The chemical bonding and spectral assignments of rhodium(III)‐catalyzed closo‐dodecaborate (RhCD) complex are systematically studied using the density functional theory calculations. It is found that the calculated main bond lengths of framework are in good agreement with experimental X‐ray observation, and the pronounced hybridization of B‐2p and Rh‐4d states is responsible for the structural stability, reflected by the large dissociation energy and HOMO–LUMO energy gap. The AdNDP chemical bonding analysis indicates that the RhCD complex can be stabilized by two H‐bridged 3c‐2e σ‐bonds (B‐H‐Rh triangles). Additionally, the theoretical calculations reproduce well the main experimental IR spectrum, and the characteristic peaks are properly assigned. These results will be helpful for further insight into the unique electronic structure of the species, and provide valuable references for potential applications in novel materials.  相似文献

PhrB from Agrobacterium fabrum is the first prokaryotic photolyase which repairs (6‐4) UV DNA photoproducts. The protein harbors three cofactors: the enzymatically active FAD chromophore, a second chromophore, 6,7‐dimethyl‐8‐ribityllumazine (DMRL) and a cubane‐type Fe‐S cluster. Tyr424 of PhrB is part of the DNA‐binding site and could provide an electron link to the Fe‐S cluster. The PhrBY424F mutant showed reduced binding of lesion DNA and loss of DNA repair. The mutant PhrBI51W is characterized by the loss of the DMRL chromophore, reduced photoreduction and reduced DNA repair capacity. We have determined the crystal structures of both mutants and found that both mutations only affect local protein environments, whereas the overall fold remained unchanged. The crystal structure of PhrBY424F revealed a water network extending to His366, which are part of the lesion‐binding site. The crystal structure of PhrBI51W shows how the bulky Trp leads to structural rearrangements in the DMRL chromophore pocket. Spectral characterizations of PhrBI51W suggest that DMRL serves as an antenna chromophore for photoreduction and DNA repair in the wild type. The energy transfer from DMRL to FAD could represent a phylogenetically ancient process.  相似文献

A phytochemical investigation of the whole plant of Liparis regnieri Finet led to the isolation of one new biphenantherene (1) and ten new nervogenic acid derivatives (211), together with nine known compounds. Their structures were elucidated mainly by extensive analyses of 1D and 2D NMR spectroscopic data and chemical reactions. Additionally, for the first time a previously unreported biphenantherene was discovered to effectively suppress TNF-α induced expression of NF-κB-Luc in Hela cells with the IC50 value of 1.80 μM and the structure-activity relationshipwas also discussed.  相似文献

Chemical investigation of the gorgonian coral Junceella fragilis resulted in the isolation of a new norditerpenoid fragilolide A (1), sixteen new briarane diterpenoids fragilolides B-Q (2–17), together with frajunolides H and N, and three known norcembranoids scabrolide D, sinuleptolide and 5-epi-sinuleptolide. The structures of new compounds were determined on the basis of extensive spectroscopic analysis, including the experimental and calculated ECD data and single-crystal X-ray diffraction for the configurational assignments. The structure of fragilolide A featured an unprecedented 4,13- and 7,11-fused tetracyclic norcembranoid, while the biogenetic relationships of the briarane analogues were postulated. Frajunolide H exerted significant inhibition against a panel of tumor cell lines, and six briarane diterpenoids (3, 6, 8, 12, 16, and frajunolide N) exhibited the inhibitory effects against the HBeAg express of hepatitis B virus in HepAD38 cells. In addition, sinuleptolide and 5-epi-sinuleptolide exerted the effects to inhibit NO production in RAW264.7 macrophage cells, in addition to the activation of ARE and the inhibition of NF-κB expression.  相似文献

