45 MeV were horizontally extracted from the cyclotron, and were delivered to the cells acutely through a home made prototype over a range of physical depths (in the form of a variable water column). The biological systems used here were two in vitro cell lines, including human embryonic kidney cells (HEK 293), and human breast adenocarcinoma cell line (MCF-7). Cells were exposed to unmodulated proton beam radiation at a dose of 50 Gy similar to that used in therapy. Resazurin metabolism assay was investigated for measurement of cell response to irradiation as a simple and non-destructive assay. In the resazurin reduction test the non-fluorescent probe dye is reduced to pink and highly fluorescent resorufin. The dose-depth distribution of proton beam obtained based on the highly sensitive laser-induced fluorometric determination of resorufin was found to coincide well with the data collected using conventional film based dosimetry. The resazurin method yielded data comparable with the optical micrographs of the irradiated cells, showing the least cell survival at the measured Bragg-peak position of 10 mm. In addition, fused silica capillary was used as a sample container to increase the probability for irradiated laser beam to probe and excite resorufin in small sample volume of the capillary. The developed method has the potential to serve as a non-destructive, sample-thrifty, and time saving tool to realize more realistic, practical dose-depth distribution of proton beam compared to conventional in vitro cell viability assessment techniques. 相似文献

We here report a new type of stationary phase for microcolumns. C18 modified silica monolith particles were prepared by grinding and sieving the silica monolith followed by C18 modification and end-capping, and were used as packing material. Ground silica monolith particles were not spherical but irregular with some residual monolithic network structure. The separation efficiency of the stationary phase made of sieved monolith particles (5-10 microm) was better than that of the stationary phase made of unsieved particles. The microcolumn packed with the sieved C18 ground monolith particles (5-10 microm) showed quite good separation efficiency (height equivalent to theoretical plate, HETP, as low as 15 microm) and it was even superior to the microcolumn packed with a commercial spherical 5 microm C18 stationary phase. The column pressure drop of C18 monolith particles was about two-third of that of the commercial spherical C18 phase. The preparation method of C18 stationary phase with ground and sieved silica monolith particles presumably suggests advantages of simplicity and convenience in modification and washing procedures compared to bulk silica monolith. The new ion-selective electrodes (ISEs) based on salphenH2 derivatives such as N,N′-(propylenedioxy)benzenebis(salicylideneimine) L1 and N,N′-4,5-(propylenedioxy)benzenebis(3,5-di-tert-butylsalicylideneimine) L2 as cation carriers are developed for a uranyl ion. The combination of these new ionophores with tris(2-ethylhexyl)phosphate (TEHP) as a plasticizer particularly shows near Nernstian slope in the wide concentration range (1.0 × 10−6 to 1.0 × 10−2 M) of UO22+ and is observed well in the pH range from 1.0 to 5.0 with a response time less than 20 s. Since the employed ionophores were confirmed to form well-defined stable 1:1 complexes with UO22+, the observed high selectivity for a uranyl ion over the other cations was attributed to the selective complexation as well as the lipophilic behavior of these ligands especially for L2. The proposed electrodes offered practically low detection limit of 6.5 × 10−7 M and reasonably good end-points within experimental error were obtained when the sensor was used as an indicator electrode for the potentiometric titration. 相似文献

We designed an A-selective fluorescent DNA probe, UFL, which bears a 2-ethynylfluorene moiety covalently attached to the base dU, and incorporated it into a central position in the hairpin loop. We describe photophysical studies of triad base pairs containing an UFL DNA probe and their application to single-nucleotide polymorphism (SNP) typing using novel quencher-free molecular beacons (MBs). A novel fluorescent receptor bearing benzimidazole moieties as recognition sites was synthesized. The recognition behaviour of the receptor towards various anions has been evaluated in CH3CN.
