Norditerpenoid Alkaloids from the Roots of Aconitum hemsleyanum Pritz.var.Pengzhouense

In continuation of our studies on Aconitum hemsleyanum Pritz.var. pengzhouense, two new norditerpenoid alkaloids, pengshenines A(1) and B(2),have isolated from the roots of the plants and their strucures were elucidated by 1D-and 2D-NMR.     

六氯合铂酸钾与金属硫蛋白的体外反应

本文报道了K₂PtCl₆与兔肝Zn₇MT-Ⅱ和apoMT-Ⅱ的反应包含一个氧化还原反应和一个取代反应。通过紫外可见光谱、园二向色谱、柱层析和X-光电子能谱研究了该反应的性质、铂在反应产物中的键合位置和氧化态。金属硫蛋白(MT)被氧化成单体、双聚和多聚产物，其中含有分子间和分子内CyS-SCy二硫键。Pt(Ⅳ)被还原成Pt(Ⅱ)然后键合于产物中。随着K₂PtCl₆与MT的反应摩尔比和反应时间的增加，键合于产物中的Pt(Ⅱ)的计量数增加而蛋白中所含Zn(Ⅱ)的量减少。当Zn₇MT与4和超过10摩尔的K₂PtCl₆反应时，分别得到了Pt₄Zn₄MT和Pt₈MT。当apoMT与7及超过25倍的K₂PtCl₆在pH 2条件下反应时，分别得到了Pt₇MT和Pt₁₅MT。动力学数据表明K₂PtCl₆与apoMT的反应比与Zn₇MT的反应快。     

阿斯美胶囊中四种组分的高效液相色谱分析

建立了反相高效液相色谱法测定阿斯美胶囊中氨茶碱、盐酸甲氧非那明、马来酸氯苯那敏及那可汀四种组分的含量。色谱柱为ＳｐｈｅｒｉｓｏｒｂＣ８，５μｍ，２００ｃｍ×４０ｍｍＩＤ，流动相为体积分数０５％的三乙胺溶液（磷酸调节ｐＨ５５）＋乙腈＋甲醇（５５０＋３６８＋８２），检测波长为２６４ｎｍ，线性范围分别为：氨茶碱３１２５～２５０ｍｇ／Ｌ，γ＝０９９９６，盐酸甲氧非那明１５６２５～１２５０ｍｇ／Ｌ，γ＝１００００，马来酸氯苯那敏２５～２００ｍｇ／Ｌ，γ＝０９９９９，那可汀８７５～７００ｍｇ／Ｌ，γ＝０９９９９；回收率（ｎ＝３）分别为１０１２％，１００８％，９９９％，９９５％；精密度：日内平均ＲＳＤ（ｎ＝６）分别为０７％，０４％，０４％，０６％；日间平均ＲＳＤ（ｎ＝３）分别为１０％，１０％，１１％，１２％。     

蜜蜂外激素9－ODA和9－HDA的合成

６－溴－２－己酮在Ｌｉ２ｃｕＣｌ４催化下与γ－溴代巴豆酸甲酯进行格氏偶联，合成了蜂王物质９－羰基－（Ｅ）－２－癸烯酸（９－ＯＤＡ），将其还原得到另一蜂王物质９－羟基－（Ｅ）－２－癸烯酸（９－ＨＤＡ）。该法是合成９－ＯＤＡ和９－ＨＤＡ的新路线。     

邻-甲氧基苯酚和α-,β-环糊精包合现象的理论与实验研究

通过紫外可见光谱法考察了水溶液中邻-甲氧基苯酚(o-Mop)和α-与β-环糊精(CD)的分子间相互作用, 利用Hildebrand-Benesi方程给出了两个包合物的稳定常数(K_s). 采用半经验PM3方法研究了α-,β-CD和o-Mop及其类似物苯酚(Phe)、丁香酚(Eug)之间的包络作用, 阐述了这些主客体包合作用过程中体系能量随主客体相对位置改变而变化的细节, 据此推断出主-客体包合物可能的分子结构, 计算了包合物的稳定化能(ΔE_s). 研究结果表明, 本文所选主客体体系而言, 当客体和同一种主体分子作用时, 超分子包合物的ΔE_s随着客体分子苯环上取代基团数目的增多而增加. 基于PM3方法优化得到的主-客体包合物在真空中的分子结构和通过实验方法在水溶液中测定的结构一致.     

One-step hexaplex immunoassays by on-line paper substrate-based electrospray ionization mass spectrometry for combined cancer biomarker screening

Mass spectrometry (MS) is attractive as a multiplexed immunoassay readout benefiting from its high sensitivity, speed and mass resolution. Here, a simple paper-based hexaplex immunoassay with an on-line MS readout was proposed, using functionalized paper as the immune substrates, along with rhodamine-based mass tags assembled on gold nanoparticles prepared as the mass probes (MPs). Simultaneous immune capture and labeling were conducted in one step on paper substrates in 96-well plates with a high throughput within 30 minutes, and the on-line efficient dissociation of the mass tags highly facilitated the hexaplex readout of the immune signals by a newly established on-line paper substrate-based electrospray ionization-MS setup. Six MPs were synthesized for the simultaneous quantification of six important cancer protein markers (cancer antigen 15-3, cancer antigen 19-9, carcinoma embryonic antigen, cancer antigen 125, human epididymis protein 4, and alpha fetoprotein) using only 10 μL serum, presenting satisfactory sensitivity, accuracy and specificity. This platform was further tested in screening for the six biomarkers in serum samples of patients with breast, liver and gastric cancers, showing its high potential for sensitive and specific early cancer diagnosis.

