Sonodynamic therapy (SDT) is a promising modality for cancer treatment, involving the synergistic interaction of ultrasound and some chemical compounds termed as sono-sensitizers. It has been found that SDT can lead to apoptotic cell death because of the induction of direct sonochemical and subsequent redox reactions. However, the detailed mechanisms are not clear. This study was to identify the cytotoxic effects of ultrasound-activated protoporphyrin IX (PpIX) on MDA-MB-231 cells. The fluorescence microscope was used to detect the sub-cellular localization of PpIX. Several distinct sonochemical effects were found after SDT treatment, including the decrease of cell viability, generation of intracellular ROS, the loss of mitochondrial membrane potential. The activation of some special apoptosis-associated proteins [Caspase-9, Caspase-3 and polypeptide poly (ADP-robose) polymerase] was evaluated by western blotting. The results show that PpIX mediated SDT (PpIX-SDT) treatment could obviously inhibit the proliferation of MDA-MB-231 cells, and which was significantly reduced by the pan-Caspase inhibitor z-VAD-fmk and the reactive oxygen species (ROS) scavenger N-acetylcysteine (NAC). Further, SDT induced a conspicuous loss of mitochondrial membrane potential (MMP) and a mass of ROS accumulation in MDA-MB-231 cells at 1 h post-treatment and the SDT-treated cells showed obvious Caspase-3 and Caspase-9 activation, and PARP cleavage at 6 h after treatment. And, the general apoptosis marker-Caspase-3 activation-was also greatly relieved by NAC. These findings primarily indicate a Caspase-depended apoptosis could be induced by PpIX-SDT in MDA-MB-231 cells, and the intracellular ROS was involved during the apoptotic process. 相似文献

(R,R)-Salen-based chiral polymer P-1 was synthesized by the polymerization of 5,5'-((2,5-dibutoxy-1,4-phenylene)bis(ethyne-2,1-diyl))bis(2-hydroxy-3-(piperidin-1-ylmethyl) benzaldehyde (M-1) with (1R,2R)-cyclohexane-1,2-diamine (M-2) via nucleophilic addition- elimination reaction, and (R,R)-salan-based polymer P-2 could be obtained by the reduction reaction of P-1 with NaBH(4). (R,R)-Salen-based chiral polymer P-1 can exhibit greater fluorescence enhancement response toward (l)-α-hydroxyl carboxylic acids, and the value of enantiomeric fluorescence difference ratio (ef) can reach as high as 8.41 for mandelic acid and 6.55 for lactic acid. On the contrary, (R,R)-salan-based chiral polymer P-2 shows obvious fluorescence quenching response toward α-hydroxyl carboxylic acids. Most importantly, (R,R)-salen-based polymer P-1 can display bright blue fluorescence color change in the presence of (l)-α-hydroxyl carboxylic acids under a commercially available UV lamp, which can be clearly observed by the naked eyes. 相似文献

New coumarin-based chiral thiourea sensor 1 was found to be an enantioselective fluorescent chemosensor for N-Boc-protected proline. The chiral sensor shows lower background fluorescence, and higher fluorescence enhancement with 18 nm blue shifts. A novel chiral (S)‐BINAM‐based fluorescent polymer sensor was designed and synthesized by the polymerization of 4,4′‐((2,5‐dibutoxy‐1,4‐phenylene)bis(ethyne‐2,1‐diyl))‐dibenzaldehyde ( M‐1 ) with (S)‐2,2′‐binaphthyldiamine (S‐BINAM, M‐2 ) via Schiff's base formation. The resulting helical chiral polymer sensor exhibited remarkable “turn‐on” bright blue fluorescence color upon the addition of trivalent metal ions under a commercially available UV lamp; this change can be clearly observed by the naked eye for direct visual discrimination at low concentration. Two perylene diimide (PDI) enantiomers ( d/l ‐PDI ) incorporating the d /l ‐alanine moiety have been designed and synthesized. d/l ‐PDI in chloroform displays bright‐yellow fluorescence that is redshifted to orange‐red when the solvent contains a methanol fraction of 99 vol %. No circular dichroism (CD) or circularly polarized luminescence (CPL) signals were observed for d/l ‐PDI enantiomers in CHCl3. Interestingly, the d/l ‐PDI enantiomers exhibit clear mirror‐image Cotton effects and CPL emission in the aggregate state. The optical anisotropy factor (glum) is as high as 0.02 at fm=99 %, which can be attributed to self‐assembly through intermolecular π–π interactions in the aggregate state. 相似文献

Combustion provides about 80% energy for our daily life and industrial production. But thermal efficiency of traditional combustion technologies is low, which causes energy waste and serious environmental pollution. In order to improve the combustion efficiency, a combined method based on non-equilibrium plasma generated by dielectric barrier discharge and OH radicals coming from water-steam additive was proposed in this work, and plasma assisted propane combustion was examined and evaluated. The results indicated that when relative humidity (RH) was 20% and applied peak voltage was fixed at 8.75 kV, the relative intensity of OH radical and the flame temperature reached the maximum value at the flame root. At the same time, propane combustion was the most complete. In addition, we found that the erosion of the inner electrode was weakened by H2O addition, and the symmetry of discharge current was changed from symmetry to asymmetry with the increase of RH. Compared with the pure air undischarged combustion, when the relative humidity was 20% and under the discharge conditions of 8.75 kV, the lean-burn extinction limit was extended to 0.4,which is far lower than the traditional lean-burn limit (0.51). 相似文献
