We analyzed ABC transporter solute-binding proteins (SBPs) of the Bacillus subtilis membrane using a proteomic approach. We prepared a washed cell membrane fraction that was insoluble in 134 mM nondetergent sulfobetaine and then extracted proteins using mixtures of detergents in a stepwise manner. The membrane proteins were resolved by three two-dimensional polyacrylamide gel electrophoresis (2-D PAGE) or two one-dimensional (1-D) PAGE procedures, electroblotted, and digested in the presence of 5% or 80% acetonitrile. Thereafter, matrix-assisted laser desorption/ionization-time of flight-mass spectrometry (MALDI-TOF MS) identified 637 proteins corresponding to 15.9% of the total cellular proteins. We predicted that among these, 256 were membrane proteins, 101 were lipoproteins or secretory proteins and 280 were soluble proteins containing peripheral proteins that function in both the cytoplasm and the cell membrane such as SecA and FtsY. Among the 637 proteins, we identified 30 SBPs among 38 importers predicted by a bioinformatic search of the genome. We confirmed expression of the genes for the 30 SBPs using DNA microarray analysis. We compared the 2-D gel separation profiles of submembrane fractions solubilized by 1% n-dodecyl-beta-D-maltoside from cells cultured on Luria Bertani (LB), S7, and S7 medium without glutamate as well as DNA microarray data on LB and S7. The results suggested that YcdH, YtmK and YurO are binding proteins for Mn(++), glutamate and glucose, respectively, and that YqiX and YxeM are binding proteins for amino acids (tryptophan in S7 medium).  相似文献

Ternary sodium borosilicide, Na(8)B(74.5)Si(17.5), was newly synthesized by heating a mixture of sodium, silicon and amorphous or crystalline boron at 1073-1273 K. The crystal structure of the black hexagonal prismatic single crystal obtained at 1273 K was analyzed. The X-ray diffraction reflections of the crystal were indexed with hexagonal cell parameters a = 10.2392(3) ? and c = 10.9215(4) ? (space group P6(3)/mmc, No. 194). The structural formula could be represented as Na(8)(B(12))(6)Si(16)[BSi](1.5)[B(2)](0.5). B(12) icosahedra form a three-dimensional framework having small triangular and large hexagonal channels along the c axis. Chains of [-Si-(Si-Si)(3)-Si-] surrounded by Na atoms are located in the large channels. Trigonal prism cages for Na atoms and for atom pairs of Si-B or B-B are alternately arrayed in the small channels. The ratio of Si-B and B-B pairs in the cage is around 3:1.  相似文献

We have made high density amorphous ice (HDA) by the pressure-induced amorphization of hexagonal ice at 77 K and measured the volume change on isobaric heating in a pressure range between 0.1 and 1.5 GPa. The volume of HDA on heating below ~0.35?GPa increases, while the volume of HDA on heating above ~0.35?GPa decreases. The polarized OH-stretching Raman spectra of the relaxed HDAs are compared with that of the unannealed HDA. The relaxed HDAs are prepared at 0.2 GPa at 130 K and 1.5 GPa at 160 K. It is found that the relatively strong totally symmetric OH-stretching vibration mode around 3100?cm(-1) exists in the depolarized reduced Raman spectrum χ(VH)(") of the unannealed HDA and that its intensity rapidly decreases by relaxation. The χ(VH)(") profiles of the relaxed HDA are similar to those of liquid water. These results indicate that the HDA reaches a nearly equilibrium state by annealing and the intrinsic state of HDA relates to a liquid state. The pressure-volume curve of the relaxed HDA at 140 K seems to be smooth in the pressure range below 1.5 GPa.  相似文献

A diversity‐oriented synthetic strategy allowed us to design a series of conjugated molecules containing multiple benzosilole units that can be utilized as efficient hole‐blocking materials for phosphorescent organic light emitting diodes (OLEDs). Some of these compounds showed a performance surpassing that of the current standard, bathocuproine. The new compounds were easily synthesized in a modular fashion from a previously reported 3‐stannyl benzosilole building unit. Studies on the properties of these compounds in solution and in the solid state indicate that they possess high electron affinity, high ionization potential, and form stable amorphous films that show high electron‐drift mobility. The correlation between their molecular properties and the efficiency of the OLED device performance is also investigated.  相似文献

