Grand canonical ensemble Monte Carlo simulation (GCMC) combined with the histogram reweighting technique was used to study the thermodynamic equilibrium of a homopolymer solution between a bulk and a slit pore. GCMC gives the partition coefficients that agree with those from canonical ensemble Monte Carlo simulations in a twin box, and it also gives results that are not accessible through the regular canonical ensemble simulation such as the osmotic pressure of the solution. In a bulk polymer solution, the calculated osmotic pressure agrees very well with the scaling theory predictions both for the athermal polymer solution and the theta solution. However, one cannot obtain the osmotic pressure of the confined solution in the same way since the osmotic pressure of the confined solution is anisotropic. The chemical potentials in GCMC simulations were found to differ by a translational term from the chemical potentials obtained from canonical ensemble Monte Carlo simulations with the chain insertion method. This confirms the equilibrium condition of a polymer solution partition between the bulk and a slit pore: the chemical potentials of the polymer chain including the translational term are equal at equilibrium. The histogram reweighting method enables us to obtain the partition coefficients in the whole range of concentrations based on a limited set of simulations. Those predicted bulk-pore partition coefficient data enable us to perform further theoretical analysis. Scaling predictions of the partition coefficient at different regimes were given and were confirmed by the simulation data.  相似文献

DNA methyltransferase 1(DNMT1) is a useful biomarker for lung cancer in early clinical diagnosis. A rapid magnetic chemiluminescence immunoassay(MCLIA) for DNMT1 in human serum has been developed.Horseradish peroxidase(HRP)-second-Ab was used to labeled polyclonal antibodies of anti-DNMT1. DNMT1 in sample integrates with specific immunomagnetic beads and can constitute a supersandwiched immunoreaction. In magnetic field, nonspecific materials can be separated. After luminescent substrate luminol-H_2O_2-BIP was added, the relative light unit(RLU) of HRP was detected and was discovered to be directly proportional to the content of DNMT1 in sample. The correlative variables involved in the MCLIA value were optimized and the methodological evaluation was carried out. After optimization, in the range of0.5–128 ng/mL, the linear regression equation was y = 0.5014 x + 1.769(x was logCDNMT1, y was relative luminescence units(RLU)/RLU0), and the limit of detection was 0.01 ng/mL. The RSD of intra-and interassays were 15.8%–16.9% and 14.3%–18.1%, respectively. The recovery was from 70.0% to 106.2%.Furthermore, paralleled with purchasable enzyme-linked immunosorbent assay(ELISA) kits, MCLEIA had lower detection limit, wider linear range and shorter detection time. Therefore, the MCLEIA established in this study could be used for the sensitive detection of DNMT1 in serum sample.  相似文献

(S)-Selective kinetic resolution was achieved through the use of a commercially available protease, which was activated with a combination of two different surfactants. The kinetic resolution (KR) process was optimized with respect to activation of the protease and to the acyl donor. The KR proved to be compatible with a range of functionalized sec-alcohols, giving good to high enantiomeric ratio values (up to >200). The enzymatic resolution was combined with a ruthenium-catalyzed racemization to give an (S)-selective dynamic kinetic resolution (DKR) of sec-alcohols. The DKR process works under very mild reaction conditions to give the corresponding esters in high yields and with excellent enantioselectivities.  相似文献

A general enantioselective synthesis of functionalized nitrocyclopropanes by organocatalytic conjugate addition of a variety of bromonitroalkanes to α,β‐unsaturated enone systems is presented. The process, efficiently catalyzed by the salts of 9‐amino‐9‐deoxyepiquinine 1 d serves as a powerful approach to the preparation of synthetically and biologically important cyclopropanes with high levels of enantio‐ and diastereoselectivities. Since only 0.6 equivalents of bromonitromethane are used as a reagent, (S)‐ 2 e is obtained enantiomerically pure by employing chiral 1 d as a highly efficient catalyst for its kinetic resolution (97 % ee at 51 % conversion, selectivity s=120).  相似文献

Multidrug resistance(MDR) is a main factor to make the failure of chemotherapy. It is closely related to the over-expression of P-glycoprotein(P-gp), multidrug resistance protein(MRP) and breast cancer resistance protein(BCRP). Herein we reported a novel method to characterize MDR, taking advantage of the electrochemical properry of chemotherapeutic drugs. Meanwhile, the definition of accumulation phase and retention phase has been improved. Furthermore, with specific modulators introduced to inhibit the relevant efflux pumps, the exact protein that mainly works in the cells employed in this study can be identified.  相似文献

脂肪酸囊泡(FAV)是一类重要的纳米容器,然而其形成pH范围较窄且偏碱性环境,限制了其应用。 本文将共轭亚油酸(CLA)与海藻酸钠(SA)在近中性环境下共同自组装囊泡化纳米容器并提高其膜稳定性。动态激光光散射(DLS)和透射电子显微镜(TEM)结果表明,当SA质量分数为25%~50%时复合体系可在近中性条件下自组装形成50~250 nm尺寸的囊泡化纳米容器,且pH=7.4时随着质量分数增加囊泡化纳米容器直径增大。 根据SA和CLA在中性环境的物种存在形式推测,二者通过氢键作用驱动形成囊泡化纳米容器。 体外模拟释放实验表明,囊泡化纳米容器具有较高包覆率和较优缓释效果,有望应用于药物传输领域。  相似文献
