Sensitive determination of bisphenol A and alkylphenols by high performance liquid chromatography with pre-column derivatization with 2-(4-carboxyphenyl)-5,6-dimethylbenzimidazole

A new and sensitive high-performance chromatographic method for the determination of bisphenol A and 8 alkylphenols with fluorescence detection is reported. Each phenol was derivatized by reaction with 2-(4-carboxyphenyl)-5,6-dimethylbenzimidazole at 40 degrees C for 60 min. The fluorescence derivatives were separated on a Wakosil 5C18 column (4.0 i.d. x 300 mm, 5 microm) with methanol:water (10:90) as mobile phase (detection wavelength: lambda(ex) 336 nm, lambda(em) 440 nm). The detection limits were in the range of 0.1-10.0 pg/mL in serum. The calibration graphs were linear to 1.0 microg/mL. The relative standard deviations were 7.2-8.9%, respectively. The proposed method was applied to the determination of bisphenol A in mother and infant rat serum.     

Search for a simpler synthetic model system for intramolecular 1,3-dipolar cycloaddition to the 5,6-double bond of a pyrimidine nucleoside

In search for a simpler model system for the study of intramolecular thermal reactions between the base and 5'-functionalized sugar moiety in nucleosides, 1-(3-azidopropyl)uracil (2), 1-(4-azidobutyl) pyrimidines (12 and 13) and 1-(5-azidopentyl)-uracil (14) was synthesized through the corresponding ω-benzoyloxy-(6,7 and 8) and ω-hydroxyalkyl-pyrimidines (9,10 and 11). Heating 2 gave 1,N⁶-trimethylene-6-aminouracil (4), while heating 12 and 13 gave N¹-C₆ cleaved addition products. 15 and 16, respectively. 15 was regiospecifically transformed to 1,2,3-triazole derivatives, 17,18 and 19. Heating 1-(4-azidobutyl)-5-bromouracil (20) yielded 3,9-tetramethylene-8-azaxanthine (22). 9 with NBA gave 1,0⁶-tetramethylene-5-bromo-6-hydroxy-5,6-dihydrouracil (24) and the 5-brominated analog of 9 (25). The 4-functionalized butyl side chain proved to serve as a substitute for the 5'-functionalized sugar moiety in pyrimidine ribonucleosides.     

Capillary electrophoretic determination of Ga(III) based on the formation of a heteropolyoxomolybdate complex

A novel capillary electrophoretic (CE) method was developed for the determination of Ga(III). The so-called Anderson-type [GaMo6O24H6]3- complex was readily formed by the reaction of Mo(VI) with Ga(III) in 0.050 M monochloroacetate buffer (pH 2.0) and the precolumn complex-formation reaction was applied to the CE determination of Ga(III) with direct UV detection at 240 nm. The peak area was linearly dependent on the concentration of Ga(III) in the range of 5.0 x 10(-7)-5.0 x 10(-5) M. Owing to the high molar absorptivity of the Anderson anion, a detection limit of 2.0 x 10(-7) M (signal-to-noise ratio=3) was achieved. The advantage of the present method is that the presence of large excesses of Al(III) and In(III) does not cause interference.     

Facile route to 3,5-disubstituted morpholines: enantioselective synthesis of O-protected trans-3,5-bis(hydroxymethyl)morpholines

[reaction: see text] (3R,5R)-1 R1 & R2 = TBDPS, (3S,5R)-2 R1 = Bn,R2 = TBDPS, (3S,5S)-3 R2 & R2 = Bn. trans-3,5-Bis(benzyl/tert-butyldiphenylsilyloxymethyl)morpholines, promising candidates for the C(2)-symmetric class of chiral reagents, were prepared with excellent optical purity. A key step in the synthesis is the coupling of a serinol derivative with 2,3-O-isopropylideneglycerol triflate or its equivalent. This methodology was extended to the synthesis of chiral trans-3-(benzyloxymethyl)-5-(tert-butyldiphenylsilyloxymethyl)morpholine, a potentially useful chiral building block.     

Fundamental equilibrium analysis on the ion-pair extraction of dibenzo-18-crown-6 complex of alkali metal ions with picrate ion into 1,2-dichloroethane

The ion-pair extraction equilibria of dibenzo-18-crown-6 (DB18C6) complexes of Na⁺, K⁺, Rb⁺ and Cs⁺ (M⁺) with picrate ion (Pic⁻) into 1,2-dichloroethane (1,2-DCE) have been studied at 25.0°C. In the case of the Rb⁺ and Cs⁺ systems, the extraction results were interpreted by taking into consideration the formation of a (DB18C6)₂ -M⁺ complex in 1,2-DCE. The thermodynamic constants of extraction, , and ion-pair formation in 1,2-DCE, , of ion pairs of the DB18C6-M⁺ complexes with Pic⁻ were determined. By using the distribution coefficient of M⁺·Pic⁻ the thermodynamic formation constants of the DB18C6-M⁺ complexes in 1,2-DCE, , were evaluated. Consequently the component equilibrium constants of the ion-pair extraction were completely determined and a contribution of these constants to the difference of value was discussed. The value in 1,2-DCE is quite high compared with that in solvating solvents and log decreases linearly with increasing Gutmann donor number of solvents.     