The development of small-molecule G-quadruplex DNA probes has attracted significant attention in recent years. However, G-quadruplexes can display a wide variety of topologies, which process different structures and functions. Therefore, selective discrimination one G-quadruplex structure over another is promising. Herein, we reported the design, synthesis and biological evaluation of a long-chain fatty amine functionalized triphenylamine-quinolinium conjugate 1b. Significant enhancement of the fluorescence intensity (over 180 fold) was observed when 1b bound with HRAS G-quadruplex DNA, while much weaker enhancements were presented in the presence of other G-quadruplexes (45–90-fold) and single/double-stranded DNAs (less than 20-fold), indicating 1b had an excellent selectivity to HRAS. The details of the interactions were investigated by UV–Vis, FID and CD analysis. The results show 1b could interact and stabilize HRAS structure mainly by π-π stacking binding mode. The introduced amine chain of the structure core was found to be better in the terms of inducing selectivity toward G-quadruplex structure. In addition, the application of 1b as a fluorescent agent for living cell imaging was also demonstrated.  相似文献

基于甲醛、苯和二氧化硫在纳米Ti3CeY2O11上的催化发光有交叉敏感现象,在3个波长处分别确定甲醛、苯和二氧化硫浓度与催化发光信号强度的响应关系,再依据发光信号强度的叠加性特征即可获取甲醛、苯和二氧化硫的准确浓度,据此建立了同时测定空气中甲醛、苯和二氧化硫的新方法.3个分析波长分别为420、535和680 nm,敏感材料表面温度为280℃,载气流速为130 mL/min.本方法对甲醛、苯和二氧化硫的检出限(3σ)分别为0.04、0.05和0.10 mg/m3,线性范围分别为0.08~75.60 mg/m3、0.10~101.40 mg/m3和0.30~115.00 mg/m3, 回收率分别为96.4%~103.7%、97.8%~102.5%和97.2%~103.3%.常见共存物,如乙醛、甲苯、硫化氢、氨、甲醇、乙醇和二氧化碳等不干扰测定.连续200 h通浓度均为50 mg/m3的甲醛、苯和二氧化硫混合气体,发光强度的相对偏差≤2%,表明此纳米级复合氧化物对甲醛、苯和二氧化硫的敏感性的寿命长.本方法充分利用了交叉敏感现象,可以实现空气中甲醛、苯和二氧化硫的在线分析.  相似文献

This study aimed at developing an efficient biotransformation process for phenylacetic acid production from phenylacetonitrile by using recombinant Escherichia coli M15 harboring a double mutant MG nitrilase (I113M/Y199G) from Burkholderia cenocepacia J2315. A yield of 2310 U/mL nitrilase was obtained by fermentation after the optimization of cultivation conditions, with a specific activity of 64 U/mg dcw. The MG nitrilase showed high substrate tolerance and completely hydrolyzed 100 mM phenylacetonitrile in 30 min under optimal conditions. To alleviate substrate inhibition, periodic or continuous batch-feeding of substrate was used during the biotransformation. Up to 164 g/L substrate was completely hydrolyzed in 9 h with continuous batch-feeding using resting cells, corresponding to 400 U/mL of nitrilase activity, and leading to production of 163.4 g/L phenylacetic acid. The hydrolysis process has potential application for phenylacetic acid production on a large scale.  相似文献

采用行波理论系统地研究了压电阶梯梁的自由振动分析以及强迫响应的分析方法. 基于分布 参数理论研究了压电阶梯梁的波传播特性,忽略柔性梁横向剪切和转动惯量的影响,给出了 梁的轴向和横向的简谐波解. 将压电阶梯梁离散化为单元,考虑压电片的刚度和质量的影响, 建立了节点散射模型. 应用位移连续和力平衡条件,推导了节点的波反射和波传递矩阵,在 此基础上,引入波循环矩阵的概念,给出波循环矩阵、波传递系数矩阵的确定方法. 应用波 循环矩阵可以有效地计算结构的固有频率. 另外,应用波传递系数研究了压电陶瓷作动器位 置对其驱动能力的影响. 得出两个主要结论:1)作动器靠近悬臂梁固定端将有较强的驱动 能力,悬臂梁边界反射行波产生弯曲消失波有利于增大压电波的模态传递系数;2)模态传 递系数与固有频率的灵敏度密切相关,波传递系数越大, 对应该处固有频率变化灵敏度越大. 另外,数值算例表明了行波方法比有限元方法具有更高的计算精度.  相似文献
