The highly refined organic chemistry in solid-phase synthesis has made it the method of choice not only to assemble peptides but also small proteins - mainly on a laboratory scale but increasingly also on an industrial scale. While conductive heating occasionally has been applied to peptide synthesis, precise microwave irradiation to heat the reaction mixture during coupling and N(α)-deprotection has become increasingly popular. It has often provided dramatic reductions in synthesis times, accompanied by an increase in the crude peptide purity. Microwave heating has been proven especially relevant for sequences which might form β-sheet type structures and for sterically difficult couplings. The beneficial effect of microwave heating appears so far to be due to the precise nature of this type of heating, rather than a peptide-specific microwave effect. However, microwave heating as such is not a panacea for all difficulties in peptide syntheses and the conditions may need to be adjusted for the incorporation of Cys, His and Asp in peptides, and for the synthesis of, for example, phosphopeptides, glycopeptides, and N-methylated peptides. Here we provide a comprehensive overview of the advances in microwave heating for peptide synthesis, with a focus on systematic studies and general protocols, as well as important applications. In this report, a new approach for the fast determination of hydrazine compounds (hy) in complex matrices is presented. The experimental protocol is based on poly(methylmethacrylate) (PMMA) microchip separations with contactless conductivity detection using a compact portable device, which integrates all separation and detection components. Three hy (hydrozine (Hy), methylhydrazine (MH), and 1,1-dimethylhydrazine (UDMH)) were separated within < 30 s at a separation voltage of 3.8 kV using a L(-)-histidine/2-(N-morpholinoethanesulfonic acid) (His/MES) buffer (25:50 mM, pH 5.87). The contactless conductivity detection enables detection limits for Hy, MH, and UDMH of 11.9, 35.5, and 337.8 ng/mL, respectively, with linear concentration dependence up to 10 μg/mL. In complex matrices such as soil samples or river water, interferences were eliminated by implementing ultrasound-assisted headspace single-drop microextraction of hy under strongly alkaline conditions, using an aqueous drop of His/MES buffer as the extractant phase. The incorporation of this miniaturized sample preparation step led to improved limits of detection for Hy, MH, and UDMH of 6.5, 15.3, and 11.4 ng/mL, respectively. Bulk polycrystalline samples of Eu2O3-doped MgB2 have been synthesized by a standard solid state reaction route and their structural and superconducting properties have been investigated. As a function of Eu2O3 content we have found a significant increase in the critical current density (Jc) and the irreversibility field (Hirr) in the magnetic field range 0–6 T. The XRD results reveal the presence of MgO and EuB6 secondary phases along with the main hexagonal phase of MgB2. The strain values and the lattice distortions have been found to increase almost linearly with the nominal Eu2O3 content. The observed significant improvement in Jc(H) and Hirr in the Eu2O3-doped MgB2 samples, thus is mainly attributed to the lattice distortions introduced by Eu2O3 doping. 相似文献

Entropy generation is the loss of energy in thermodynamical systems due to resistive forces,diffusion processes, radiation effects and chemical reactions. The main aim of this research is to address entropy generation due to magnetic field, nonlinear thermal radiation, viscous dissipation, thermal diffusion and nonlinear chemical reaction in the transport of viscoelastic fluid in the vicinity of a stagnation point over a lubricated disk. The conservation laws of mass and momentum along with the first law of thermodynamics and Fick's law are used to discuss the flow, heat and mass transfer, while the second law of thermodynamics is used to analyze the entropy and irreversibility. The numbers of independent variables in the modeled set of nonlinear partial differential equations are reduced using similarity variables and the resulting system is numerically approximated using the Keller box method. The effects of thermophoresis,Brownian motion and the magnetic parameter on temperature are presented for lubricated and rough disks. The local Nusselt and Sherwood numbers are documented for both linear and nonlinear thermal radiation and lubricated and rough disks. Graphical representations of the entropy generation number and Bejan number for various parameters are also shown for lubricated and rough disks. The concentration of nanoparticles at the lubricated surface reduces with the magnetic parameter and Brownian motion. The entropy generation declines for thermophoresis diffusion and Brownian motion when lubrication effects are dominant. It is concluded that both entropy generation and the magnitude of the Bejan number increase in the presence of slip. The current results present many applications in the lubrication phenomenon,heating processes, cooling of devices, thermal engineering, energy production, extrusion processes etc. 相似文献

Two basic disadvantages of the symmetric rank one (SR1) update are that the SR1 update may not preserve positive definiteness
when starting with a positive definite approximation and the SR1 update can be undefined. A simple remedy to these problems
is to restart the update with the initial approximation, mostly the identity matrix, whenever these difficulties arise. However,
numerical experience shows that restart with the identity matrix is not a good choice. Instead of using the identity matrix
we used a positive multiple of the identity matrix. The used positive scaling factor is the optimal solution of the measure
defined by the problem—maximize the determinant of the update subject to a bound of one on the largest eigenvalue. This measure
is motivated by considering the volume of the symmetric difference of the two ellipsoids, which arise from the current and
updated quadratic models in quasi-Newton methods. A replacement in the form of a positive multiple of the identity matrix
is provided for the SR1 update when it is not positive definite or undefined. Our experiments indicate that with such simple
initial scaling the possibility of an undefined update or the loss of positive definiteness for the SR1 method is avoided

The development of cellulase production technology has greatly contributed to the successful use of cellulosic materials as
renewable carbon sources. In this study, a putative endoglucanase IV (EG IV) complementary DNA was cloned from the mycelium
of a strain of the filamentous fungus Trichoderma viride using a PCR-based exon-splicing method and expressed in both a silkworm BmN cell line and in silkworm larvae. Western blot
analysis detected a band of 42 kDa in BmN cells after infection with a recombinant mBacmid/BmNPV/EG IV baculovirus. Sequence
alignment analysis of the T. viride EG IV gene showed two domains that were highly conserved with glycosyl hydrolases and a funga-type cellulose-binding domain.
Analysis of variance showed that silkworms infected with recombinant baculoviruses exhibited significantly higher enzyme activity
that was 48.84% higher than silkworms infected with blank baculoviruses and 46.61% higher than normal silkworms. The expressed
bioactive EG IV was also stable at the pH range from 5.0 to 10.0. The availability of large quantities of bioactive EG IV
in silkworm provided a possibility to produce cellulase transgenic silkworm, which express bioactive cellulase specially in
its digestive tract and improve its metabolism efficiency of mulberry leaves. Its application in the sericulture industry
