We report a novel automated device for nucleic acid extraction, which consists of a mechanical control system and a disposable cassette. The cassette is composed of a bottle, a capillary tube, and a chamber. After sample injection in the bottle, the sample is lysed, and nucleic acids are adsorbed on the surface of magnetic silica beads. These magnetic beads are transported and are vibrated through the washing reagents in the capillary tube under the control of the mechanical control system, and thus, the nucleic acid is purified without centrifugation. The purified nucleic acid is automatically extracted in 3 min for the polymerase chain reaction (PCR). The nucleic acid extraction is dependent on the transport speed and the vibration frequency of the magnetic beads, and optimizing these two parameters provided better PCR efficiency than the conventional manual procedure. There was no difference between the detection limits of our novel device and that of the conventional manual procedure.  相似文献

This paper investigates the relationship between the pre-annealing conditions and the thermal stability of uniaxially-drawn poly(vinylidene fluoride) (PVDF) films in order to clarify their technical limits in terms of temperatures that can be used for assembly processes and for practical applications. Specimens that are pre-annealed below their melting temperature apparently shrink in the stretch-direction when they are exposed to elevated temperatures above the pre-annealing temperature. Since the content of β-PVDF in the films decreases simultaneously with the shrinkage, their piezoelectric properties also deteriorate. In addition, there is a suggestion that the level of polarization in the remaining β-phase decreases significantly during annealing above 90-100 °C. However, the dimensions and the piezoelectric coefficients of the films remain stable during annealing below the pre-annealing temperature. Therefore, the thermal stability of PVDF films can be controlled practically by using the appropriate pre-annealing temperature. By contrast, the films were softened at 90-100 °C when the pre-annealing treatment was conducted above the melting temperature. The softening of films that are pre-annealed above the melting temperature is a different phenomenon from that observed in specimens that are pre-annealed below the melting temperature.  相似文献

Monthly variation in chlorophyll concentration of living ginko tree leaves 65 m away from a system was remotely estimated by a laser-induced fluorescence imaging lidar. The combination of a pulsed laser and a short-time gated CCD using an image intensifier made it possible to monitor the weak fluorescence signal from the ginko tree leaves as an image. By applying the experimental idea that a ratio of intensity of the chlorophyll fluorescence at 740 nm to that at 685 nm showed a linear correlation to the chlorophyll concentration, the fluorescence image of the ginko tree obtained by the lidar was converted to the chlorophyll concentration distribution image.  相似文献

Mesostructured calcium phosphates constructed by ionic frameworks were synthesized using carboxylic acid- and amine-type surfactants in mixed solvent systems of ethanol and water. A lamellar mesostructured calcium phosphate was prepared using palmitic acid as an anionic surfactant, as in the case using n-alkylamines. A wormhole-like mesostructured calcium phosphate can be obtained using dicarboxyl N-lauroyl- l-glutamic acid, whose headgroup is larger than that of palmitic acid. Similar mesostructured product was obtained using 4-dodecyldiethylenetriamine with a large headgroup containing two primary amine groups. Interactions of carboxyl and primary amino groups in the surfactant molecules with inorganic species are quite important for the formation of mesostructured calcium phosphates. The Ca/P molar ratio of mesostructured calcium phosphates was strongly affected by the molecular structure of surfactants containing carboxyl and primary amino groups. Ca-rich materials can be obtained using carboxylic acid-type surfactants (Ca/P approximately 1.7) rather than amine-type surfactants (Ca/P approximately 1.0).  相似文献

Background

In drug discovery research, cell-based phenotypic screening is an essential method for obtaining potential drug candidates. Revealing the mechanism of action is a key step on the path to drug discovery. However, elucidating the target molecules of hit compounds from phenotypic screening campaigns remains a difficult and troublesome process. Simple and efficient methods for identifying the target molecules are essential.

Results

2-Amino-4-(3,4-(methylenedioxy)benzylamino)-6-(3-methoxyphenyl)pyrimidine (AMBMP) was identified as a senescence inducer from a phenotypic screening campaign. The compound is widely used as a Wnt agonist, although its target molecules remain to be clarified. To identify its target proteins, we compared a series of cellular assay results for the compound with our pathway profiling database. The database comprises the activities of compounds from simple assays of cellular reporter genes and cellular proliferations. In this database, compounds were classified on the basis of statistical analysis of their activities, which corresponded to a mechanism of action by the representative compounds. In addition, the mechanisms of action of the compounds of interest could be predicted using the database. Based on our database analysis, the compound was anticipated to be a tubulin disruptor, which was subsequently confirmed by its inhibitory activity of tubulin polymerization.

Conclusion

These results demonstrate that tubulin is identified for the first time as a target molecule of the Wnt-activating small molecule and that this might have misled the conclusions of some previous studies. Moreover, the present study also emphasizes that our pathway profiling database is a simple and potent tool for revealing the mechanisms of action of hit compounds obtained from phenotypic screenings and off targets of chemical probes.
