Various repertoires of membrane protein interactions determine cellular responses to diverse environments around cells dynamically in space and time. Current assays, however, have limitations in unraveling these interactions in the physiological states in a living cell due to the lack of capability to probe the transient nature of these interactions on the crowded membrane. Here, we present a simple and robust assay that enables the investigation of transient protein interactions in living cells by using the single-molecule diffusional mobility shift assay (smDIMSA). Utilizing smDIMSA, we uncovered the interaction profile of EGFR with various membrane proteins and demonstrated the promiscuity of these interactions depending on the cancer cell line. The transient interaction profile obtained by smDIMSA will provide critical information to comprehend the crosstalk among various receptors on the plasma membrane.Subject terms: Fluorescence imaging, Super-resolution microscopy, Single-molecule biophysics  相似文献

An efficient and feasible synthetic approach was developed for the synthesis of an array of new flavane derivafives from the substituted benzaldehyde with the reduction of chalcones and subsequent cyclization as the key steps. The purity and structure of the products were confirmed by the elemental analysis and a combination of its IR, ^1H and ^13C NMR, and mass spectra. These synthetic compounds were tested for xanthine oxidase (XO) inhibitions and antifungal actions against Candida albicans, Cryptococcus neoformans, Aspergillus sp. and Trichophyton rubrum. 7-Hydrazinocarbonylmethoxy-4'-methoxyflavane (9) was found to be the most XO inhibitory with IC50=76.4 μmol/L, and the most potent antifungal compound was 4'-hydrazinocarbonylmethoxyflavane (12) with minimal inhibition concentration MIC=8 μg/mL against Trichophyton rubrum.  相似文献

建立了使用超高效液相色谱-串联质谱(UHPLC-MS/MS)高效、快速直接测定茶叶中游离氨基酸的方法。通过对质谱、色谱条件及氨基酸提取条件的优化,以含0.2%(体积分数)甲酸的5 mmol/L乙酸铵水溶液和甲醇为流动相进行梯度洗脱,在电喷雾离子(ESI)源正离子扫描模式下检测,通过UHPLC-MS/MS测定,共解析了茶叶中的20种氨基酸。结果表明,茶氨酸(Thea)、Arg、Asn和Asp在50~500 μg/L范围内线性关系良好,其他氨基酸在10~250 μg/L范围内线性关系良好,相关系数均大于0.99;加标回收率为92.3%~109.2%,相对标准偏差为2.00%~9.88%,检出限为0.001~0.011 mg/L,定量限为0.010~0.053 mg/L。该方法灵敏、准确,具有良好的重复性和稳定性,可有效检测出茶叶中的20种氨基酸及氨基类成分。  相似文献

Three new hypocrellin derivatives, amino- or amino acid-substituted on the side ring of hypocrellin B (HB), were synthesized by the reactions of HB with 3-methoxypropylamine, 6-aminohexanoic acid and gamma-amino-n-butyric acid, respectively. The structures of these compounds were characterized with proton nuclear magnetic resonance spectra, infrared spectra and mass spectra. The UV-visible absorption spectra, singlet oxygen-generating quantum yield and amphiphilicities of hypocrellin derivatives were measured and compared with HB, the parent compound. These derivatives showed strong absorption in the domain of the phototherapeutic window (600-900 nm) and improved amphiphilicity. HB and the derivatives were preliminarily tested for their photodynamic effects on human oral cavity epithelial carcinoma KB cell line in vitro. Two amino acid-substituted hypocrellins showed phototoxicity to the KB cell line. At an inhibitory dosage of 50% killing only 0.51 mumol L-1 compound 3 (or 0.88 mumol L-1 compound 2) and 0.5 J cm-2 irradiation were required. The hypocrellins exhibited some dark toxicity to the KB cell line. HB and amino acid-substituted hypocrellins showed lower dark toxicity to the KB cell line than amino-substituted hypocrellins in the assessment of cell survival.  相似文献

By using a micro-bomb calorimeter, the standard enthalpies of combustion of C60 and C70 have been determined to be - (25 947.1±8.5) and - (29 956.1 ± 8.9) kJ/mol respectively. A g. 1. c. analysis indicated that the amounts of residual organic solvents in the samples were very small, and their effects on the final results were negligible. The energy of combustion ofC60 determined in this work is in agreement in the uncertainty interval with that determined by means of traditional calorimeter using macro amount of sample. The enthalpies of formation of these two substances have been derived. The strain energies in the molecules of C60 and C70 were estimated by a bond energy scheme and by using corannulene as the model compound, the results estimated from different methods are very close. A novel diterpene derivative, 5-methyl-5-(4,8,12-trimethyl-tridecyl)-dihydro-furan-2-one (1) and a new chromone, 5-hydroxy-7-methoxy-3-methyl-chromen-4-one (2), along with a known compound, phytol (3) have been isolated from the aerial parts of Hypericum perforatum. Their structures were established on the basis of spectroscopic analysis and by comparison with published values.  相似文献
