The keto–enol tautomerism of cyameluric acid, both in gas phase and in water and methanol solution, has been studied at the B3LYP/6-31++g(d,P) level of theory in this paper. The harmonic frequencies of all the structures are calculated. The results show that the transition states of the tautomerism are 4-membered ring conformations in gas phase, whereas 6-membered ring conformations in solution. In the first proton transfer, activation energy ΔE# is 56.4 and 50.9 kJ/mol for water and methanol solution, respectively, which is much lower than that in gas phase (163.2 kJ/mol). Solvent molecules (water and methanol) produce an important catalytic effect in the tautomerism, especially for methanol-solvated system. NBO analysis shows that there is a strong interaction between cyameluric acid and solvent molecules in transition states. AIM charge analysis indicates that the keto–enol tautomerism shows a certain degree of proton transfer character. From the reaction enthalpy and reaction rate point of view, keto–enol tautomerism in water-solvated and methanol-solvated system is easier than that in gas phase. The keto–enol tautomerisms are endothermic both in gas phase and in solution, so the enol forms are less stable than the keto ones. 相似文献

In this paper, we studied polyetheneglycol (PEG) pretreatment effect on the mature embryo culture in vitro by using barley (Hordeum vulgare L.) seeds. Meanwhile, we analyzed and assayed its mineral element and endogenous hormone level. The experimental results were as follows: (1) PEG-6000 imbibition could obviously slow down the water timecourse absorbed by barley seeds; (2) 10% PEG-6000 treatment of barley seeds for 3 h had a positive effect on germination in vitro and callus induction of the barley seed mature embryos; (3) 10% PEG-6000 treatment inhibited soluble leakage from the seeds; (4) N leakage was mainly from the endosperms, Mn2+ leakage from embryos; (5) PEG-6000 treatment changed greatly the hormone level (ABA, IAA, GAs), which influenced the percentage of plantlets from the mature embryo callus. The results can provide some clues to scientific sowing of crop seeds, pretreatment for the purpose of uniform seedlings, and the explant response quality in plant tissue culture. 相似文献

A new thiol weak-fluorescence probe, 5-maleimidyl-2-(m-methylphenyl)benzoxazole (MMPB), gives a highly fluorescence product in the presence of Cys. In this paper, MMPB has been developed for the fluorimetric determination of cysteine (Cys). At lambda(ex)/lambda(em) = 305.6/425.6 nm, the linear range is from 0 to 3.3 x 10(-7) mol l(-1) and the detection limit (sigma = 3) of 6.2 x 10(-10) mol l(-1). The main advantage of this method lies in the relative high selectivity compared with the methods using other N-substituted maleimide type of thiol reagents, in which 0.15-fold (molar ratio) of GSH is allowed and most of other amino acids at 100-fold (molar ratio) level had no obvious effect on the results. The determination of four volatile halocarbons (CHCl3, CCl4, C2HCl3 and C2Cl4) in water by headspace liquid-phase microextraction (HS-LPME) with gas chromatography using a micro electron capture detector (GC-μECD) is described. The effects of the type and volume of the extraction solvent, headspace volume, stirring rate, extraction temperature and time and ionic strength on the extraction performance are investigated and optimized. The developed protocol yields a linear calibration curve in the concentration range from 0.05 to 50 µg L?1 for the target analytes; the detection limits ranged from 0.003 to 0.146 µg L?1 and the relative standard deviation (R.S.D.) values below 8.45%. The results demonstrate that HS-LPME followed with GC-μECD is a simple and reliable technique for the determination of volatile halocarbons in water samples. 相似文献
