Cisplatin has been clinically used for treatment of solid tumors such as non–small-cell lung cancer for decades. However, tumor resistance may be acquired with losing the antitumor activity of cisplatin. As cellular membrane is the first barrier that cisplatin has to overcome before its further action inside the cells, the membrane composition must play a vital role in the cisplatin uptake and excretion, which further influences cisplatin sensitivity. In this work, we applied time-of-flight secondary ion mass spectrometry (ToF-SIMS) surface analysis combined with principle component analysis to distinguish the differences of cell membrane composition between non–small-cell lung cancer cells (A549) and its cisplatin resistant counterpart A549/DDP cells. The decreased phosphatidylcholine content and more abundant cholesterol were observed in the drug resistant cell surfaces, indicating the decreased membrane fluidity of A549/DDP cells. Moreover, we further compared membrane composition of A549 and A549/DDP cells after being treated with different concentrations of cisplatin. A higher composition level of proteins was discovered on all groups of A549/DDP cell membranes. The altered surface chemistry of cellular membranes induced by cisplatin indicates the significance of membrane structures in the drug resistance, which deserves further investigations to this regard. 相似文献

Single molecule spectroscopy is applied in studies of diffusion and surface adsorption in sol-gel-derived mesoporous silica thin films. Mesoporous films are obtained by spin casting surfactant-templated sols onto glass substrates. Small-angle X-ray diffraction results are consistent with hexagonally ordered mesophases in as-synthesized (i.e., surfactant-containing) films. Upon calcination, a 30% contraction and disordering of these structures occurs. Nile Red is used as a fluorescent probe of both the as-synthesized and calcined films. It is loaded into the samples at subnanomolar levels either prior to spin casting or after calcination. Fluorescence imaging and single-point fluorescence time transients show the dye molecules to be relatively mobile in the as-synthesized samples. In contrast, the molecules appear entrapped at fixed locations in dry calcined films. In calcined films rehydrated under high humidity conditions, the Nile Red molecules again become mobile. Time transients obtained from the as-synthesized and rehydrated samples provide clear evidence for frequent reversible adsorption of the dye to the silica surfaces. Autocorrelations of the time transients provide quantitative data on the mean diffusion coefficients (D = 2.4 x 10(-10) and 2.6 x 10(-10) cm2/s) and mean desorption times (1/k = 25 and 40 s) for the as-synthesized and rehydrated films, respectively. The second-order nonlinearities in thermally poled lead borate glasses were studied. After poling with different electrodes, both bulk and near surface second-order nonlinearities were obtained. The second harmonic generation signal from near surface effect was two orders of magnitude larger than the bulk effect. By simulating the poling process with a multiple carrier model, we concluded that the bulk second-order nonlinearity was caused by the deficiency of Pb2+ in a thick layer after poling, and the near surface second-order nonlinearity was caused by the depletion of H+ in a thin layer when H+ injection from the atmosphere was blocked. 相似文献

One-dimensional transverse oscillations in a layer of a non-linear elastic medium are considered, when one of the boundaries is subjected to external actions, causing periodic changes in both tangential components of the velocity. In a mode close to resonance, the non-linear properties of the medium may lead to a slow change in the form of the oscillations as the number of the reflections from the layer boundaries increases. Differential equations describing this process were previously derived. The equations obtained are hyperbolic and the change in the solution may both keep the functions continuous and lead to the formation of jumps. In this paper a model of the evolution of the wave patterns is constructed as integral equations having the form of conservation laws, which determine the change in the functions describing the oscillations of the layer as “slow” time increases. The system of hyperbolic differential equations previously obtained follows from these conservation laws for continuous motions, in which one of the variables is slow time, for which one period of the actual time serves as an infinitesimal quantity, while the second variable is the real time. For the discontinuous solutions of the same integral equations, conditions on the discontinuity are obtained. An analogy is established between the solutions of the equations obtained and non-linear waves propagating in an unbounded uniform elastic medium with a certain chosen elastic potential. This analogy enable discontinuities which may be physically realised to be distinguished. The problem of steady oscillations of an elastic layer is discussed. 相似文献

A pentagon-shaped undoped BaTiO3 crystal was used to construct a high-quality mutually pumped phase conjugator. It yields two very stable phase-conjugate outputs (ǃ.7% fluctuation in intensity) with high reflectivity (the highest ᅝ%) when the two incident beams are of equal intensities. The response time of the conjugator is rather fast (ƻ s) and the tolerance of the acceptance angle is large too (걄°). The delayed luminescence was applied to detect the intermediate processes of the excitedstate decay in the selectively excited phycobiliproteins. Phosphorescence spectra of the five types of phycobiliproteins, R-PE, CPC, APC, R-PC, PEC were reported in this article. The five phycobiliproteins showed different phosphorescence yields, the sequence of which was the same as that of the singlet oxygen yields. Based on the observation, it can be concluded that each of the chromophores possesses a characteristic phosphorescence emission. The delayed luminescence spectra of APC at different aggregation states (trimer, monomer and denatured APC) are researched. The lower aggregation APC showed less phosphorescence because of relative loose structures and less interaction with the surrounding proteins, while the denatured APC showed delayed fluorescence instead of phosphorescence because of triplet-triplet annihilation. 相似文献