On-line paper substrate based electrospray ionization mass spectrometry for hexaplex immunoassays.     

Carbene-catalyzed enantioselective annulation of dinucleophilic hydrazones and bromoenals for access to aryl-dihydropyridazinones and related drugs

4,5-Dihydropyridazinones bearing an aryl substituent at the C6-position are important motifs in drug molecules. Herein, we developed an efficient protocol to access aryl-dihydropyridazinone molecules via carbene-catalyzed asymmetric annulation between dinucleophilic arylidene hydrazones and bromoenals. Key steps in this reaction include polarity-inversion of aryl aldehyde-derived hydrazones followed by chemo-selective reaction with enal-derived α,β-unsaturated acyl azolium intermediates. The aryl-dihydropyridazinone products accessed by our protocol can be readily transformed into drugs and bioactive molecules.

Polarity inversion of arylidene hydrazones to react with bromoenals via carbene organic catalysis is disclosed. The reaction enantioselectively affords 6-aryl-4,5-dihydropyridazinones and related drugs with proven commercial applications.     

Antidepressant-like effect of triterpenoids extracts from Poria cocos on the CUMS rats by 16S rRNA gene sequencing and LC–MS metabolomics

Abstract

Poria cocos is an edible medicinal mushroom with the effects of inducing diuresis, excreting dampness, invigorating the spleen and tranquilizing the mind. The triterpenoids of Poria cocos as the main active ingredients have shown health benefits of the central nervous system of the human body. Accordingly, this study aimed at further understanding the antidepressant-like effect and a potential mechanism of the triterpenoids extracts from Poria cocos (TPC). As a result, the TPC significantly ameliorated depression-like behaviors on chronic unpredictable mild stress (CUMS) rats, and restored the levels of brain-derived neurotrophic factor and nerve growth factor in the hippocampus of rats. Gut microiota analysis revealed that TPC could increase the biodiversity and markedly regulate the relative abundance of [Prevotella], Allobaculum and Ochrobactrum of CUMS rats. The cecal contents metabolomics pointed to thirteen biomarkers associated with TPC antidepressant effect, which involved in primary bile acid biosynthesis, taurine and hypotaurine metabolism, arginine and proline metabolism. Correlation analysis further showed that there was a strong correlation relationship between the gut microbiota and the cecal contents metabolites, especially compounds involved in energy metabolism, inflammation and immunity. In conclusion, TPC showed a potential antidepressant effect, which was possibly mediated the gut microbiota and cecal contents metabolism.     

两种含锌化合物对慢性染铅大鼠海马NADPH-d阳性神经元影响的比较

为探讨硫酸锌(ZnSO4)和牛磺酸锌(TZC)拮抗铅对学习记忆的损害的不同，采用NADPH-黄递酶(NADPH-d)组化，观察了饮用含0.2g／L醋酸铅(PbAc)饮水和含不同剂量(5．0，15．0g／kg)ZnSO4、(5．9、17．7g／kg)TZC饲料喂养的大鼠海马一氧化氮合酶(NOS)活性的变化。结果表明，各补锌组均不同程度地缓解铅致NOS活力减低，铅 低TZC组、铅 高ZnSO4组和铅 低ZnSO4组大鼠海马神经元的NOS活性明显高于染铅组，其中铅 低TZC组的保护作用最显著，而铅 高TZC组的NOS活性明显低于对照组，而接近染铅组水平。结论是锌对抗铅的毒性作用与锌的化学结合形式、锌的剂量密切相关。     

2,3,7‐Triazabicyclo[3.3.0]octenes Prepared by Tandem Cascade Reaction of Allyl Azides and Olefinic Dipolarophiles

Tandem cascade reactions of allylazides and olefinic dipolarophiles to give cis‐fused 2,3,7‐triazabicyclo [3.3.0]octenes ( 5, 6 or 7 ) are reported. Therein, an intermolecular dipolar cycloaddition of azide and alkene gave a triazoline which was followed by isomerization of the triazoline to a diazoester ( 4 ) and then an intramolecular dipolar cycloaddition from the diazo functional group and the double bond in 4 to give 5 . Compound 5 may further more undergo a Michael addition to give 7‐substituted‐ 2,3,7‐ triazabicyclo [3.3.0]oct‐2‐ene ( 6 ) or a tautomerization to give 2,3,7‐triazabicyclo[3.3.0]oct‐3‐ene ( 7 ). The reaction may be manipulated to stop at a particular stage by adopting a suit able solvent or an appropriate temperature.
