The steady-state and time-resolved fluorescece spectroscopy is one of the most powerful method to detect and analyze subtle
conformation change and interaction between peptide elements in protein. Phytocystatin Scb isolated from sunflower seeds includes
a single Trp residue at position 85. In an attempt to investigate the interaction of the N-terminal region of Scb with the
first and second hairpin loops by fluorescence spectroscopy of Trp residue, two Scb mutants in which single Trp locates at
position 52 and 58, respectively, and their N-terminal removed mutants were generated. The N-terminal truncation changed the
fluorescence decay kinetics of Trp52 from the triple exponential to double. Furthermore, the time-resolved fluorescence anisotropy
residue indicated that the segmental motion of Trp52 was significantly enhanced by its N-terminal truncation. In contrast,
Trp58 and Trp85 had little influence. The N-terminal successive truncations of Scb and its mutants resulted in the weaken
inhibitors to papain. These results suggested that the N-terminal region of Scb interacts with the peptide segment preceding
the first hairpin loop, thereby stabilizing the conformation of the hairpin loop structure. 相似文献

An optical mono-stable multivibrator laser diode (MM-LD) is realized by using a multi-electrode distributed feedback laser diode. All-optical pulse-width conversion of ultra-short pulses to non-return-to-zero (NRZ) is achieved using an MM-LD. The MM-LD is adopted for a wide range of bit-rates between 2-10 Gbit/s by tuning the DC bias. Data format transformation from 10-Gbit/s return-to-zero optical signals to NRZ optical-signals is achieved with error free operation. Converted optical signals, which have a narrower spectral bandwidth and lower peak power than when input, are transmitted using a 1.3-μm zero dispersion fiber (1.3Aλ0-SMF). 相似文献

A micellar solution of cetylpyridinium chloride (CPC) can separate into two phases due to a temperature change or to the addition of salts. Platinum(II), (IV) and palladium(II) reacted with chloride ions to form stable anionic complexes of PtCl4(2-), PtCl6(2-) and PdCl4(2-), respectively, and were adsorbed onto the CPC gel phase. The CPC phase plays the role of an ion-exchange adsorbent for the anionic complexes. By such a procedure, the precious metals of platinum and palladium could be separated from base metals such as copper, zinc and iron. The kinetic separation was performed by a ligand exchange reaction of the palladium(II) chloro-complex with EDTA at 60 degrees C. The anionic palladium(II)-EDTA complex could not bind the opposite charged CP+ and was desorbed from the CPC phase. In the aqueous phase, the recovery of palladium(II) by the double-desorption was 101.1 +/- 1.2%. The platinum(II) and (IV) chloro-complexes were stable for at least 30 min and remained in the CPC phase. 相似文献

Fluorous chiral BINOLs and BINAP were prepared and used as the ligands for an asymmetric addition of Et2Zn to aromatic aldehydes and an asymmetric Heck reaction, respectively. The enantioselectivities were similar in homogeneous system to those of the original non-fluorous reactions. Consecutive reactions were examined by utilizing fluorous–organic biphase and fluorous solid phase extraction techniques. Enantioselectivities in consecutive reactions were close to that attained in the non-fluorous system. Intense photostimulated luminescence (PSL) with a peak at 420 nm is observed in ultraviolet (UV) Iight-irradiated europium-doped potassium chloride (KCl: Eu) crystalline phosphors. The PSL characteristics of UV-irradiated KCl: Eu phosphor for optical memory application were studied. Vacancy-mediated lateral manipulations of intrinsic adatoms of the Si(111)-(7x7) surface at room temperature are reported. The topographic signal during the manipulation combined with force spectroscopy measurements reveals that these manipulations can be ascribed to the so-called pulling mode, and that the Si adatoms were manipulated in the attractive tip-surface interaction regime at the relatively low short-range force value associated to the manipulation set point. First-principles calculations reveal that the presence of the tip induces structural relaxations that weaken the adatom surface bonds and manifests in a considerable local reduction of the natural diffusion barriers to adjacent adsorption positions. Close to the short-range forces measured in the experiments, these barriers are lowered near the limit that enables a thermally activated hopping at room temperature. 相似文献

Background
Peptides with cytoprotective functions, including antioxidants and anti-infectives, could be useful therapeutics. Carnosine, β-alanine-histidine, is a dipeptide with anti-oxidant properties. Tripeptides of Ala-His-Lys, Pro-His-His, or Tyr-His-Tyr are also of interest in this respect.Results
We synthesized several histidine-containing peptides including glycine or alanine, and tested their cytoprotective effects on hydrogen peroxide toxicity for PC12 cells. Of all these peptides (Gly-His-His, Ala-His-His, Ala-His-Ala, Ala-Ala-His, Ala-Gly-His, Gly-Ala-His (GAH), Ala-His-Gly, His-Ala-Gly, His-His-His, Gly-His-Ala, and Gly-Gly-His), GAH was found to have the strongest cytoprotective activity. GAH decreased lactate dehydrogenase (LDH) leakage, apoptosis, morphological changes, and nuclear membrane permeability changes against hydrogen peroxide toxicity in PC12 cells. The cytoprotective activity of GAH was superior to that of carnosine against hydrogen peroxide toxicity in PC12 cells. GAH also protected PC12 cells against damage caused by actinomycin D and staurosporine. Additionally, it was found that GAH also protected SH-SY5Y and Jurkat cells from damage caused by hydrogen peroxide, as assessed by LDH leakage.Conclusion
Thus, a novel tripeptide, GAH, has been identified as having broad cytoprotective effects against hydrogen peroxide-induced cell damage.108.
To develop the redox-switching system of intramolecular magnetic interaction, 1,2-bis[3-(N-tert-butyl-N-oxylamino)phenyl]-1,2-bis[4-{N,N-bis(4-methoxyphenyl)amino}phenyl]ethylene, tetraarylethylene with two nitroxide radical groups at the meta-position, was synthesized, and characterized by the electrochemical method and ESR spectroscopy. Cyclic voltammetry showed the tetraarylethylene core has the lower oxidation potential than the substituted nitroxide radical moiety. ESR spectroscopy in frozen solution revealed that the neutral form shows the fine-structured spectrum characteristic of the spin triplet species, while the dicationic form shows the anisotropic hyperfine-structured spectrum characteristic of the randomly-oriented nitroxide radical, indicating the drastic change of intramolecular magnetic interaction. 相似文献

Much attention has been paid to the cross-coupling reaction of organosilicon compounds due to their stability, non-toxicity, and natural abundance of silicon. In addition, the silicon-based cross-coupling has many advantages over other cross-coupling protocols. Successful examples of the silicon-based cross-coupling reaction are reviewed, focusing especially on the advances made in the last decade. Having had a number of highly effective palladium catalysts developed mainly for other cross-coupling reactions, the development of the silicon-based protocol owes heavily to the design of organosilicon reagents which effectively undergo transmetalation, a key elemental step of the silicon-based cross-coupling reaction. This tutorial review thus classifies various organosilicon reagents depending on substituents on silicon and surveys their cross-coupling reactions with various electrophiles. 相似文献