Friction coefficients of spheres having sticky or hairy surfaces

The theoretical equations for friction coefficients of spheres having sticky or hairy surfaces are presented. In the present treatment, the sticky surface is represented by a higher viscosity of fluid in the surface layer and the hairy surface is assumed to have a porous structure. The friction coefficients are given as functions of the thickness of the surface layer, the fluid viscosity in the surface layer and the segmental density of hairy chains.     

Properties of χ-alumina as related to catalysis

-Alumina was synthesized by thermal decomposition of aluminium(III) isopropoxide, and its properties were investigated. The change in its surface area with increasing calcination temperature resembled that of -alumina. Both aluminas had the same acid strength, and their activities in isomerization of 1-butene were the same. -Alumina as a support for molybdenum catalyst in the epoxidation of allyl alcohol also behavedin the same manner as -alumina. Thus -alumina has almost the same catalytic properties as -alumina.     

Determination of a new cerebral vasodilator 2,6-dimethyl-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methylamino)]-ethyl ester 5-methyl ester hydrochloride (YC-93) in plasma by electron capture gas chromatography

A highly sensitive method for the quantitative determination of 2,6-dimethy-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylic acid 3-[2-(N-benzyl-N-methyl-amino)]-ethyl ester 5-methyl ester hydrochloride (YC-93) in plasma is described. After extraction, YC-93 was oxidized to pyridine analogue with nitrous acid and detected by electron capture gas chromatography. The sensitivity was 2–3 ng/ml, which is suficient to determine plasma concentrations of YC-93 after oral administration of clinical doses to humans.     

Polycyclic N-heterocyclic compounds. XLI. Synthesis of 4-substituted 6,7-dihydro-5H-pyrimido[5,4-d][1]benzazepines, 1,2,5,6-tetrahydro-4H-imidazo[1′,2′:1,6]pyrimido[5,4-d][1]benzazepines and their related compounds as a series of potential blood platelet aggregation inhibitors

As a series of polyheterocyclic compounds for exploitation as anti-platelet agents, tricyclic heterocyclic compounds, 4-substituted 6,7-dihydro-5H-pyrimido[5,4-d][1]benzazepines 3–6, 9, 12–14 , and 16–26 , having nitrogen, oxygen, or sulfur containing functional groups at the 4-position, were prepared. In addition, tetra-cyclic heterocyclic compounds, 3-methyl-1,2,5,6-tetrahydro-4H-imidazo[1′,2′:1,6]pyrimido[5,4-d][1]benzaze-pinium chloride ( 7 ), 1,2,5,6-tetrahydro-4H-imidazo[1′,2′:1,6]pyrimido[5,4-d][1]benzazepines 10a-e , 2,3,6,7-tetrahydro-1H 5H-pyrimido[1′,2′:1,6]pyrimido[5,4-d][1]benzazepine ( 11 ), and 1,2,5,6-tetrahydro-4H-thiazolo-[3′,2′:1,6]pyrimido[5,4-d][1]benzazepinium chloride ( 15 ) via ring closure of 4-(hydroxyalkylamino)- 6, 9a-e , and 3c , and 4-(2-hydroxyethylthio)-6,7-dihydro-5H-pyrimido[5,4-d][1]benzazepine ( 14 ) with phosphoryl chloride or thionyl chloride, respectively, were also prepared. Their inhibitory activities against collagen-induced aggregation of rabbit blood platelets in vitro were investigated. Among them, compound 5 having a morpholino group at the 4-position on the tricyclic nucleus, which enhanced the activity more than 14-fold as compared with aspirin, was found to have the most satisfactory in inhibitory activity.     

Studies on the metabolism of gomisin A (TJN-101). II. Structure determination of biliary and urinary metabolites in rat

After oral administration of gomisin A (1) to rats, the bile and urine were collected and treated with beta-glucuronidase and arylsulfatase. Seven metabolites, met B (2), met A-III (3), met E (4), met D (5), met F (6), met G (7), and met H (8) were isolated from the bile treated with the enzymes. Eight metabolites 2-8, and met A-II (9) were isolated from the urine treated with the enzymes. A major metabolite 2, and two minor metabolites 3 and 9 were identified as met B, met A-III, and met A-II, respectively, which are oxidative products of 1 formed by rat liver S9 mix. The structures of five new metabolites 4-7, and 8 were determined on the basis of chemical and spectral studies.
