Methylglyoxal-induced oxidative stress and cytotoxicity are the main factors causing neuronal death-related, diabetically induced memory impairment. Antioxidant and anti-apoptotic therapy are potential intervention strategies. In this study, 25 flavonoids with different substructures were assayed for protecting PC-12 cells from methylglyoxal-induced damage. A structure–activity relationship (SAR) analysis indicated that the absence of the double bond at C-2 and C-3, substitutions of the gallate group at the 3 position, the pyrogallol group at the B-ring, and the R configuration of the 3 position enhanced the protection of flavan-3-ols, and a hydroxyl substitution at the 4′ and meta-positions were important for the protection of flavonol. These SARs were further confirmed by molecular docking using the active site of the Keap1–Nrf2 complex as the receptor. The mechanistic study demonstrated that EGCG with the lowest EC50 protected the PC-12 cells from methylglyoxal-induced damage by reducing oxidative stress via the Nrf2/Keap1/HO-1 and Bcl-2/Bax signaling pathways. These results suggested that flavan-3-ols might be a potential dietary supplement for protection against diabetic encephalopathy. 相似文献

Summary: Like in many other cases, block copolymers bearing alkoxysilyl groups in one segment self‐assemble into aggregates. However, they may allow one to study the sol‐gel reaction by making use of the gelable groups located in the domains of the aggregates, therefore, the gelation process can be made in a selected domain of the nanoscale. As a result, the organic nanostructure is transformed into an organic/inorganic hybrid. The process of using covalently grafted alkoxysilane groups along the block copolymer precursors is applicable not only to the aggregates formed in solution, but also to the other forms of aggregates in melts, thin films, and interfaces. In this feature article, this emerging field is introduced, mainly focusing on the gelation that occurs inside the preformed block copolymer aggregates in solution. The morphologically fixed hybrid nanoparticles, such as spheres, hollow particles, and complex hollow particles, are presented and discussed. This technology has great potentials in the fields of nanomaterials and nanotechnologies, since various organic/inorganic hybrid nanoparticles and nanostructures can be effectively generated using this process.

Self‐healing cyanate ester resins (CE) were developed by adding low molecular weight poly(phenylene oxide) (PPO) resin, yielding a high performance CE/PPO system via a low‐temperature process. The addition of PPO improved the flexural strength and fracture toughness of the CE matrix without sacrificing thermal properties. CE/PPO formulations with 5, 10, and 15 wt.% PPO showed 43%, 65%, and 105% increase in fracture toughness due to a combination of crack deflection, crack pinning, and matrix cavitation around second‐phase particles. When PPO was introduced into the CE, dielectric properties were either unchanged or declined. During thermal treatment to heal damaged CE, liquid PPO flowed into cracks, and during subsequent cooling, solidified to bond the crack surfaces. The self‐healing efficiency for CE with 15 wt.% PPO after heating to 220°C for 1 h exhibited a recovery of 73% in toughness and 81% in microtensile strength. Novel amphiphilic comb‐dendronized diblock copolymers composed of hydrophobic Percec‐type dendronized polystyrene block and hydrophilic comb‐like poly(ethylene oxide) grafted polymethacrylate P(PEOMA) block were designed and synthesized via two steps of atom transfer radical polymerization (ATRP). The comb‐like P(PEOMA) prepared by ATRP of macromonomers (PEOMA) with two different molecular weights (Mn = 300 and 475) were used to initiate the sequent ATRP of dendritic styrene macromonomer (DS). The molecular weights and compositions of the obtained block copolymers were determined by 1H NMR analysis. The copolymers with relatively narrow polydispersities (1.27–1.38) were thus obtained. The bulk properties of comb‐dendronized block copolymers were studied by using differential scanning calorimetry, polarized optical microscopy and wide‐angle X‐ray diffraction (WAXD). Similar to dendronized homopolymers, the block copolymers exhibited hexagonal columnar liquid‐crystalline phase structure. By using such amphiphilic comb‐dendronized block copolymers as building blocks, the rich self‐assembly morphologies, such as twisted string, vesicle, and large compound micelle (LCM), were obtained in a mixture of CH3OH and THF. © 2008 Wiley Periodicals, Inc. A new screening and analysis method that combines in vitro metabolism with high-performance liquid chromatography-mass spectrometry
(HPLC-MS) was developed for the screening and analysis of an antineoplastic compound, coniferyl ferulate, which is present
in the rhizome of Rhizoma Chuanxiong. Infrared (IR), ultraviolet visible spectroscopy (UV-Vis), nuclear magnetic resonance (NMR) and element analysis were used
to identify the molecular structure of coniferyl ferulate. The quantitative analysis of coniferyl ferulate in different extracts
of Rhizoma Chuanxiong was carried out, and the metabolism of coniferyl ferulate was investigated by in vitro incubation with rat liver homogenate.
The metabolite of coniferyl ferulate, ferulic acid ethyl ester, was identified by HPLC-MS, UV-Vis and IR. In addition, antineoplastic
activities of coniferyl ferulate and ferulic acid ethyl ester were detected by the MTT assay. The observed inhibition rate
of coniferyl ferulate on the activity of HeLa cells was over 80% at 5.4 ng μl−1. However, its metabolite, ferulic acid ethyl ester, showed no antineoplastic activity in vitro.

Thermal rearrangement reactions of (aminomethyl)silane H(3)SiCH(2)NH(2) were studied by ab initio calculations at the G3 level. The results show that two dyotropic reactions could happen when H(3)SiCH(2)NH(2) is heated. In one reaction, the silyl group migrates from the carbon to the nitrogen atom while a hydrogen atom shifts from the nitrogen to the carbon atom, forming (methylamino)silane CH(3)NHSiH(3) (reaction A). This reaction can proceed via three paths: a path involving two consecutive steps with two transition states and one intermediate metastable carbene species (A-1); and two concerted paths (A-2 and A-3). In the other reaction, the amino group migrates from the carbon to the silicon atom while a hydrogen atom shifts from the silicon to the carbon atom, via a double three-membered ring transition state, forming aminomethylsilane CH(3)SiH(2)NH(2) (reaction B). Reaction rate constants, changes (DeltaS(#), DeltaH, and DeltaG) in thermodynamic functions and equilibrium constants of the reactions were calculated with the MP2(full)/6-311G(d,p) optimized geometries, harmonic vibrational frequencies and G3 energies of reactants, transition states, intermediates and products with statistical mechanical methods and the conventional transition-state theory (TST) with Wigner tunneling approximation over a temperature range 400-1800 K. 相似文献

In this paper, an adaptive fuzzy output feedback approach is proposed for a single-link robotic manipulator coupled to a brushed direct current (DC) motor with a nonrigid joint. The controller is designed to compensate for the nonlinear dynamics associated with the mechanical subsystem and the electrical subsystems while only requiring the measurements of link position. Using fuzzy logic systems to approximate the unknown nonlinearities, an adaptive fuzzy filter observer is designed to estimate the immeasurable states. By combining the adaptive backstepping and dynamic surface control (DSC) techniques, an adaptive fuzzy output feedback control approach is developed. Stability proof of the overall closed-loop system is given via the Lyapunov direct method. Three key advantages of our scheme are as follows: (i) the proposed adaptive fuzzy control approach does not require that all the states of the system be measured directly, (ii) the proposed control approach can solve the control problem of robotic manipulators with unknown nonlinear uncertainties, and (iii) the problem of “explosion of complexity” existing in the conventional backstepping control methods is avoided.