X-ray crystallographic and cross-polarization/magic angle spinning 13C nuclear magnetic resonance techniques have been used to study an ethylenediamine (EDA)-cellulose I complex, a transient structure in the cellulose I to cellulose IIII conversion. The crystal structure (space group P2 1 ; a = 4.546 Å, b = 11.330 Å, c = 10.368 Å and γ = 94.017°) corresponds to a one-chain unit cell with one glucosyl residue in the asymmetric unit, a gt conformation for the hydroxymethyl group, and one EDA molecule per glucosyl residue. Unusually, there are no O–H···O hydrogen bonds between the cellulose chains; the chains are arranged in hydrophobic stacks, stabilized by hydrogen bonds to the amine groups of bridging EDA molecules. This new structure is an example of a complex in which the cellulose chains are isolated from each other, and provides a number of insights into the structural pathway followed during the conversion of cellulose I to cellulose IIII through EDA treatment.  相似文献

All four stereoisomers of p-tert-butylsulfinylcalix[4]arene [4(rccc), 4(rcct), 4(rctt), and 4(rtct)], arising from the disposition of the four sulfinyl groups with respect to the mean plane of the four bridging sulfur atoms, have been prepared via oxidation of p-tert-butylthiacalix[4]arene (2) or its tetra-O-benzyl ethers 5 of defined conformations. Thus, treatment of 2 with 4.4 molar equiv of NaBO(3).4H(2)O gave the rtct and rctt isomers in 27% and 17% yields, respectively, while oxidation of cone 5 (5(C)) and partial cone 5 (5(PC)) proceeded stereoselectively to give, after debenzylation of the resulting tetrasulfoxides 12 and 15, the rccc and rcct isomers in 56% and 28% yields, respectively, based on 5. The sulfinylcalix[4]arenes 4 were treated with iodomethane in the presence of a base to give the corresponding tetramethyl ethers 16, the structures of which in regard to the disposition of the sulfinyl groups and the conformation of the phenol units were determined by X-ray crystallography. Also reported is the synthesis of all four conformational isomers of tetra-O-benzyl ether of 2 (5(C), 5(PC), 5(1,2-A), and 5(1,3-A)).  相似文献

The magnetic property of Co2B2O5 and the optical property of M2B2O5 (M = Mn, Co) were investigated. Co2B2O5 showed antiferromagnetic behavior below the Néel temperature of TN ≈ 45 K, and the Weiss temperature was TW = +7.7 K. The effective magnetic moment of Co was 4.96 μB, which indicated that Co was divalent and in a high-spin state. Absorptions attributed to the d–d transitions in Mn2+ and Co2+ ions were observed at 250–650 nm in the diffuse reflection spectra. The optical absorption edges of Mn2B2O5 and Co2B2O5 were at 243 nm (5.11 eV) and 299 nm (4.15 eV), respectively.  相似文献

Di‐stereoblock polylactides (di‐sb‐PLA: PLLA‐b‐PDLA) having high molecular weight (Mn > 100 kDa) were successfully synthesized by two‐step ring‐opening polymerization (ROP) of L ‐ and D ‐lactides using tin(2‐ethylhexanoate) as a catalyst. By optimizing the polymerization conditions, the block sequences were well regulated at non‐equivalent feed ratios of PLLA and PDLA. This synthetic method consisted of three stages: (1) polymerization of either L ‐ or D ‐lactide to obtain a PLLA or PDLA prepolymer with a molecular weight less than 50 kDa, (2) purification of the obtained prepolymer to remove residual lactide, and (3) polymerization of the enantiomeric lactide in the presence of the purified prepolymer. Their 13C and 31P NMR spectra of the resultant di‐sb‐PLAs strongly supported their di‐stereo block structure. These di‐sb‐PLAs, having weight‐average molecular weights higher than 150 kDa, were fabricated into polymer films by solution casting and showed exclusive stereocomplexation. The thermomechanical analysis of the films revealed that their heat deformation temperature was limited probably because of their low crystallinity owing to the non‐equivalent PLLA/PDLA ratio. The blend systems of the di‐sb‐PLAs having complementary stereo‐sequences (the one with a long PLLA block and the other with long PDLA block) were also prepared and characterized to enhance the sc crystallinity. © 2010 Wiley Periodicals, Inc.
